Iodine is an essential element necessary for the production of the thyroid hormones T3 and T4. These hormones are necessary for metabolism and are especially important for brain development in infants and young children. Iodine is transported into the thyroid by the sodium iodide symporter. Perchlorate is known to inhibit iodine uptake into the thyroid by binding to the sodium iodide symporter, thus acting as a potential neurotoxicant for developing children.The research in this dissertation consists of two goals. Pyrrolizidine Alkaloids (PAs) are known plant toxins which can cause hepatic veno-occlusive disease (HVOD) in both humans and livestock when ingested. Although PAs are primarily known for their hepatotoxicity they also induce genotoxicity, carcinogenicity and pneumotoxicity. PAs are consumed through plants as food, for medicinal purposes, or as contaminants of agricultural crops. Aside from ingesting the plants directly PA exposure may occur through the consumption of honey produced by bees that visit PA-containing plants or by drinking milk produced by animals that have consumed PA-containing plants. Possible PA contamination in our food chain is a potential health risk.

Cyanobacteria, also known as blue-green algae, are a common component of many freshwater and marine ecosystems and have a potential to produce several different classes of biotoxins. The most common class of cyanobacterial toxins are the microcystins (MCs) and cylindrospermopsins (CYLs). MCs are cyclic heptapeptides that display potent hepatotoxicity and are tumour promoters. CYLs are alkaloids that are cytotoxic and can cause illness and display hepatotoxic effects in animals and humans. Worldwide, these toxins have caused fatal poisonings to animals, birds and fish. Herceptin, a typical monoclonal antibody, was immobilized on the surface of CdSe/ZnS core-shell quantum dots (QDs) to enhance their specific interactions with breast cancer cells (SK-BR3). the mean size of the core-shell quantum dots (28 nm), as determined by dynamic light scattering, increased to 86 nm after herceptin immobilization. the in vitro cell culture experiment showed that the keratin forming cancer cells (KB) proliferated well in the presence of herceptin-conjugated QDs (QD-Her, 5 nmol/mL), whereas most of the breast cancer cells (SK-BR3) had died. to clarify the mechanism of cell death, the interaction of SK-BR3 cells with QD-Her was examined …

Sutherlandia frutescens (L.) R.Br. (SF) is a South African plant that is widely used to treat stress, infections, cancer, and chronic diseases, many of which involve oxidative stress. The aim of the study was to quantitatively assess the antioxidant potential of SF extracts in cell-free system as well as in cell lines. Dried SF vegetative parts were extracted using six different solvents, and the extracts were assessed for total phenolic and flavonoid contents, total reducing power, iron chelating capacity, and free radical scavenging power, including, scavenging of hydroxyl radicals, superoxide anions, nitric oxide, and hydrogen peroxide. The hyporheic zone is the subsurface area below and adjacent to a stream where groundwater mixes with stream water, through vertical, lateral, and longitudinal flows. The hyporheic zone connects the stream to uplands and other terrestrial environments. It is a zone of distinct faunal communities, high biological diversity and ecological complexity, and is the site of chemical processing and transformations of ground- and stream waters. The molecular characteristics of aerosol organic matter (OM) determines to a large extent its impacts on the atmospheric radiative budget and ecosystem function in terrestrial and aquatic environments, yet the OM molecular details of aerosols from different sources are not well established. Mass spectrometry is being increasingly used in biomarker research mainly due to its ability to achieve high selectivity coupled with high sensitivity. This dissertation focuses on quantitative mass spectrometric investigations of two protein biomarkers i.e. serum thymidine kinase 1 (TK1) and human osteopontin (OPN).

Determination of trace amounts of uric acid (UA) in serum is of great importance since its level in serum may show disorders in purine metabolism. With this method, trace amounts of UA in blood serum can be determined by a fast and simple procedure using differential pulse polarography (DPP). The limit of detection was estimated to be 2 \times 10^{-7} M (S/N = 3), lower than that given in previous studies. Four 6-aminopenicillanic acid moieties were grafted at either rim of calix[4]arene, giving 2 novel generations of penicillin, which were named calixpenam. Antibiotic tests showed that they have amplified activity with respect to the corresponding penams against 3 gram-positive nonpenicillinase-producing strains of Streptococcus.

New 2-(imidazo[1,2-a]pyridin-2-ylcarbonyl)-N-substituted hydrazinecarbothioamides (4a--j), N'-(3-substituted-4-oxo-1,3-thiazolidin-2-ylidene)imidazo[1,2-a]pyridine-2-carbohydrazides (5a--f), and N-(nonsubstituted/4-substituted phenyl)-5-(imidazo[1,2-a]pyridine-2-yl)-1,3,4-oxadiazole-2-amines (6a--d) were synthesized from imidazo[1,2-a]pyridine-2-carbohydrazide (3) and evaluated for antifungal activity against Microsporum gypseum NCPF 580, M. canis, Trichophyton tonsurans NCPF 245, T. rubrum, Candida albicans ATCC 10231, and C. parapsilosis ATCC 22019 using amphotericin B as the standard. The chemical structures of the compounds were confirmed by elemental analysis, IR, ^1H NMR, ^{13}C NMR, HMBC (^{13}C, ^1H), and mass spectra. This study was the first fatty acid analysis and biological activity report on an endemic Achillea species, Achillea cappadocica Hausskn. et Bornm. The major compounds in the fatty acid were identified as oleic (34.7 %), palmitic (23.1 %), and linoleic acids (20.6 %). The methanol and water extracts indicated higher ABTS cation radical scavenging activity than that of \alpha-tocopherol and BHT at 100 \mu g/mL. The acetone extract showed 70.62 % inhibitory activity against butyrylcholinesterase at 200 \mu g/mL. The acetone and methanol extracts exhibited moderate antimicrobial activity.

Three new polysubstituted isoindole-1,3-diones were prepared from 2-ethyl-5-hydroxy-3a,4,5,7a-tetrahydro-isoindole-1,3-dione. The reaction of 2-ethyl-5-hydroxy-3a,4,5,7a-tetrahydro-isoindole-1,3-dione with m-CPBA gave the corresponding epoxide. The triacetate derivative was obtained via cis-hydroxylation using OsO_4, followed by acetylation. An aromatic derivative, a secondary reaction product, was also formed during the acetylation. Finally, a tricyclic derivative from 2-ethyl-5-hydroxy-3a,4,5,7a-tetrahydro-isoindole-1,3-dione was synthesized via dichloroketene addition under microwave irradiation. The exact structures of epoxide and tricyclic derivatives were determined by X-ray diffraction analysis.

The inclusion complexation of flavanone with \beta-cyclodextrin was studied by ultraviolet absorption, steady state fluorescence, time-resolved fluorescence, and 2D ROESY nuclear magnetic resonance spectroscopic techniques. A 1:1 stoichiometric ratio was determined for the inclusion of flavanone with \beta-cyclodextrin. The Stern--Volmer constant for the accessible fraction of the binding of flavanone with \beta-cyclodextrin, and the binding constant for the flavanone--\beta-cyclodextrin complex are reported. The flavanone--\beta-cyclodextrin inclusion complex was characterized by 2D ROESY NMR spectroscopy. A new capillary electrophoresis method was developed for the simultaneous determination of dexpanthenol (DEX), lidocaine (LID), and mepyramine (MEP) in pharmaceutical preparations. The best results were obtained using 20 mM (pH 3.0) phosphate buffer as the background electrolyte. Separation was obtained using a fused-silica capillary (75 \mu m internal diameter, 50 cm total length, 41 cm effective length) and a potential of +30 kV at 20 °C. A novel green approach for the synthesis of silver nanoparticles using erythromycin as a reducing/capping agent is presented. Erythromycin-derived silver nanoparticles were characterized by ultraviolet-visible spectroscopy, scanning electron microscopy, high resolution transmission electron microscopy, X-ray powder diffraction, and Fourier transform infrared spectroscopy. Monodispersed silver nanoparticles showed excellent and promising catalytic activity for reduction of 3 differently charged dyes (eosin B, methylene blue, and rose bengal) in the presence of NaBH_4. Rosmarinic acid is a water-soluble ester of caffeic acid and 3,4-dihydroxyphenyllactic acids, and is mainly found in plant species including Boraginaceae and Lamiaceae. In this research, we determined the inhibition property of rosmarinic acid on carbonic anhydrase isoenzymes I and II (hCA I and II) purified from human erythrocytes by using Sepharose-4B affinity column chromatography. hCA I and II isoenzymes were obtained with a yield of 57.9% and 67.2% and 76.5- and 509.3-fold purification of each isoenzyme, respectively. The synthesis of novel zinc, cobalt, indium, and metal-free phthalocyanines carrying four 3-(4-phenyloxy)couma- rins in the periphery/nonperiphery were prepared by cyclotetramerization of 3-[4-(3,4-dicyanophenyloxy)phenyl]coumarin (2)/3-[4-(2,3- dicyanophenyloxy)phenyl]coumarin (3). The novel chromogenic compounds were characterized by elemental analysis, ^1H NMR, mass spectra, F-IR, and UV-vis spectral data. Here, liquid-exfoliated graphene is functionalized with p-aminophenyl moiety using p-nitrophenyl diazonium salts which were diazotized in situ then reduced by tin(II) chloride. The aminophenyl-graphene thus produced is further modified to form polyaniline-graphene (PANI-GNH) by in situ oxidative graft polymerization of aniline using ammonium peroxydisulfate as oxidizing agent. The properties of the PANI-GNH were compared with polyaniline/graphene blends by Raman, infrared and UV-Visible spectroscopy, and cyclic voltammetry. Parkinson's disease (PD) is the second most common neurodegenerative disorder after Alzheimer's disease, affecting approximately 0.3% of the total U.S. population, and its prevalence increases with age. Two neuropathological hallmarks of PD are the loss of dopaminergic neurons in the substantia nigra pars compacta, a region in the midbrain involved in initiating and sustaining movement, and the presence of cytosolic inclusions called Lewy bodies (LBs) in various brain regions. LBs are enriched with fibrillar forms of the presynaptic protein &agr;-synuclein (aSyn). A discrete, heterobimetallic Tb3+-Eu3+ -containing complex has been synthesized, and energy transfer was observed in the complex without using an antenna. This result represents a new way of observing lanthanide emission using lanthanide excitation in discrete, soluble complex. The occurrence of energy transfer in the mixed heterobimetallic Tb3+-Eu3+ complex was characterized using steady state luminescence spectroscopy and luminescence-decay studies. These findings are likely to be useful for fundamental mechanistic studies of energy transfer in lanthanide-containing systems.

The Dimerization Initiation Sequence (DIS) is a conserved hairpin-loop motif on the 5' UTR of the HIV-1 genome. It plays an important role in genome dimerization through formation of a "kissing complex" intermediate between two homologous DIS sequences. This bimolecular kissing complex ultimately leads to the formation of an extended RNA duplex. Understanding the kinetics of this interaction is key to exploiting DIS as a possible drug target against HIV. Chronic kidney disease (CKD) is known as a heterogeneous disorder which currently is on the rise and lately has been classified as a public health issues in the United State and worldwide. CKD is an irreversible and progressive disease which can lead to kidney failure, and this is depicted by the advanced stage of the disorder when it reaches the point, that is classified as end stage of renal disease (ESRD) (Stage 5 of CKD) (eGRF <15 mL/min/ 1.73 m2 working capacity), where both organs are in a total or permanent kidney failure. End-Stage renal disease patients, on hemodialysis have been associated to experience an accelerated form of atherosclerosis, which is induced by inflammation, impairment of antioxidant system and elevated oxidative stress. Since the problem effecting ESRD patients is multifactorial, the objective of this investigation is to explore and look at the effects of supplementing with vitamin E-tocotrienol rich fraction (TRF), a micronutrient which has anti-inflammatory, antioxidant, and lipid lower capabilities into tackling these comorbid conditions experienced by this population. Therefore the aims of this investigation will be to explore changes in lipid profiles, inflammatory markers, and oxidative status, as well as look at any changes in metabolomic profiles. It was hypothesized that by supplementing with TRF a vitamin E, for 16 weeks in ESRD patients undergoing hemodialysis, it may help reverse and/or improve, oxidative status, inflammatory markers, increase antioxidants status and improve lipid profiles.

Multidrug membrane transporters (efflux pumps) can selectively extrude a variety of structurally and functionally diverse substrates (e.g., chemotoxics, antibiotics), leading to multidrug resistance (MDR) and ineffective treatment of a wide variety of diseases. In this study, we have designed and constructed a fusion gene (egfp-mexB) of N-terminal mexB with C-terminal egfp, inserted it into a plasmid vector (pMMB67EH), and successfully expressed it in the Δ MexB (MexB deletion) strain of Pseudomonas aeruginosato create a new strain that expresses MexA-(EGFP-MexB)-OprM.
