Isoquinoline-Substituted Triazole And Pyran Derivatives: Synthesis And Computational Studies, Duygu Yeni̇dede, Selçuk Gümüş, Ayşegül Gümüş
Turkish Journal of Chemistry
The one-pot synthesis of novel 1,4-disubstituted 1,2,3-triazoles from isoquinoline-substituted homopropargyl alcohol backbone is described (42%-88% yields). A ring closing metathesis reaction and an intramolecular Pauson-Khand reaction of enyne system derived from a homopropargyl alcohol backbone to afford the corresponding isoquinoline-substituted dihydropyran and cyclopentenone-pyran, respectively, are also described (54% and 78% yields). Information about the structural, electronic, and physico-chemical properties of the novel compounds, obtained by density functional theory application, is also reported.

A fluorescent probe based on benzoyl hydrazine was synthesized and characterized as an Al$^{3+}$-selective fluorescent probe. This probe showed good selectivity towards Al$^{3+\, }$compared to other common ions. Under optimized experimental conditions, the probe exhibited a linear dynamic response for Al$^{3+}$ from 5.0 $\times $ 10$^{-7}$ to 4.5 $\times $ 10$^{-6}$ M with a detection limit of 1.3 $\times $ 10$^{-7}$ M in ethanol-water solution (9:1, v:v, pH 6.8, 20 mM HEPES). Furthermore, it was used for imaging of Al$^{3+}$ in living cells with satisfying results.

New furanoside-based NHC precursor salts (2) were synthesized using amino alcohols from the chloralose derivatives of glucose (a), galactose (b), and mannose (c). The novel compounds were fully characterized by $^{1}$H NMR, $^{13}$C NMR, and elemental analyses. The catalytic activities of these salts were tested in the arylation of aldehydes as catalysts that were generated in situ from [RhCl(COD)]$_{2}$. In addition, 2a was converted to the rhodium complex 3a in order to compare the results obtained in situ. The newly synthesized compounds were very efficient in terms of yield; nevertheless they did not exhibit a chiral induction.

The $N$-heterocyclic carbene (NHC)--Ag(I) complexes 3a-c were synthesized from benzimidazolium chlorides (2a-c = NHC.HCl), incorporating benzyl derivatives and a 2-methoxyethyl group (a: $^{3}$N = 2,4,6-trimethylbenzyl; b: $^{3}$N = 2,3,5,6-tetramethylbenzyl; c: $^{3}$N = pentamethylbenzyl). The compounds were characterized by spectroscopic and elemental analyses. The synthesized NHC-Ag(I) complexes (3a-c) were tested as catalysts for the catalytic three-component coupling reaction of aldehyde, alkyne, and amine to propargylamines in various solvents. All complexes were active catalysts for catalytic three-component coupling reactions with good yields under neat and mild conditions (after 4 h, yields of up to 94%).

L-cysteine-capped Mn-doped ZnS quantum dots (QDs) were used for the determination of glutamic acid in foodstuffs. This method is based on measurement of the quenching of the phosphorescence intensity of the QDs after interacting with glutamic acid. A linear response was observed from 50 to 500 ng mL$^{-1}$ glutamic acid with a limit of detection of 6.79 ng mL$^{-1}$. Room temperature phosphorescence (RTP) intensity of the QDs was quenched rapidly upon the addition of the quencher and the reaction reached equilibrium within 2 min. A carboxymethylcellulose (CMC)-degrading bacterium was isolated from soil, identified as Bacillus methylotrophicus according to the physiological properties and analyses of 16S rRNA and a partial sequence of the gyrase A (gyrA) gene, and named as B. methylotrophicus Y37. The CMCase enzyme was purified to homogeneity by 20.4-fold with 21.73% recovery using single-step hydrophobic interaction chromatography and biochemically characterized. CMCase showed a molecular weight of approximately 50 kDa as determined by SDS-PAGE. The activity profile of the CMCase enzyme exhibited optimum activity at 45 $^{\circ}$C and pH 5.0. This review focuses on the speciation analysis of metals, metalloids, and organometals by solid phase extraction before spectrometric detection, and all aspects of analytical speciation such as determination of oxidation states and organometallic compounds. A series of novel positional oximes (2a-2d), $O$-methyl oxime ethers (3a-3d), and $O$-benzyl oxime ethers (4a-4d) were synthesized in high yields starting from their corresponding methyl 3-, 4-, 6-, and 13-keto tetradecanoates. The synthesized compounds were characterized by $^{1}$H NMR, $^{13}$C NMR, FT-IR, mass, and elemental analyses for their structures and $(E)$/$(Z)$ isomerizations. Their DNA binding abilities were investigated in vitro by agarose gel electrophoresis. The antibacterial and antifungal activities were tested also in vitro against eleven bacterial strains and three fungal strains. This study is focused on the synthesis of chromate anion imprinted sorbent supported on silica gel for nonchromatographic Cr speciation in surface waters. The preparation procedure is based on grafting of 3-methyl-1-trimethoxysilylpropylimidazolium, preliminarily coordinated to CrO$_{4}^{2-}$ as a template ion, onto the surface of silica gel. Sorption and desorption characteristics of surface-imprinted sorbent toward Cr(III) and Cr(VI) were examined by batch solid-phase extraction. An excellent separation of Cr(VI), selectively retained on the sorbent, from Cr(III) remained in the solution, was achieved at pH 2-3 for 20 min. Hexavalent chromium, Cr(VI), is a highly toxic species, while trivalent chromium, Cr(III), is an essential trace element for humans and other organisms. In the present work, a simple, rapid, and efficient procedure for water treatment of Cr(VI) is described. Active carbon (AC) was chemically treated with HCl and NaOH for the formation of surface modified Acidic-AC and Basic-AC sorbents, respectively. Cloud point extraction (CPE), which is the most popular coacervate-based extraction method, was utilized for the separation and preconcentration of trace amounts of cadmium, lead, and nickel. Quinalizarin was used as complexing agent and Triton X-114 was used as surfactant. After extraction, analyte ions were determined by flame atomic absorption spectrometry. The detection limits (3$s)$ of 1.8 $\mu $g L$^{-1}$ for Cd(II), 3.2 $\mu $g L$^{-1}$ for Pb(II), and 2.8 $\mu $g L$^{-1}$ for Ni(II) were obtained using 50 mL of solutions. This study reports 2 new simple derivatization-based dispersive liquid-liquid microextraction (DLLME) methods for spectrophotometric ultratrace determination of asulam and sulfide. 1-Naphthol (in the presence of nitrite) and N,N-diethyl-p-phenylenediamine (in the presence of Fe(III)) were used to derivatize asulam and sulfide, respectively. In the enrichment methods, the formed derivatives were preconcentrated into microdroplets of the in situ formed water insoluble ionic liquid (IL), 1-hexyl-3-methylimidazolium hexafluorophosphate. Monitoring was performed at 526 nm for asulam and at 664 nm for sulfide, after dissolution of the IL-rich phases into the basic ethanolic solution and ethanol for asulam and sulfide, respectively. The title compounds C17H14BrNO2, (I), and C17H15NO3, (II), were obtained from the reaction of 6-methoxy-3,4-dihydro-2H-naphthalen-1-one and 2-bromonicotinaldehyde in ethanol. Compound (I) was the expected product and compound (II) was the oxidation product from air exposure. In the crystal structure of compound (I), there are no short contacts or hydrogen bonds. The structure does display [pi]-[pi] interactions between adjacent benzene rings and adjacent pyridyl rings. Compound (II) contains two independent molecules, A and B, in the asymmetric unit; both are non-planar, the dihedral angles between the methoxybenzene and 1H-pyridin-2-one mean planes being 35.07 (9)° in A and 35.28 (9)°in B. This research was performed to examine the physiological and biochemical effects of bare (nCeO2) and citric acid coated (nCeO2+CA) cerium oxide nanoparticles in tomato (Solanum lycopersicum). Plants were germinated and grown to full maturity in soil amended with 0-500 mg/kg and periodically analyzed using ICP-OES/MS and other spectroscopic and biochemical techniques. Although Ce in tissues increased as the external nCeO2 increased, there were no visible signs of toxicity. In mature plants, nCeO2 induced developmental changes and altered nutritional quality (essential elements) in fruits. On the other hand, nCeO2+CA affected fruit carbohydrate composition (sugars and starch). This study gives insights on …

The use of cell penetrating peptides (CPPs) as biomolecular delivery vehicles holds great promise for therapeutic and other applications, but development has been stymied by poor delivery and lack of endosomal escape. We have developed a CPP-adaptor system capable of efficient intracellular delivery and endosomal escape of user-defined protein cargos. The cell penetrating sequence of HIV transactivator of transcription was fused to calmodulin, which binds with subnanomolar affinity to proteins containing a calmodulin binding site. Our strategy has tremendous advantage over prior CPP technologies because it utilizes high affinity noncovalent, but reversible coupling between CPP and cargo. Three different cargo …

Two-photon microscopy is a fluorescence imaging technique which provides distinct advantages in three-dimensional cellular and molecular imaging. The benefits of this technology may extend beyond imaging capabilities through exploitation of the quantum processes responsible for fluorescent events. This study utilized a two-photon microscope to investigate a synthetic photoreactive collagen peptidomimetic, which may serve as a potential material for tissue engineering using the techniques of two-photon photolysis and two-photon polymerization. Additive manufacturing (AM), now more commonly known as 3D printing, has been classified as efficient, fast, and practical in the prototyping sector of product development. In the work presented here, we will use one of the AM techniques known as Material extrusion 3D printing (ME3DP), which has all the advantages of AM. The protozoan parasite, Trypanosoma cruzi, the etiologic agent of Chagas disease (ChD), has a cell surface covered by immunogenic glycoconjugates. A synthetic glycoarray containing non-reducing α-galactopyranosyl moieties related to mucin O-glycans of the parasite T. cruzi was evaluated by a chemiluminescent enzyme-linked immunosorbent assay (CL-ELISA) with sera from patients with chronic ChD, revealing the disaccharide Galα(1,3)Galβ as the immunodominant glycotope (Chapter 2). This disaccharide comprises the two terminal sugars of the known immunodominant glycotope Galα(1,3)Galβ(1,4)GlcNAcα, which is expressed on glycosylphosphatidylinositol-anchored mucins of the infective trypomastigote stage of T. cruzi and triggers high levels of protective anti-α-Gal antibodies (Abs) in infected …

It is imperative to not only understand the impact of engineered nanomaterials (ENMs) in edible plants, but also the interactions they have with various additives used in agriculture. In this study, kidney bean plants were grown in potting soil treated with nano copper (nCu), bulk copper (bCu), and copper chloride (CuCl2) at concentrations of 0, 50, and 100 mg/kg for 55 and 90 days. At 15 days of growth, 0, 10, and 100 μm of kinetin (KN) were applied to plants. Plant tissue samples were harvested at 55 days and seeds were reaped at 90 days. Persistent organic pollutants (POPs) are toxic chemicals that adversely affect the health of human populations as well as the environment. The need to develop remediation methods and materials as well as understand the mechanisms in which they can degrade POPs is necessary. Virophages are satellite-like dsDNA viruses that parasitize giant viruses of the family Mimiviridae. Mavirus is the second virophage discovered that associates with its host virus Cafeteria roenbergensis Virus (CroV). When co-infecting their common host cell Cafeteria roenbergensis, a marine zooplankton that is widely spread throughout the oceans, mavirus will inhibit CroV's replication. In addition, mavirus was shown to share high similarities to the Maverick/Polinton eukaryotic DNA transposons. A coding sequence in mavirus genome (MV02) reveals high homology to retroviral integrases such as those found in HIVs. The putative integrase MV02 is predicted to integrate mavirus DNA into the host genome. …

Chagas disease, caused by the parasite Trypanosoma cruzi, is an endemic life-threatening disease that affects mainly the heart. It remains the leading cause of heart failure in Latin American countries. Since current treatments against this parasite are highly toxic and somewhat ineffective, novel and more efficacious types of interventions are desired. The oxidation of N-acetylthiourea (ACTU) by acidic bromate has been studied by observing formation of bromine in excess bromate conditions. The reaction displays an induction period before formation of bromine. The stoichiometry of the reaction was determined to be 4:3: 4BrO 3 – +3(CH 3 CO)NH(NH 2 )C=S+3H 2 O®4Br – +3(CH 3 CO)NH(NH 2 )C=O+3SO 4 2– +6H + (A) with a complete desulfurization of ACTU to its urea analogue. One main aspect of high energy density material (HEDM) design is to obtain greener alternatives for HEDMs that produce toxic byproducts. Primary explosives lead azide, lead styphnate, and mercury fulminate contain heavy metals that cause heavy metal poisoning. Leaching of the widely used tertiary explosive NH4ClO4 into groundwater has resulted in human exposure to ClO4– ions, which cause disruptions of thyroid related metabolic pathways and even thyroid cancer. Detection of trace metals has great importance in environmental and biological applications. While traditional electrochemical techniques have played critical roles in this field, their usefulness is limited by temporal resolution, Hg toxicity and stability concerns. Recently, we developed a method using fast-scan cyclic voltammetry (FSCV) at carbon-fiber microelectrodes (CFMs) to achieve rapid measurement of metal ions with high sensitivity, selectivity, and stability. Through optimizations this method showed strengths in real-time trace metal analysis.

The first step in the biosynthesis of nearly all plant membrane phospholipids and storage triacylglycerols is catalyzed by a glycerol-3-phosphate acyltransferase (GPAT). The requirement for an endoplasmic reticulum (ER)-localized GPAT for both of these critical metabolic pathways was recognized more than 60 years ago. However, identification of the gene(s) encoding this GPAT activity has remained elusive. Here, we present the results of a series of in vivo, in vitro, and in silico experiments in Arabidopsis (Arabidopsis thaliana) designed to assign this essential function to AtGPAT9.
