We developed a new fluorogenic bioorthogonal reaction that is based on the inverse electron-demand Diels–Alder reaction between styrene (an unstrained alkene) and a simple tetrazine. The reaction forms a new fluorophore with no literature precedent. We have identified an aminoacyl-tRNA synthetase/tRNA pair for the efficient and site-specific incorporation of a styrene-containing amino acid into proteins in response to amber nonsense codon. Fluorogenic labeling of purified proteins and intact proteins in live cells were demonstrated. The fluorogenicity of the styrene–tetrazine reaction can be potentially applied to the study of protein folding and function under physiological conditions with low background fluorescence interference.

Little information exists on the occurrence and the ultimate fate of pharmaceuticals in the water bodies in India despite being one of the world leaders in pharmaceutical production and consumption. This paper has reviewed 19 published reports of pharmaceutical occurrence in the aquatic environment in India [conventional activated sludge wastewater treatment plants (WTPs), hospital WTPs, rivers, and groundwater]. Carbamazepine (antipsychoactive), atenolol (antihypertensive), triclocarban and triclosan (antimicrobials), trimethoprim and sulfamethoxazole (antibacterials), ibuprofen and acetaminophen (analgesics), and caffeine (stimulant) are the most commonly detected at higher concentrations in Indian WTPs that treat predominantly the domestic sewage. Molecular dynamics (MD) simulations and computational chemistry allow for an atomistic understanding of protein-protein and protein-ligand binding motifs. Through the use of MD, medicinally relevant complexes can be examined in detail unattainable by experimental methods. Within this work, systems pertinent to both Alzheimer’s Disease and HIV-1 are probed and thoroughly sampled to help elucidate potential therapeutic pathways. We used molecular dynamics and free energy estimations to gauge the affinity for the binary and ternary complexes of KLC1, APP and JIP1, three proteins all believed to be involved in the propagation of Alzheimer’s Disease. Recent studies reveal 2-aminoadipic acid (2-AAA) is both elevated in subjects at risk for diabetes and mechanistically linked to glucose homeostasis. Prior studies also suggest enrichment of protein-bound 2-AAA as an oxidative post-translational modification of lysyl residues in tissues associated with degenerative diseases of aging. While in vitro studies suggest redox active transition metals or myeloperoxidase (MPO) generated hypochlorous acid (HOCl) may produce protein-bound 2-AAA, the mechanism(s) responsible for generation of 2- AAA during inflammatory diseases are unknown. A novel chemosensor for the detection of inorganic phosphate (Pi) in environmental water samples is outlined. The sensing solution is comprised of a luminescent lanthanide, terbium (Tb3+), chelated to ethylenediaminetetraacetic acid (EDTA) acid in solution with cetyltrimethylammonium bromide (CTAB)- capped gold nanoparticles (AuNPs). The Tb-EDTA and AuNPs undergo a Fluorescence resonance energy transfer (FRET) mechanism in which the Tb3+ luminescence is quenched. Upon the addition of inorganic phosphate (Pi), the AuNPs begin to aggregate and precipitate out of solution. The aggregation of AuNPs results in the restoration of the Tb-EDTA signal which can then be correlated to Pi concentration in …

Some of this damage may not be able to be repaired by cellular machineries causing DNA replication to be blocked. Autophagy is a cellular process conserved in eukaryotes that involves the trafficking of intracellular material from the cytosol to the vacuole/lysosome. This material is wrapped in a double membraned vesicle called an autophagosome, which ls constructed at the pre-autophagosomal structure. The emergence of multidrug-resistant bacterial and fungal strains poses a threat to human health that requires the design and synthesis of new classes of antimicr obial agents. We evaluated bis(N-amidinohydrazones) and N-(amidino)-N'-aryl-bishydrazones for their antibacterial and antifungal activities against panels of Gram-positive/Gram-negative bacteria as well as fungi. We investigated their potential to develop resistance against both bacteria and fungi by a multi-step, resistance-selection method, explored their potential to induce the production of reactive oxygen species, and assessed their toxicity. A compound was specifically designed to further aid research on the detection and treatment of folate receptor positive cancer cells. A small but complex peptide chain was commercially produced to have a flexible glycine spacer and a six-histidine tag. Histidine tagging was chosen for its ability to bind to nickel. The compound in this report was purified and analyzed by HPLC and confirmed via mass spectrometry. In this work, microfluidic systems coupled with Raman detection for analytical applications were developed using both off-chip and on-chip synthesized nanoporous Ag microstructures (np-AgMSs) as SERS substrates. Off-chip synthesized np-AgMSs were used as on-chip SERS substrates for determination of glutathione (GSH) using a reversed reporting technique. The np-AgMSs were drop casted onto the cross channel of a microfluidic device. R6G (a Raman reporter) interacted with the embedded np-AgMSs in the microchannel to form R6G-np-AgMSs which provided high SERS signal. Then, R6G in the R6G-np-AgMSs complex was replaced by GSH, causing the decrease in SERS signal which was related to the …

Perchlorate (ClO4) in the environment is of concern, because of potential health risks to humans, among other reasons. Evidence suggests that the majority of environmental perchlorate is formed in the atmosphere (likely in the stratosphere), in chemical processes involving ozone and stratospheric chlorine. Synthesis and characterization of 1-ethyl-3- methylimidazolium alkylbenzenesulfonate ionic liquids for extraction of lignin from prairie cord grass have been studied. The ionic liquids (ILs) 1-ethyl-3- methylimidazolium benzenesulfonate (EBS), 1-ethyl-3- methylimidazolium toluenesulfonate (ETS) and 1-ethyl-3- methylimidazolium xylenesulfonate (EXS) have been synthesized in this research. An extensive structural, physical, thermal and toxicological characterization has been performed to understand the behavior of these ionic liquids. The reaction yield for the synthesis of EBS, ETS, and EXS ionic liquids was determined to be 91.2 ± 1.5 %, 96.1 ± 0.7 %, 99.0 ± 0.5 % respectively. Spectral analysis using NMR and FTIR confirms the …

Certain foundational concepts, including acid-base theory, chemical bonding and intermolecular forces (IMFs), appear throughout the undergraduate chemistry curriculum. The level of understanding of these foundational concepts influences the ability of students to recognize the relationships between sub-disciplines in chemistry. The purpose of this study was to investigate the relationship between student attitudes towards chemistry and their abilities to recollect and transfer knowledge of IMFs, a foundational concept, to their daily lives as well as to other classes. Lignin forms about 30% of lignocellulosic material, and is the second most abundant non-fossil organic carbon source in the biosphere. Chirality (or handedness) is the property that a structure is “different” from its mirror image. Topology can be used to provide a rigorous framework for the notion of chirality. This project examines various types of chirality and discusses tools to detect chirality in graphs and knots. Notable theorems that are discussed in this work include ones that identify chirality using properties of link polynomials (HOMFLY polynomials), rigid vertex graphs, and knot linking numbers. Physicochemical properties of coordination compounds can be exploited for molecular recognition of biomolecules. The inherent π-π stacking properties of [Pt(chelate)(N-donor)]2+([PtN4]) complexes were modulated by systematic variation of the chelate (diethylenetriamine and substituted derivatives) and N-donor (nucleobase or nucleoside) in the formally substitution-inert PtN4 coordination sphere. Macrophage colony stimulating factor receptor (CSF-1R or MCSFR) is a receptor tyrosine kinase essential for the growth and function of macrophages. Understanding the mechanisms that regulate CSF-1R activation and deactivation will provide insights to clinical treatment of macrophage related diseases including chronic inflammation and cancer. Previously, our laboratory showed that CSF-1R undergoes a novel membrane trafficking route that involves macropinocytosis to deactivate CSF-1R signaling. This thesis makes the discovery that the ubiquitin ligases Cbl and Cbl-b cooperate to regulate CSF-1R endocytosis and traffic to macropinosome in macrophages. Cyanide poisoning by accidental or intentional exposure poses a severe health risk. The current FDA approved antidotes for cyanide poisoning can be effective, but each suffers from specific major limitations. Dimethyl trisulfide (DMTS), a sulfur donor that detoxifies cyanide by converting it into thiocyanate, is a promising next generation cyanide antidote. Although a validated analytical method to analyze DMTS is not currently available from any matrix, one will be vital for the approval of DMTS as a therapeutic agent against cyanide poisoning. Dihydrofurans serve as building blocks for other compounds in organic synthesis. The main goal of this project was to discover an efficient and relatively inexpensive pathway for producing monosubstituted dihydrofurans in high yield from cyclic boronic half acids. Aldehydes were converted to homoallylic alcohols by the addition of allylmagnesium bromide. The alcohols were then transformed into cyclic boronic half acids using ringclosing metathesis with Grubbs 1st Generation Catalyst and alkenyl boronic esters. Finally, monosubstituted dihydrofurans were produced using a palladium (II) catalyst with a base. Within the context of North American Indigenous culture, certain Elders are respected gatekeepers to Indigenous science, also known as traditional knowledge. Yet, while North American born minorities such as Black Americans, Amerindians, and Latin Americans may hail from cultures with a similar appreciation of their own Indigenous science Elders, these minority groups are especially underrepresented in Western science, technology, engineering, and math (STEM)—both in academia and in the workforce. North American underrepresented minorities experience high attrition rates in academia generally, and in STEM specifically. As the K to 12 Science program was formally implemented, interventions to enhance competence and confidence of teachers in teaching science in a spiral progression approach are main concerns. This study aims to assess the chemistry content knowledge and self-efficacy of 38 in-service teachers enrolled in a graduate program from a teacher education institution using a content knowledge test (CKT) and a self-efficacy beliefs scale (SeS) using a mixed-method approach. Quantitative findings reveal that the least mastered topics in chemistry of the teacher-respondents include solutions, chemical bonding, the mole concept, gas laws, and chemical reactions. Quinolinol-based compounds are a promising starting point for discovery o f effective inhibitors o f the clostridial neurotoxin, botulinum neurotoxin type A light chain (BoNT/A LC). Insights into the mechanism o f inhibition by quinolinol compounds facilitate interpretation o f docking data and inhibitor optimization. In this study, a fluorogenic substrate o f BoNT/A, SNAPtide, was used to study the mechanism by which two new quinolinol compounds, MSU58 and MSU84, with IC50 values of 3.3 uM and 5.8 uM, respectively, inhibit BoNT/A LC. (-)-Epigallocatechin gallate (EGCG) is a polyphenolic natural product that inhibits the key chaperone protein Hsp90 by binding to the carboxy terminal portion of the protein. The natural product has modest activity as an Hsp90 inhibitor and there is little information in the literature on structure activity relationships for EGCG analogs. Previous studies from this laboratory provided initial data on the effect of a limited range of changes associated with functional groups, substituents and stereochemistry.
