Synthesis Of 1,2-Naphthoquinone Derivatives As Α-Glucosidase Inhibitors, Trang Pham Thi

For a couple of decades the bioactivity of quinone has been considered as privileged structure-based drug design. In this research, 4-phenylamino, 4-alkylamino, 4-alkylphenylamino, 4-phenoxy and other derivatives of 1,2-naphthoquinone were synthesized, evaluated for their anti α-glucosidase activity and elucidated for their structure-activity relationship. The results indicated that multiple factors affecting to the α-glucosidase inhibition included π-π interaction, hydrophobic interaction and hydrogen bonding with the enzyme. In general, the main factor affecting the activity was the more favorable 1,2-dione production. The presence of electron withdrawing substituents on the phenyl ring and heteroatom linker bound to the quinone skeleton increased the activity. …

Diabetes mellitus occurs from deficiencies in insulin secretion or insulin resistance which leads over time to high risk for long-term macro-and microvascular complications. Diabetes mellitus type 2 contributes to more than 90% of all cases worldwide. One approach of treatment for this disorder is utilizing α-glucosidase inhibitors. Certain sulfonamides and chalcones are considered as viable candidates that are effective in reducing hyperglycemia. Forty-seven sulfonamide chalcones were synthesized by the Claisen-Schmidt reaction with various substituents on the A, B, and C-rings furnishing the desired products with 50-95 %yield. This dissertation focused on the development of electrochemical paper-based immunosensor for determining essential biomarkers by using a label-free immunoassay. This dissertation can be divided into four main parts. In the first part, an origami paper-based analytical device (oPAD) was developed for c-reactive protein (CRP) detection. oPAD was designed to combine the preparation of immunosensor and detection step into a single device. A gold nanoparticle modified graphene screen-printed carbon electrode (AuNPs/G/SPCE) was utilized to enhance the sensitivity of the electrode. Mercury ion (Hg2+) is one of the most challenging problems due to its high toxicity at low concentration and bioaccumulative effects affecting human health. Herein, we propose a strategy to exploit the fluorescence quenching of carbon quantum dots (CQDs) combined with surface-enhanced Raman scattering (SERS) technique to detect Hg2+. CQDs exhibit strong fluorescence emission interfering SERS measurement. However, with the presence of Hg2+, fluorescence from CQDs can be quenched, and the SERS spectrum can be collected. ในงานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาการสังเคราะห์คอมพอสิตไคโตซาน-พอลิไวนิลแอลกอฮอล์ (CS:PVA) โดยใช้สารเชื่อมขวาง เตตระเอธิลออร์โธซิลิเกต (TEOS) และกรดออกซาลิก (OA) และการกราฟต์หมู่ฟังก์ชันเอมีนโดยใช้สาร 3-อะมิโนโพรพิลไตรเอทอกซีไซเลน (APTES) ในการกำจัดสีแอคทีฟเรด 120 ในน้ำ โดยตรวจสอบหมู่ฟังก์ชันบนคอมพอสิตด้วยเทคนิคฟลูเรียร์ทรานส์ฟอร์ม อินฟราเรดสเปกโทรสโกปี (FTIR) ปรากฏการเปลี่ยนแปลงหมู่ฟังก์ชันบนคอมพอสิตเมื่อมีการเติมสารเชื่อมขวาง และเมื่อพิจารณาลักษณะสัณฐานวิทยาโดยใช้กล้องจุลทรรศน์อิเล็กตรอนแบบส่องกราด (SEM) แสดงให้เห็นว่าไคโตซานและพอลิไวนิลแอลกอฮอล์สามารถรวมเข้ากันได้ดี และสัดส่วนคอมพอสิตไคโตซาน-พอลิไวนิลแอลกอฮอล์พบว่า ผลของการดูดซับที่สัดส่วนระหว่างไคโตซานและพอลิไวนิลแอลกอฮอล์บนคอมพอสิตและการเติมสารเชื่อมขวางส่งผลต่อการดูดซับโดยร้อยละการกำจัดจะขึ้นอยู่กับพฤติกรรมการบวมตัวของคอมพอสิต การบวมตัวสูงทำให้เวลาในการเข้าสู่สมดุลได้เร็ว และประจุบนพื้นผิวเป็นศูนย์ของคอมพอสิต (PZC) พบว่าการกำจัดสีจะขึ้นอยู่กับประจุที่ผิวคอมพอสิตและประจุของสี สำหรับผลการศึกษาตัวแปรต่าง ๆ พบว่าคอมพอสิต 3CS:7PVA_1.5TEOS_0.5OA_0.1APTES ที่ความเข้มข้นเริ่มต้นของสีแอคทีฟเรด 120 เท่ากับ 100 มิลลิกรัมต่อลิตร ความเป็นกรด-ด่างเท่ากับ 7 ปริมาณคอมพอสิตที่ 0.5 กรัม และที่อุณหภูมิ 30 องศาเซลเซียส ใช้เวลาในการเข้าสู่สมดุลภายในเวลา 120 นาที และมีร้อยละการกำจัดสี 100.0 เมื่อเวลาผ่านไป 480 นาที การเพิ่มความเข้มข้นเริ่มต้นของสีสามารถกำจัดสีได้อย่างสมบูรณ์ และสามารถลดเวลาในการเข้าสู่สมดุลได้ที่ภาวะเป็นกลาง นอกจากนี้การเพิ่มปริมาณคอมพอสิตเป็นการเพิ่มพื้นผิวสัมผัสในการดูดซับ และมีประสิทธิภาพในการดูดซับที่อุณหภูมิปกติ

งานวิจัยนี้ทำการศึกษาการผลิตไฮโดรเจนจากกระบวนการแกซิฟิเคชันด้วยไอน้ำของใบอ้อยโดยใช้ NiO/CaO-Li4SiO4 โดยการทดลองถูกดำเนินการในเครื่องปฏิกรณ์แบบเบดนิ่งชนิดสองขั้น จากผลการทดลองพบว่าการใช้ตัวดูดซับลิเทียมออโทซิลิเกต (Li4SiO4) ร่วมกับตัวดูดซับแคลเซียมออกไซด์ (CaO) ผลได้และความเข้มข้นของไฮโดรเจนมีค่าสูงกว่าการใช้ตัวดูดซับลิเทียมออโทซิลิเกตหรือแคลเซียมออกไซด์เพียงอย่างเดียว บ่งชี้ถึงการทำงานร่วมกันของแคลเซียมออกไซด์และลิเทียมออโทซิลิเกต ปริมาณของแคลเซียมออกไซด์ที่เพิ่มขึ้นส่งผลให้ผลิตภัณฑ์แก๊สที่ได้มีแนวโน้มของผลได้และความเข้มข้นของไฮโดรเจนที่สูงขึ้น เนื่องจากการดูดซับของคาร์บอนไดออกไซด์ที่เพิ่มขึ้น การใช้ตัวเร่งปฏิกิริยา NiO/CaO-Li4SiO4 ให้ผลิตภัณฑ์แก๊สที่มีผลได้และความเข้มข้นของไฮโดรเจนสูงขึ้นเป็นผลมาจากการเร่งปฏิกิริยาการแตกตัวและรีฟอร์มมิงด้วยไอน้ำของสารระเหย การศึกษาผลของอุณหภูมิ พบว่าการเพิ่มขึ้นของอุณหภูมิส่งให้ผลิตภัณฑ์แก๊สรวมมีค่าสูงขึ้นจากการแตกตัวด้วยความร้อนเพิ่มขึ้น แต่ความเข้มข้นของไฮโดรเจนลดลงจากการดูดซับคาร์บอนไดออกไซด์ที่ลดลง นอกจากนี้จากการศึกษาผลของวิธีการเตรียมตัวเร่งปฏิกิริยาที่ต่างกันพบว่าวิธีอิมเพรกเนชันร่วมจะให้ความเข้มข้นของไฮโดรเจนสูงสุด

งานวิจัยนี้ศึกษากัมมันตภาพในการผลิตไฮโดรเจนพร้อมกับการสลายสารอินทรีย์ในน้ำเสียจากการผลิตไบโอดีเซลโดยใช้ตัวเร่งปฏิกิริยาเชิงแสงไทเทเนียมไดออกไซด์เจืออโลหะ ตัวแปรที่ศึกษา คือ ความเข้มข้นเริ่มต้นของน้ำเสีย ชนิดอโลหะ ได้แก่ คาร์บอน (C) ซิลิคอน (Si) และฟอสฟอรัส (P) ที่เจือลงบนตัวเร่งปฏิกิริยาเชิงแสงไทเทเนียมไดออกไซด์ ปริมาณฟอสฟอรัสร้อยละ 1 3 5 7 และ 9 โดยน้ำหนัก และความสามารถในการนำตัวเร่งปฏิกิริยาเชิงแสงกลับมาใช้ซ้ำ พบว่าตัวเร่งปฏิกิริยาเชิงแสง P1/T400ให้กัมมันตภาพในการผลิตไฮโดรเจนและลดค่าซีโอดีสูงกว่าตัวเร่งปฏิกิริยาเชิงแสง C1/T400 และ Si1/T400 เท่ากับ 6.43 มิลลิโมลต่อกรัมตัวเร่งปฏิกิริยา และร้อยละ 26.0 ตามลำดับ ภายใต้ภาวะความเข้มข้นเริ่มต้นของน้ำเสียที่ไม่ผ่านการเจือจาง ปริมาณตัวเร่งปฏิกิริยา 4 กรัมต่อลิตร ความเข้มแสง 5.93 มิลลิวัตต์ต่อตารางเซนติเมตร ที่อุณหภูมิห้อง เป็นเวลา 4 ชั่วโมง เนื่องจากตัวเร่งปฏิกิริยาเชิงแสง P1/T400 มีแถบช่องว่างพลังงานที่แคบและมีตำแหน่งแถบเวเลนซ์และแถบนำในโครงสร้างที่เหมาะสมต่อการผลิตโปรตอน (H+) และตัวออกซิไดซ์ (HO• และ O2•-) ซึ่งส่งผลต่อการผลิตไฮโดรเจนและการลดค่าซีโอดี ปริมาณฟอสฟอรัสที่เจือลงบนตัวเร่งปฏิกิริยาเชิงแสง T400 ที่ให้กัมมันตภาพในการผลิตไฮโดรเจนและลดค่าซีโอดีสูงที่สุด ได้แก่ P7/T400 โดยสามารถผลิตไฮโดรเจนได้สูงถึง 8.34 มิลลิโมลต่อกรัมตัวเร่งปฏิกิริยา และลดค่าซีโอดีได้เท่ากับร้อยละ 50.6 นอกจากนี้ตัวเร่งปฏิกิริยาเชิงแสง P7/T400 ยังคงให้กัมมันตภาพในการผลิตไฮโดรเจนและลดค่าซีโอดีสูงกว่าตัวเร่งปฏิกิริยาเชิงแสง T400 แม้ผ่านการนำกลับมาใช้ซ้ำ 4 ครั้ง

แบตเตอรี่สังกะสีไอออนแบบประจุกลับได้ฐานน้ำ (aqueous rechargeable zinc-ion batteries, ARZIBs) เป็นเทคโนโลยีกักเก็บพลังงานที่ได้รับความสนใจอย่างแพร่หลายเนื่องจากขั้วที่ทำจากสังกะสีมีความจุพลังงานสูง มีความปลอดภัยในการใช้งานสูง เป็นมิตรต่อสิ่งแวดล้อม และใช้สังกะสีเป็นวัตถุดิบที่ต้นทุนต่ำและพบง่ายในประเทศ อย่างไรก็ตาม การใช้งานจริงของแบตเตอรี่ประเภทนี้ยังมีข้อจำกัดของอายุการใช้งานที่สั้นและมีประสิทธิภาพในการคายประจุที่ต่ำ ซึ่งส่วนใหญ่เป็นผลมาจากการเกิดการพอกพูนสังกะสีแบบกิ่งก้าน (zinc dendrites) บนขั้วแอโนด งานวิจัยนี้ศึกษาผลของสารเติมแต่งในอิเล็กโทรไลต์ฐานน้ำต่อสมรรถนะและอายุการใช้งานของแบตเตอรี่สังกะสีไอออนแบบประจุกลับได้ จากการศึกษาผลของการเติมเอทิลีนคาร์บอเนต (ethylene carbonate, EC) ในอิเล็กโทรไลต์ฐานน้ำโดยมี ZnSO4 ความเข้มข้น 2 โมลาร์และ MnSO4 ความเข้มข้น 0.5 โมลาร์เป็นเกลือ พบว่าการเติมเอทิลีนคาร์บอเนต 6% น้ำหนัก/ปริมาตร เป็นสัดส่วนที่เหมาะสมที่สุดที่ช่วยเพิ่มสมรรถนะของแบตเตอรี่ได้สูงที่สุด โดยในรอบแรกมีความจุในการคายประจุ 204.0 mA h/g และมีประสิทธิภาพความจุ 98.9% ที่ความหนาแน่นกระแสเท่ากับ 0.1 A/g และมีอัตราการอัดประจุย้อนกลับ (recovery rate) สูงถึง 99.0% หลังจากเปลี่ยนความหนาแน่นกระแสกลับจาก 5.0 A/g เป็น 0.1 A/g ซึ่งสูงกว่าอิเล็กโทรไลต์ฐานน้ำที่ปราศจากเอทิลีนคาร์บอเนตโดยมีความจุในการคายประจุในรอบแรก 111.5 mA h/g และประสิทธิภาพความจุ 97.8% ที่ความหนาแน่นกระแส 0.1 A/g และมีอัตราการอัดประจุย้อนกลับ 90.1% หลังจากเปลี่ยนจากความหนาแน่นกระแสกลับ 5.0 A/g เป็น 0.1 A/g และเมื่อทดสอบ แบตเตอรี่แบบครึ่งเซลล์ (Zn/Zn symmetric cells) เพื่อศึกษาคุณสมบัติการพอกพูนและการละลายสังกะสีในอิเล็กโทรไลต์ พบว่าแบตเตอรี่ที่มีเอทิลีนคาร์บอเนตมีอัตราการพอกพูนและละลายของสังกะสีที่เสถียรตลอดกระบวนการ ในขณะที่แบตเตอรี่ที่ปราศจากเอทิลีนคาร์บอเนตมีความไม่สม่ำเสมอของศักย์ไฟฟ้า และเมื่อวิเคราะห์ลักษณะทางกายภาพของขั้วแอโนดหลังทดสอบแบตเตอรี่แบบครึ่งเซลล์ พบว่าเอทิลีนคาร์บอเนตสามารถยับยั้งการเกิดการพอกพูนสังกะสีแบบแบบกิ่งก้านได้ดี

งานวิจัยนี้มีจุดมุ่งหมายเพื่อศึกษาผลของสารเพอร์ออกไซด์อินทรีย์และภาวะที่เหมาะสมในการผลิตออกซิไดซ์พอลิเอทิลีนแวกซ์โดยใช้เครื่องปฏิกรณ์แบบแบตซ์ เพื่อให้ได้ค่าความเป็นกรดที่มากกว่า 13 มิลลิกรัมโพแทสเซียมไฮดรอกไซด์ต่อกรัม เนื่องจากเป็นค่าที่เหมาะสมในการนำมาใช้เป็นสารหล่อลื่นในภาคอุตสาหกรรม ทำให้สามารถเพิ่มประสิทธิภาพการผลิตและลดต้นทุนการผลิตได้ โดยใช้สารเพอร์ออกไซด์อินทรีย์คือ หนึ่ง,หนึ่ง-บิส-(เติร์ท-เอมิลเพอร์ออกซี)ไซโคลเฮกเซน และ ได-เติร์ท-บิวทิลเพอร์ออกไซด์ เป็นสารริเริ่มปฏิกิริยา (Initiator) ที่ความเข้มข้นร้อยละ 80 โดยน้ำหนัก และใช้แก๊สอากาศ (Air zero) ในการทำปฏิกิริยาออกซิเดชัน โดยใช้เวลาในการทำปฏิกิริยาในช่วง 60 ถึง 180 นาที อุณหภูมิที่ใช้ในการทำปฏิกิริยาในช่วง 90 ถึง 130 องศาเซลเซียส และปริมาณของสารเพอร์ออกไซด์อินทรีย์ในช่วงร้อยละ 2 ถึง 8 โดยน้ำหนัก จากการทดลองพบว่าภาวะที่เหมาะสมในการผลิตออกซิไดซ์พอลิเอทิลีนแวกซ์จากปริมาณพอลิเอทิลีนแวกซ์ 100 กรัม ได้แก่ เวลาในการทำปฏิกิริยา 180 นาที อุณหภูมิที่ใช้ในการทำปฏิกิริยา 123 องศาเซลเซียส และปริมาณของสารเพอร์ออกไซด์อินทรีย์ หนึ่ง,หนึ่ง-บิส-(เติร์ท-เอมิลเพอร์ออกซี)ไซโคลเฮกเซน ร้อยละ 6 โดยน้ำหนัก จะได้ค่าความเป็นกรด 15.23 มิลลิกรัมโพแทสเซียมไฮดรอกไซด์ต่อกรัม นอกจากนี้จากการวิเคราะห์ด้วยเทคนิค FTIR และ NMR ยังยืนยันว่าพอลิเอทิลีนแวกซ์ถูกออกซิไดซ์ในเครื่องปฏิกรณ์แบบแบตซ์

Arsenic is one of the contaminants in wastewater which becomes a worldwide environmental problem. Therefore, several simple and cost-effective techniques have been developed for arsenic removal. This study reports on the removal of arsenic by coprecipitation method with variable combinations of the well-known metal salts, such as FeCl3 or AlCl3 with the environmentally friendly inorganic metal salts, such as MgCl2 or CaCl2, which is called "mixed metal system". Interaction between particles and different surfaces under ambient condition is studied using AFM-based force spectroscopy. This work focuses on the adhesion mechanism and factors affecting particle-surface adhesion. The particles of interest consist of magnesium silicate (talcum powder) and zinc oxide (ZnO) particles, which have very attractive properties and are widely used in several applications. The adhesion force measurements were first carried out using silicon/silicon coated with DLC probes pressed on talc particles modified with hydrochloric acid or different organosilanes. Heparan sulfate (HS), a type of glycosaminoglycan (GAG), has been identified as a ligand for interactions with polynuclear platinum complexes (PPCs). HS is involved in numerous biological processes, and often considered to be extracellular DNA. PPCs interact with HS through the formation of a “sulfate clamp”, where multiple anionic sulfate groups cluster around the cationic PPC. The masking of HS results in the inhibition of enzymatic activity, protein-HS interactions, and other biological processes. PPCs use HS proteoglycans as an method of cellular internalization. By targeting sulfated-GAGs, PPCs can offer precision medicine not currently observed with the FDA approved platinum therapies. …

The study of organophosphorus and nitroaromatic compounds is essential due to their suitability as models for understanding the dynamics of radiation induce damage to DNA sugar-phosphate backbone, and the detection of chemical warfare agents as well as nitroaromatic explosives (such as TNT). The ultrafast dynamics of these polyatomic radicals were studied using femtosecond time-resolved mass spectrometry (FTRMS), a versatile method that is capable of monitoring the ultrafast dynamics of rapid dissociation in the gas phase after the removal of an electron. C-H activation is considered the “holy grail” of chemistry. Remote, selective C-H activation has many applications such as modifying precursors, adding complexity to molecules, and modifying pre- existing structures in a reduced number of steps. Previous work by Swift and Gronert identified an iridium(III) phenanthroline complex that can activate C-H bonds of cyclohexane. The objective of this dissertation is to study the mechanism and regioselectivity of this complex in common precursors, namely 1-butanol, 1-methoxybutane and butyl acetate.

In the title molecule, C22H16O, the indanone ring system is approximately planar with a dihedral angle between the fused rings of 5.13 (14)°. Two benzene rings are linked together at one side of a double bond, sitting on either side of the indanone ring system and making dihedral angles of 70.30 (12) and 44.74 (13)° with it. In the crystal, hydrogen bonding is not present, but weak C—H⋯π or π–π interactions occur and molecules form a sheet-like structure in the bc plane.

Oxidative stress-induced neuroinflammation is the prominent feature of neurodegenerative disorders, and is characterized by a gradual decline of structure and function of neurons. Many biochemical events emerge thanks to the result of this neurodegeneration, and ultimately provoke neuroinflammation, activation of microglia, and oxidative stress, leading to neuronal death. Eastern Kentucky is home to many private ponds used for agricultural and recreational purposes. Each year, the owners of these ponds observe harmful algal blooms (HABs) that release toxins into the water, potentially limiting the use of these ponds. A pilot study in summer 2018 observed a harmful algal bloom (HAB) occurring in one pond in Madison Country that roughly correlated with a rise in water sulfate levels with no detectable levels of nitrate or phosphate present. The use of SERMs (Selective Estrogen Receptor Modulators) – most notably the drug Tamoxifen – has significantly increased among athletes over the past decade to enhance athletic performance and/or negate certain side effects of using anabolic steroids. Capillary electrophoresis-mass spectrometry is a powerful technique for high-throughput and high efficiency separations combined with structural identification. Electrospray ionization is the primary interface used to couple capillary electrophoresis to mass analyzers; however, improved designs continue to be reported. A new interfacing method based on vibrating sharp-edge spray ionization is presented in this work to overcome the challenges of decoupling applied voltages and to enhance the compatibility with separations performed at near-neutral pH. Ergot alkaloids are fungal tryptophan derived toxins which affect mammalian circulation and neurotransmission. These compounds are biosynthesized by a conserved genetic pathway, known as the ergot alkaloid synthesis (EAS) pathway by fungi belonging to the ascomycete families Trichocomaceae and Clavicipitaceae. Several Ipomoea species and related plants in the morning glory family harbor vertically transmitted symbiotic fungi in the genus Periglandula, also members of Clavicipitaceae, that produce ergot alkaloids. Metabolomic analysis of seeds identified a previously uncharacterized glycoside form of the pharmaceutically important ergot alkaloid, ergonovine. Capillary electrophoresis has many advantages that make it a powerful technique for the analysis of proteins, which is challenging due to the natural heterogeneity of proteins. The small sample volume required for each analysis (nano- and pico-liter), fast separation times (<30 >min), and ability to couple the separation to mass spectrometry are all factors that make capillary electrophoresis an ideal technology for protein separations. Capillary zone electrophoresis and capillary gel electrophoresis are the two most commonly employed modes for protein analysis and allow for rapid, high resolution separations that can be automated. The synthesis and electronic structures of the square-planar high-spin FeII complexes (MesPDPPh)Fe(OEt2) and (MesPDPPh)Fe(thf) were investigated. Analysis of these compounds using a combination of 1H NMR and 57Fe Mössbauer spectroscopies, in conjunction with computational analysis, has resulted in a rationalization for their unusual electronic structures, which arise from the unique coordination environment enforced by the pyridine dipyrrolide chelate ligand.

Huntington’s Disease (HD) is a genetic, neurodegenerative disease characterized by an abnormal polyglutamine (polyQ) expansion in the first exon of the huntingtin protein (htt). The polyQ domain facilitates aggregation and initiates the formation of a diverse collection of aggregate species, including fibrils, oligomers and annular aggregates. The first 17 amino acids of htt (Nt17) directly flank the polyQ domain and is a key factor in htt’s association to membranous structures. In addition to Nt17 being an amphipathic αhelix, it also promotes aggregation through self-association and contains numerous posttranslational modifications (PTMs) that can modulate toxicity and subcellular localization.
