Synthesis And Characterization Of Enantioenriched Deuterated And Fluorinated Small Molecules, Mitchell D. Mills
Synthesis And Characterization Of Enantioenriched Deuterated And Fluorinated Small Molecules, Mitchell D. Mills
Master's Theses (2009 -)
The development of novel deuterated and fluorinated bioisosteres has significantly impacted the pharmaceutical and agricultural industry and created a growing demand for new synthetic methodology. Both deuterated and fluorinated small molecules exist at the forefront of drug discovery due to their unique ability to attenuate the pharmacokinetic properties of new and currently existing drugs. Selective, high-throughput methods for deuteration and fluorination are scarce in the literature, especially methods that introduce D or F atoms asymmetrically. Benzylic CH bonds are oftentimes key sites for enzymatic manipulation in metabolic processes, alteration of CH bonds to CD or CF bonds at the benzylic …

A Proposed Treatment Of Mixed Connective Tissue Disease By Competitive Inhibition Of Autoantibodies, Thomas Russell
Senior Honors Theses
Mixed Connective Tissue Disease is an autoimmune disease characterized by Raynaud’s phenomenon and arthritis among other symptoms. It is primarily caused by antibodies that target the U1-RNP 70K peptide. The treatment proposed in this paper uses competitive inhibition to prevent the binding of the anti-U1-RNP 70K antibodies with the U1-RNP 70K peptide. A method for testing the designed treatment in silico is proposed using AutoDock Vina docking software.

The Development Of Inhibitors For Sars-Cov-2 Orf8, My Thanh Thao Nguyen
The Development Of Inhibitors For Sars-Cov-2 Orf8, My Thanh Thao Nguyen
CSB and SJU Distinguished Thesis
An unexpected outbreak of SARS-CoV-2 caused a worldwide pandemic in 2020. Many repurposed drugs were tested, but there are currently only three FDA approved antivirals (Merck’s antiviral Molnupiravir, Pfizer’s antiviral Paxlovid, and Remdisivir).1 Most of the antiviral drugs tested SARS-CoV-2 main protease and RNA-dependent RNA polymerase. However, it is important to explore different drug targets of SARS-CoV-2 to prepare for the virus mutations of the future. This research looks at an alternative approach in which SARSCoV- 2 Open Reading Frame 8 (ORF8), which has been shown to be a rapidly evolving hypervariable gene, was chosen to be the protein of …

CSB and SJU Distinguished Thesis
Severe acute respiratory syndrome (SARS-CoV-2) led to the COVID-19 global pandemic, with over 460 million cases of infection and over 6 million deaths since the start of the pandemic. SARS-CoV-2 is a retrovirus that utilizes a main protease (Mpro). Mpro is a catalytic cys/his protease. Several treatments were proposed to stop the pandemic including repurposing drugs to inhibit the Mpro. Another retrovirus that uses a protease is human immunodeficiency virus (HIV-1) which has been a global epidemic for 40 years and is a devastating disease that attacks the immune system. HIV-1 has infected 79.5 million people and has killed an …

Block Copolymer Directed Self-Assembly: Exploring The Efficacy Of Applications In Semiconductor Fabrication, Jakin Bryce Delony
USF Tampa Graduate Theses and Dissertations
Over the course of the past 80 years, semiconductor devices have become increasingly ubiquitous in everyday life. From constructing mainframes that encompassed entire rooms during the 1940s, to inventing personal computers in the 1980s, to developing progressively faster smartphones and wearable technology in the 2010s, the primary driving force behind the Digital Revolution has been increasing transistor counts, and thus computing power, via incremental improvements in optical lithography. In 1965, Intel co-founder Gordon Moore boldly predicted that the transistor density of semiconductor devices would double approximately every 18-24 months. While this prediction -- now colloquially referred to as Moore's Law -- …
Adsorption Of Microcystins To Sediment Affects Their Bioavailability And Remediation, Ashley Womer
Adsorption Of Microcystins To Sediment Affects Their Bioavailability And Remediation, Ashley Womer
Theses and Dissertations
The freshwater harmful algae Microcystis spp produces a family of toxins called microcystins1. These species of algae are known to release microcystins from their cells directly into the surrounding water2. This creates an array of human health risks depending on the fate and transport of the toxins in the water column3,4. Risk modelling and previous literature has shown that sediment is a sink for free microcystin, microcystin can be stably adsorbed to sediment for hundreds of years, and its adsorption to sediment is determined by the sediment adsorption partitioning coefficient (Kd)5,6,7. In this work it is hypothesized that the sediment …

Chemistry & Biochemistry Faculty Publications and Presentations
Within the last 2 decades, severe acute respiratory syndrome coronaviruses 1 and 2 (SARS-CoV-1 and SARS-CoV-2) have caused two major outbreaks; yet, for reasons not fully understood, the coronavirus disease 2019 pandemic caused by SARS-CoV-2 has been significantly more widespread than the 2003 SARS epidemic caused by SARS-CoV-1, despite striking similarities between these two viruses. The SARS-CoV-1 and SARSCoV-2 spike proteins, both of which bind to host cell angiotensin-converting enzyme 2, have been implied to be a potential source of their differential transmissibility. However, the mechanistic details of prefusion spike protein binding to angiotensin-converting enzyme 2 remain elusive at the …

Fragmentation Method For Computing Quantum Mechanics And Molecular Mechanics Gradients For Force Matching: Validation With Hydration Free Energy Predictions Using Adaptive Force Matching, Dong Zheng, Ying Yuan, Feng Wang
Chemistry & Biochemistry Faculty Publications and Presentations
A fragmentation approach referred to as a simple overlapping region method for force matching (SORForM) is presented. SORForM is designed to enable efficient computation of quantum mechanical (QM) forces for large molecules and is validated in the framework of adaptive force matching (AFM) to develop solute models in water. The SORForM method divides a molecule into overlapping QM regions with each region containing a gradient zone and a buffer zone. The buffer zone ensures that the atoms in the gradient zone have their surroundings unchanged with fragmentation. The performance of the method is validated with mefenamic acid and linalyl acetate …

Tanzania Journal of Science
Bulk geochemical data for the Twangiza-Namoya Gold Belt, Democratic Republic of Congo, are presented in order to classify the rock assemblages, elucidate their petrogenesis and tectonic settings. Nb/Yb and Zr/Ti constraints reveal two suites for the rocks: sub-alkaline to andesitic basalts (Suite-1); and alkaline basalts (Suite-2). Ratios of Ti/Zr = 22–70; Zr/Hf = 35–42; Nb/Ta = 12–13; Nb/Th = 1–2 and La/Nb = 2.4–3.8 suggest arc-generated mantle melts with crustal inputs and arc-fluids metasomatism for Suite-1. The samples have positive LILE and negative Nb, Ta, Ti anomalies, enrichments in the LREE over middle and HREE (La/Yb)CN = 3.6–8.9 with negative …

Attempted Synthesis & Antibacterial Properties Of Apt-6k Against Ndm-1 K. Pneumoniae, Alec Bofetiado
Honors Theses
NDM-1 K. pneumoniae is a highly resistant bacterial organism that is capable of causing debilitating nosocomial infections in immunocompromised patients. Only "last-resort" antibiotics--such as colistin--work against this organism. Therefore, new antibiotics are needed to help fight against these types of infections. APT-6K is a novel compound that was demonstrated to be effective against MRSA with nanomolar concentrations in a prior study. Novel methods of APT-6K synthesis and its testing for antibiotic effects against NDM-1 K. pneumoniae were attempted in this research. APT-6K synthesis was unsuccessful. Commercially-prepared APT-6K also did not demonstrate growth inhibition against NDM-1 K. pneumoniae nor against a …

The design of bioinspired polymers has long been an area of intense study, however, applications to the design of concrete admixtures for improved materials performance have been relatively unexplored. In this work, we functionalized poly(acrylic acid) (PAA), a simple analogue to polycarboxylate ether admixtures in concrete, with dopamine to form a catechol-bearing polymer (PAA-g-DA). Synthetic routes using hydroxybenzotriazole (HOBt) as an activating agent were examined for their ability in grafting dopamine to the PAA backbone. Previous literature using the traditional coupling reagent 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC) to graft dopamine to PAA were found to be inconsistent and the sensitivity of EDC coupling …

LSU Doctoral Dissertations
Globally cell culture is an $18.98 billion industry as of 2020, with an 11.6 percent annual growth rate. Drug discovery has an estimated worth of $69.8 billion in 2020 and is predicted to grow to $110.4 billion by 2025. Three-dimensional (3D) cell culture of cancer cells is one of the rapidly growing felids since it better recapitulates in vivo conditions compared to two-dimensional (2D) models. However, it is challenging to grow 3D tumor spheroids outside the body, and some of the existing technology can generate these spheroids outside the human body but poorly mimic in vivo tumor models. Therefore, there …

Small-Molecule Ras Inhibitors As Anticancer Agents: Discovery, Development, And Mechanistic Studies, Shaila A. Shetu, Debasish Bandyopadhyay
School of Integrative Biological & Chemical Sciences (Formerly Dept. of Chemistry)
Mutations of RAS oncogenes are responsible for about 30% of all human cancer types, including pancreatic, lung, and colorectal cancers. While KRAS1 is a pseudogene, mutation of KRAS2 (commonly known as KRAS oncogene) is directly or indirectly associated with human cancers. Among the RAS family, KRAS is the most abundant oncogene related to uncontrolled cellular proliferation to generate solid tumors in many types of cancer such as pancreatic carcinoma (over 80%), colon carcinoma (40–50%), lung carcinoma (30–50%), and other types of cancer. Once described as ‘undruggable’, RAS proteins have become ‘druggable’, at least to a certain extent, due to the …

An Additive Incorporated Non-Nucleophilic Electrolyte For Stable Magnesium Ion Batteries, Mao-Ling Xie, Jun Wang, Chen-Ji Hu, Lei Zheng, Hua-Bin Kong, Yan-Bin Shen, Hong-Wei Chen, Li-Wei Chen
Journal of Electrochemistry
Non-nucleophilic electrolytes are promising next-generation highly stable electrolytes for magnesium-ion batteries (MIBs). However, a passivation layer on Mg metal anode usually blocks Mg2+ diffusion, leading to poor reaction kinetics and low Coulombic efficiency of the Mg plating/stripping in these electrolytes. Here we explore the utilization of phenyl disulfide (PDF) as a film-forming additive for non-nucleophilic electrolytes to regulate the interfacial chemistry on Mg metal anode. Phenyl-thiolate generated from the PDF additive was found to suppress the unfavorable surface blocking layer, resulted in a high Coulombic efficiency of up to 99.5% for the Mg plating/stripping process as well as a …
