Acetylpyrazine is an important spice and also plays important roles in biology, medicine and other fields. However, the wastewater containing ammonium persulfate, ferric ion (Fe3+), pyrazine, and a large amount of ammonium sulfate was produced during the production of acetylpyrazine. In this study, ammonium sulfate in acetylpyrazine wastewater was electrolytically converted into ammonium persulfate for economic benefits. Using platinum as an anode and graphite as a cathode, the influence factors of the electrolysis process (including the composition of the anolyte, the current density, the temperature, etc.) were firstly investigated without interference from other compositions. The role of phosphorus in biochemistry is well understood. However, the route by which phosphorus was incorporated into early biomolecules on the prebiotic Earth is uncertain. Phosphate, the most prevalent species of phosphorus found in Earth’s geological record, is insoluble and unreactive with organics in aqueous environments. While the most abundant biogenic elements (C, N, H, O, and S) can be found in a volatile phase under terrestrial conditions, phosphorus cannot, suggesting that minerals must have been the main sources of phosphorus on the early Earth. Parkinson’s disease (PD) is a progressive neurodegenerative disorder for which there is no successful prevention or intervention. The pathological hallmark for PD involves the self-assembly of functional Alpha-Synuclein (αS) into non-functional amyloid structures. One of the potential therapeutic interventions against PD is the effective inhibition of αS aggregation. However, the bottleneck towards achieving this goal is the identification of αS domains/sequences that are essential for aggregation. Using a protein mimetic approach, we have identified αS sequences-based targets that are essential for aggregation and will have significant therapeutic implications. Understanding the dynamics of peatland methane (CH4) emissions and quantifying sources of uncertainty in estimating peatland CH4 emissions are critical for mitigating climate change. The relative contributions of CH4 emission pathways through ebullition, plant-mediated transport, and diffusion, together with their different transport rates and vulnerability to oxidation, determine the quantity of CH4 to be oxidized before leaving the soil. Notwithstanding their importance, the relative contributions of the emission pathways are highly uncertain. In particular, the ebullition process is more uncertain and can lead to large uncertainties in modeled CH4 emissions. To improve model simulations of CH4 emission and its pathways, …

Water contact angles are used to determine hydrophilicity, which is a material's attraction to water. A water contact angle of greater than 90 degrees indicates a hydrophobic material that repels water, whereas a contact angle of less than 90 degrees indicates a hydrophilic material [Fig. 1]. Water contact angles are measured through a goniometer, which can cost between $2000 to $10,000.

We explored the ability of ADP-glucose pyrophosphorylase (ADP-Glc PPase) from different bacteria to use glucosamine (GlcN) metabolites as a substrate or allosteric effectors. The enzyme from the actinobacteria Kocuria rhizophila exhibited marked and distinctive sensitivity to allosteric activation by GlcN-6P when producing ADP-Glc from glucose-1-phosphate (Glc-1P) and ATP. This behavior is also seen in the enzyme from Rhodococcus spp., the only one known so far to portray this activation. GlcN-6P had a more modest effect on the enzyme from other Actinobacteria (Streptomyces coelicolor), Firmicutes (Ruminococcus albus), and Proteobacteria (Agrobacterium tumefaciens) groups. Upon infection, DNA viruses can be sensed by pattern recognition receptors (PRRs), leading to the activation of type I and III interferons to block infection. Therefore, viruses must inhibit these signaling pathways, avoid being detected, or both. Papillomavirus virions are trafficked from early endosomes to the Golgi apparatus and wait for the onset of mitosis to complete nuclear entry. This unique subcellular trafficking strategy avoids detection by cytoplasmic PRRs, a property that may contribute to the establishment of infection. However, as the capsid uncoats within acidic endosomal compartments, the viral DNA may be exposed to detection by Toll-like receptor 9 …

An alkyne-functionalized elastomer derived from sebacic acid, 1,3-propanediol, and alkyne-functionalized serinol is synthesized via melt condensation. A low-power UV lamp triggers the cross-linking rapidly via thiol-yne click chemistry. The cross-linking behavior is studied by photorheology and NMR spectroscopy. The resultant elastomer possesses mechanical properties similar to those of human soft tissues and exhibits in vitro degradability and good cytocompatibility.

Platelets are key players in our body’s inflammatory processes upon activation, which can create physiological problems. Researchers are actively seeking solutions by targeting platelets with therapeutics. The objective of the research in this thesis was to investigate a new and potentially less invasive drug delivery platform using nanoparticles. The topic of the project was to explore the world of ionic liquids and their abilities, when coated on nanoparticles, to bind to platelets. In the presence of trimethylsilyl trifluoromethanesulfonate (TMSOTf) and 2,6-lutidine, acylated thiones undergo Aldol and Mannich reactions with aldehydes and nitrones to yield complex nitrogen-containing compounds. The use of a chiral metal-ligand complex is hypothesized to accelerate the reaction and increase stereochemical control during product formation. N-Alkylindoles undergo Friedel-Crafts alkylation with acetates in the presence of TMSOTf and 2,6-lutidine to produce 3-alkyl indoles. Varying Substitution on the indole at the 5 position and removal of the alkyl group from the 1 position both show continued reactivity at the 3 position even in the presence of TMSOTf.

Fast photochemical oxidation of proteins (FPOP) is a useful tool in proteomics because of the ability for modifications to occur on the scale of microseconds which reduces the modifications to tertiary and quaternary structure allowing for more accurate labeling of the protein. Labels for FPOP are generated from various radicals in our experiments which include hydroxyl radicals and trifluoromethyl radicals. Hydroxyl radicals are easily generated by using an excimer laser (KrF laser, 248 nm) or a UV flash lamp (as a part of the Fox™ System) by the photolysis of hydrogen peroxide. TUG-UBL1 (Tether Containing UBX Domain for GLUT-4 Ubiquitin Like 1) is a glucose transporter 4 (GLUT4) regulating protein which facilitates the release of GLUT4 transport vesicles when insulin stimulation is present. To understand the role of TUG-UBL1 in glucose metabolism, especially for type two diabetic patients for whom increases in insulin concentration do not correlate with increased glucose uptake, we explored the research question of whether TUG-UBL1 binds to insulin. Initially, models of full-length TUG-UBL1 (amino acids 1-473), the GLUT4 binding domain of TUG-UBL1 (amino acids 86-337), and the insulin heterodimer were built using PHYRE2. In a clinical setting, some patients are exposed to an anti-cancer chemotherapy agent, paclitaxel. Cancerous cells undergo rapid, continuous cell division without control. Chemotherapy treatments try to slow and stop the uncontrollable cell division cycles and eliminate cancerous cells in the process. Paclitaxel serves as a treatment for some types of cancers, including lung, melanoma, bladder, and esophageal. Because it targets the cytoskeleton, paclitaxel can also influence cell migration. To better understand how peer review can be used to support students when designing experiments, the current thesis examined how the structure of the peer review template affects the kinds of feedback students give and the revisions that they make. I utilized a quasi-experimental design to investigate the effects that the peer review template had on the outcome of an experimental design task. A sample of 195 students enrolled in an Organic Chemistry I course participated in the study. This dissertation entails design and development of two new synthetic methodologies made possible through the generation of cationic species, namely oxyallyl and amidoallyl cations. Chapter 1 involves literature survey on generation and trapping of oxyallyl cations. Mostly known for cycloaddition reactions with these elusive species, the most recent approach of direct nucleophilic capture of oxyallyl cations are discussed. Apt design of substrates and reaction conditions by Kartika Group to access a-electrophilic carbon towards the synthesis of a variety of value-added compounds, i.e., 1,4- and 1,6-dicarbonyls, carbazoles, densely substituted pyrroles and furans are elaborated.

Azaborines are fascinating compounds because they possess valuable properties such as photochemical stability, have high molar absorption coefficient and high fluorescent quantum yields, as well as large Stokes shifts and tunable absorption/emission spectra. Here, we designed, synthesized, and will examine a novel reaction-based azaborine fluorescent probe capable of selectively detect carbon monoxide (CO) based on palladium-mediated carbonylation chemistry. This novel azaborine fluorescent probe will exhibit high selectivity for CO and display a robust turn-on fluorescent response in the presence of CO in aqueous buffer solution.

Flat-structured heteroaromatic polycyclic compounds with extended conjugated π-systems such as azaborines are in high demand in the material and imaging technology markets because of their unique features such as simultaneous tunability of fluorescence color and intensity. In this study, we combine all-atom MD simulations and comprehensive mutational scanning of S-RBD complexes with the angiotensin-converting enzyme 2 (ACE2) host receptor in the native form as well as the S-RBD Delta and Omicron variants to (a) examine the differences in the dynamic signatures of the S-RBD complexes and (b) identify the critical binding hotspots and sensitivity of the mutational positions. Transition metal complexes ligated by Donor-Acceptor (D-A) biradicals are of interest for probing the degree of electronic coupling between chemically biased radical fragments. The unique nature of their electronic coupling has allowed these systems to serve as ground state analogues of charge separated excited states generated by photoexcitation. Di(2-ethylhexyl) phthalate (DEHP) is a widely used chiral plasticizer whose primary metabolite (MEHP) and secondary oxidative metabolites have been shown to have peroxisome proliferating and endocrine disrupting effects in mice. The synthesis of these oxidative metabolites is necessary to study their binding affinity to nuclear receptor PPAR. Here, the first step in these syntheses, the construction of the alkyl side chains, was optimized by comparing GC-MS data showing percent composition of the desired alkylation product under varying reaction conditions. Within this dissertation, photochemical systems that bear significance to next-generation photonic materials and devices are explored. Notable advances in the design, synthesis, and characterization of three distinct groups of photoactive molecules are achieved through molecular design and spectroscopic analysis. First, novel ruthenium sulfoxide complexes bearing substituted phosphine ligands are found to provide extraordinary control over photoisomerization quantum yields. A comparison of these complexes reveals ground-state characteristics that are instrumental in this reactivity, while a novel spectroscopic technique provides rare structural evidence for an O-bonded metastable isomer. One of the primary symptoms is dilated cardiomyopathy, and alongside the cardiovascular disease that arises, patients often experience metabolic abnormalities such as 3-methylglutaconic aciduria, growth retardation, and neutropenia. Extended-spectrum β-lactamases (ESBLs) of different Enterobacteriaceae (e.g. Escherichia coli, Klebsiella pneumoniae) have developed significant resistance mechanisms against antibiotics. ESBLs, like class A CTX-Ms (cefotaximases) enzymes hydrolyze the β-lactam ring of different antibiotics such as penicillins and third-generation oxyimino-cephalosporins. Although CTX-Ms are repeatedly found in many clinical and community settings, molecular-level information on their function/activity is currently limited to structural studies performed by X-ray crystallography. A new method of determining the cation exchange capacity (CEC) of clay minerals was examined. This method involved exchanging the smectite clay with organic cations and using bomb calorimetry to measure the energy of combustion of the material. Because the clay is noncombustible, the energy is completely dependent on the amount of organic present, which is dependent on the CEC of the clay. Two organic cations, tetrapentylammonium and octadecyltrimethylammonium , with similar size but much different shape were used.
