We are developing energy-efficient and reversible carbon capture and release (CCR) systems that mimic the Lys201 carbamylation reaction in the active site of ribulose-1,5-bisphosphate carboxylase-oxygenase (RuBisCO). The multiequilibria scenario ammonium ion Xa ⇌ amine Xb ⇌ carbamic acid Xc ⇌ carbamate Xd requires the presence of both free amine and CO2 for carbamylation and is affected by the pKa(Xa). Two fluorination strategies aimed at ammonium ion pKa depression and low pH carbamylation were analyzed with (2,2,2-trifluoroethyl) butylamine 2b and 2,2-difluoropropylamine 3b and compared to butylamine 1b. Complementary alternative medicine approaches are growing treatments of diseases to standard medicine practice. Many of these concepts are being adopted into standard practice and orthomolecular medicine. Age-related diseases, in particular neurodegenerative disorders, are particularly difficult to treat and a cure is likely a distant expectation for many of them. Poly(hydroxybutyrate) is a biocompatible, biodegradable polyester synthesized naturally in a variety of microbial species. A greener alternative to petroleum-based plastics and sought after for biomedical applications, poly(hydroxybutyrate) has failed to break through as a leading material in the plastic industry due to its high cost of production. Invasive fungal species provide a challenge to the current drug landscape, only partiallymet by approved drugs. The Acetyl Coenzyme Synthetase (ACS) enzymes are an attractive target for antifungal compounds because normal mammalian cells do not rely on ACS for energy metabolism while fungal cells do rely on ACS. This work focuses on the design and synthesis of inhibitors of various fungal ACS enzymes. The first class of inhibitors is based on the 2-aminothiazole scaffold originating from a saturation transfer difference (STD) nuclear magnetic resonance (NMR) study and previous whole-organism assays. Mo and W enzymes are ubiquitous in living organisms and crucial for sustaining life. Mo and W enzyme variety is diverse, but they contain a highly conserved aspect of functionality in these enzymes, the pyranopterin-dithiolene (PDT) ligand. A total of eight possible electron redox equivalents occurs between the metal center and PDT, making the molybdenum cofactor (Moco) and the tungsten cofactor (Tuco) the most redox rich cofactors in all of biology. Chlorate has become a concern in the food and beverage sector, related to chlorine sanitizers in industrial food production and water treatment. It is of particular concern to regulatory bodies due to the negative health effects of chlorate exposure. This study investigated the fate of chlorate in raw milk and isolated bacterial strains of interest responsible for chlorate breakdown. Unpasteurized milk was demonstrated to have a chlorate-reducing capacity, breaking down enriched chlorate to undetectable levels in 11 days. Block copolymers (BCP) with balanced interactions and their resultant bulk-phase self-assembly have become increasingly important in advancing nanotechnology, separation, and energy applications. However, a few reports have addressed the synthesis challenge and bulk-phase self-assembly of such triblock copolymers. This thesis presents a facile route for preparing triblock copolymer via controlled radical and organocatalytic ring-opening polymerization that allows precise control over the incorporation of individual moieties in resultant polymer, responsible for balanced hydrophilic, hydrophobic/lipophilic, and fluorophilic interactions. This work is a computational study of a reaction mechanism for the trifluoromethylation of boronic acids. Three steps of the proposed reaction mechanism are studied, •CF3 addition to a copper catalyst center, base promoted transmetalation between copper and aryl boronic acid, and bond forming reductive elimination in which the CF3 and aryl substituent form a bond. Tuberculosis (TB) is a disease that is caused by a bacteria called Mycobacterium tuberculosis and primarily attacks the lungs but can spread to other areas of the body as well. TB can develop resistance to drugs that may be used to cure the disease. Multidrug resistant TB is especially a growing issue as it does not respond to the two most powerful anti-TB drugs, isoniazid and rifampicin. The subarctic shield near Yellowknife, Northwest Territories (NWT), is populated with thousands of small lakes (<1.5 km2) and several large lakes. Historic mining activities in the region have left a legacy of environmental impacts and widespread arsenic (As) contamination in both aquatic and terrestrial environments. In particular, several small subarctic lakes near Yellowknife have been previously documented to be contaminated with high levels of As. Gold nanoclusters with diameters in the quantum size regime (< ~2 nm) are promising building blocks for the design of novel nanomaterials as they exhibit unique size-dependent properties that can be altered and fine-tuned. The research outlined in this PhD thesis employs density functional theory to construct and analyze small ligand-protected cage and rod-shaped nanoclusters. Chapter 2 reports stable halide-protected gold nanocages that were engineered to have a closed-shell valence electron count of 18. This study finds that nanocages comprised of 19 and 20 gold atoms can be converted into stable magic number species containing 18 valence electrons by modifying their charged states via adsorption of halide ligands to the cage’s surface. Chapter 3 reports stable ligand-protected gold nanoclusters with a tetrahedral Au4 core that were engineered to have a closed-shell valence electron count of 2. This study investigates the structural and electronic effects of halide and alkoxy ligands on the tetrahedral nanocluster and concludes that the results support the broader conclusion that it’s possible to fine-tune the stability and electronic properties of small gold nanoclusters using appropriate ligands. Chapter 4 reports stable gold nanorods that have diameters in the quantum regime that were constructed from elementary building blocks that contain “halide-staples”. The electrochemical oxidation of aripiprazole was explored at a carbon paste electrode modified with aluminium oxide nanoparticles by cyclic voltammetry and square-wave anodic adsorptive stripping voltammetry. Experimental parameters such as carbon paste composition, scan rate, buffer pH, accumulation time, and accumulation potential were optimized in order to obtain high analytical performance. The incorporation of aluminium oxide nanoparticles into the carbon paste matrix enhanced the effective surface area of the carbon paste electrode and improved the sensitivity. Polymer brushes are promising many applications as smart materials and biocompatible surfaces. Surface-initiated reversible addition-fragmentation chain transfer (RAFT) polymerization is one of the most effective techniques for synthesis of well-defined polymer brushes. Herein, a biocompatible, uniform and stable poly(N-acryloylmorpholine)-silicon hybrid system was achieved using surface-initiated RAFT polymerization. Evidence of a well-controlled surface-initiated RAFT polymerization was confirmed by a linear increase of number average molecular weight (M$_{n}$) with overall monomer conversions. Bis(4-(4-nitrobenzyl)pyridine)dichloropalladium(II), [PdCl$_{2}$L$^{1}$$_{2}$], bis(2-amino-5-bromopyridine)dichloropalladium(II), [PdCl$_{2}$L$^{2}$$_{2}$], bis(2,4-dimethylpyridine)dichloropalladium(II), [PdCl$_{2}$L$^{3}$$_{2}$], bis(3,4-dimethylpyridine)dichloropalladium(II), [PdCl$_{2}$L$^{4}$$_{2}$] were prepared. The spectroscopic techniques (FT-IR and $^{1}$H-NMR, $^{13}$C-NMR) were used to characterize the compounds. Theoretical calculations were used to validate the experimental results. The LanL2DZ-based DFT/B3LYP method was used to define the most stable possible molecular structure for the complexes. The present work aimed to investigate inclusion complexes of squalene with various cyclodextrins (native ß-cyclodextrin and methyl-ß-cyclodextrin). The production of squalene-ß-cyclodextrin inclusion complex was obtained using Response Surface Methodology and obtained inclusion complexes were studied with FTIR spectroscopy, X-ray diffractometry, thermal analysis, and 1H-NMR spectrometry. At the same time, squalene content was determined by reversed-phase high-performance liquid chromatography. All results confirmed that squalene was successfully involved in the cyclodextrin cavities. Optimizing the condition in preparation for the squalen-ß-cyclodextrin inclusion complex yielded 54.3% with squalene content of 9.01%. In the present study, we herein report the conductance behavior, effect of temperature, and chain-length of two environmentally friendly imidazolium cationic capric and stearic surfactants. The conductance behavior has been carried out in aqueous solvent (H2O) at four different temperatures such as 24 °C, 29 °C, 34 °C, and 39 °C. The normal micelles were formed in an aqueous solvent and critical micelle concentration (CMC) can be estimated through conductivity parameters. In this study, some of the critical fundamental properties, which are holding importance in usage areas, of low-density polyethylene (LDPE) film were studied by embedding size-graded and polydopamine-coated halloysite nanotubes into the polymer matrix. This concept evaluated the importance of the well-dispersion of nanoparticles in the composite system and interfacial adhesion between nanofiller and polymer matrix on the degree of crystallinity and mechanical properties. In this paper, a new pharmaceutical cocrystal was synthesized using apigenin (AP) and pharmaceutically acceptable conformer nicotinamide (Nico), and the drug delivery between AP-Nico pharmaceutical cocrystal and human serum albumin (HSA) in vivo was studied at atomic scale. The pharmaceutical cocrystal was characterized using Fourier-transform infrared (FTIR) spectroscopy, 1H NMR spectroscopy, differential scanning calorimetry (DSC), and powder X-ray diffraction (PXRD), and the self-assembling mechanism was explored. Copper nano/microparticles were synthesized in octadecene at 290 °C by thermal decomposition method. Copper acetate monohydrate, stearic acid, and 1-octadecanol were used as copper precursor, capping and mild reducing agents, respectively, at the synthesis. Borosilicate glass substrates submerged into the reaction solution during the synthesis were coated by copper nano/ microparticles. Thermal decomposition and coating techniques were combined in this study.
