A series of 3-hydroxy-6-methyl-2-[(substitutedpiperidine-1-yl) methyl]-4H-pyran-4-one structured compounds were synthesized by reacting 5-hydroxy-2-methyl-4H-pyran-4-one with suitable piperidine derivatives using Mannich reaction conditions. Antibacterial activities of the compounds for E. coli ATCC 25922, S. paratyphi ATCC BAA-1250, S. flexneri ATCC 12022, E. gergoviae ATCC 33426, and M. smegmatis ATCC 14468 were assessed in vitro by the broth dilution method for determination of minimum inhibitory concentration (MIC). In addition, their inhibitory effects over DNA gyrase enzyme were evaluated using a DNA gyrase supercoiling assay. Using glucose oxidase as a model enzyme, a novel rhodanine derivative chemiluminescence biosensor for the determination of glucose was formed based on multilayer-enzyme membrane as receptor, which was assembled via layer-by-layer assembly of sol-gel and glucose oxidase-gold nano-particles inside a glass tube. Compared with the traditional chemiluminescence biosensor, the proposed biosensor had some remarkable advantages, such as good selectivity of substrate, good response performance, good stability, good sensitivity, and longer service life. The linear range was from 1.0 \times 10^{-6} to 5.0 \times 10^{-2} mol L^{-1} (r = 0.9991). The detection limit was 5.0 \times 10^{-7} mol L^{-1}. In this …

Synthesis And Spectroscopic Studies Of Some New Organometallic Chelates Derived From Bidentate Ligands, Kiran Singh, Parvesh Puri

A series of organosilicon(IV) and organotin(IV) complexes having the formulae R_2MCl(L^1), R_2MCl(L^2), R_2MCl(L^3), R_2MCl(L^4), R_2M(L^1)_2, R_2M(L^2)_2, R_2M(L^3)_2, and R_2M(L^4)_2, where M = Si and Sn, and R = CH_3, were synthesized with Schiff bases 5-mercapto-4-(pyridine-2-carboxalideneamino)-s-triazole (HL^1), 4-(furfuralideneamino)-5mercapto-s-triazole (HL^2), 4-(2-chlorobenzylideneamino)-5-mercapto-s-triazole (HL^3), and 4-(4-flourobenzylideneamino)-5-mercapto-s-triazole (HL^4). These complexes were characterized by elemental analysis, molar conductance, and spectroscopic studies, including UV, IR, ^1H, ^{13}C, ^{29}Si, and ^{119}Sn NMR spectroscopy. On the basis of these studies, the resulting complexes are proposed to have trigonal bipyramidal and octahedral geometries.

This article describes the preparation and characterization of anticancer drug-loaded poly(lactide-co-glycolide) (PLGA) microparticles. PLGA microparticles loaded with doxorubicin HCl (DOX) were prepared via o/w emulsion solvent evaporation. The release characteristics, encapsulation efficiency, size, and morphology of the PLGA microparticles were also determined. A cytotoxicity test was performed by using Glioma RG2 cancer cells to investigate the cytotoxicity of DOX-loaded PLGA microparticles. The DOX-loaded PLGA microparticles had an average diameter of 500 \pm 9 nm. The DOX encapsulation efficiency and drug loading were 22.75% and 0.78%, respectively. A series of new 4-(3,5-disubstitue-4H-1,2,4-triazol-4-yl)- 5-(thiophen-2-yl-methyl)-2H-1,2,4-triazol-3(4H)-ones (3a-c) were obtained by reaction N'-1-ethoxy-2-thiophen-2-yl-ethylydene hydrazino carboxylic acid ethyl ester (1) and 4-amino-4H-1,2,4-triazoles (2). 4-(3,5-disubstitue-4H-1,2,4-triazol-4-yl)-2-(2-oxo-2-arylethyl)-5- (thiophen-2-yl-methyl)-2H-1,2,4-triazol-3(4H)-ones (4a-e) and ethyl 2-(4-(3,5-disubstitue-4H-1,2,4-triazol-4-yl)-5-oxo-3-(thiophen-2-ylmethyl)- 4,5-dihydro-1,2,4-triazol-1-yl)acetates (6a-c) were obtained by reaction of compounds 3 and bromoacetophenon derivatives and bromo ethylacetate, respectively. Compounds 5a-e were synthesized from the reaction of corresponding compounds 4a-e with NaBH_4. Compounds 7a-c were obtained by the reaction compounds 6 and LiAlH_4. Seventeen new compounds were synthesized and characterized by elemental analyses, IR, ^1H-NMR, and ^{13}C-NMR spectral data. Some new unsymmetrical and optically active 2,5-disubstituted 1,3,4-oxadiazoles 5a-j were efficiently synthesized by cyclization reaction of diacylhydrazides 4a-j. The synthesis of the title compounds was achieved by the reaction of acyl hydrazides 3a-b and N-phetaloyl-L-amino acids 1a-e in the presence of the phosphoroxy chloride (POCl_3) as an anhydrous reagent.

A pH optical sensor was developed based on the use of a mixture of Malachite Green Oxalate and Bromocresol Green indicators immobilized in a triacetylcellulose membrane, which had been previously hydrolyzed. Requirements of the pH indicators were investigated, including the effects of the coupling pH, indicator ratio, and concentrations of the indicators. The pH sensor performed with a very fast response (10 s) and long term stability with no significant leaching of the dyes. An investigation of cause was started in the 1990s at a bleached Kraft mill (BKM), located in Saint John NB, Canada. The 5th effect chemical recovery condensates waste stream (derived from wastewater generated during the pulping process) was identified as having the greatest potential for affecting mummichog (Fundulus heteroclitus), compared to the rest of the inmill waste streams. A solid phase extraction (SPE) protocol was utilized to isolate the causative chemicals in the condensate stream. High density lipoprotein (HDL), the carrier of so-called “good” cholesterol, serves as the major athero-protective lipoprotein and has emerged as a key therapeutic target for cardiovascular disease. We applied small angle neutron scattering (SANS) with contrast variation and selective isotopic deuteration to the study of nascent HDL to obtain the low resolution structure in solution of the overall time-averaged conformation of apolipoprotein AI (apoA-I) versus the lipid (acyl chain) core of the particle. The macrocyclic ligand 3,6,9-tris(methylenebutyl phosphonic acid)-3,6,9−15-tetraazabicyclo [9.3.1]pentadeca-1(15),11,13-triene (PCTMB) was synthesized and complexes of Eu3+, Tb3+, and Gd3+ studied by X-ray crystallography, luminescence, and relaxometry. In the crystal these complexes are dimeric and possess 8-coordinate Ln3+ centers that are linked by bridging phosphonates. The rigidity introduced by the pyridyl nucleus forces the EuPCTMB and TbPCTMB to adopt a twisted snub disphenoid (TSD) coordination geometry. Examination of the 5D0 → 7F0 luminescent transition of EuPCTMB in the solid state confirmed the existence of a single distinct Eu3+ coordination environment, whereas two …

Metal ions like Ca2+, Mn2+, Tb3+, Eu3+ etc., can bind at the active site and have varying effects on the enzymatic activity. In a crystal structure with DNA, two Ca2+ ions were bound to the active site of each PvuII monomer, and the metals share the ligands. Ca2+ ions promote DNA binding and not cleavage in many nucleases. We explored the role of Mg2+ and Ca2+ binding at the metal binding sites of PvuII to elucidate the role of each metal site. We monitored the cleavage kinetics of PvuII in presence of mixed metals. Lyconadin A is a pentacyclic Lycopodium alkaloid isolated from the club moss Lycopodium complanatum with antitumor properties. We have developed a novel 7-exo/6-exo acyl radical cascade cyclization as a method of making the bicyclo[5.4.0]undecane ring system of lyconadin A. The model products are trans-fused ring systems, while a cis-fused ring system is needed in lyconadin A. We have discovered a method to convert the trans-fused model cascade cyclization product into the desired cis isomer. Background: Mammalian spermatogenesis involves formation of haploid cells from the male germline and then a complex morphological transformation to generate motile sperm. Focusing on meiotic prophase, some tissue-specific transcription factors are known (A-MYB) or suspected (RFX2) to play important roles in modulating gene expression in pachytene spermatocytes. The current work was initiated to identify both downstream and upstream regulatory connections for Rfx2.
Results: Searches of pachytene up-regulated genes identified high affinity RFX binding sites (X boxes) in promoter regions of several new genes: Adam5, Pdcl2, and Spag6. We confirmed a strong promoter-region X-box for Alf, a germ cell-specific variant of …

The goal of the first project was to evaluate strategies for determining the in vitro intrinsic clearance (CLint) of dextrorphan (DR) as metabolized by the UGT2B7 enzyme to obtain dextrorphan glucuronide (DR-G). A direct injection liquid chromatography-mass spectrometry (LC-MS) method was used to monitor products using the pseudo-first-order (PFO) model. Standard enzymatic incubations were also quantified using LC-MS. These data were fit utilizing both PFO and Michaelis-Menten (MM) models to determine estimates of kinetic parameters. The CLint was determined to be 0.28 (± 0.08) µL/min/mg protein for a baculovirus insect cell-expressed UGT2B7 enzyme. This is the first confirmation that dextrorphan …

Diaroyl sulfides were prepared in one-pot reaction from ethylenediamine and carbon disulfide, via reaction of in situ generated ethylene-bis- carbamodithioates with aroyl chlorides. The reaction of diaroyl sulfides with acetylenic and ethylenic π-deficient compounds such as dimethyl acetylenedicarboxylate, ethyl propiolate, (E)-1,4-diphenylbut-2-ene-1,4-dione and 1,4-diphenylbut-2-yne-1,4-dione afforded the corresponding thionylated products. Disodium 3β,6β-dihydroxy-5α-cholestane disulfate (1) was synthesized in 4 steps with a high overall yield from cholesterol. First, cholesterol (4a) was converted to cholest-4-en-3,6-dione (5a) via oxidation with pyridinium chlorochromate (PCC) and then 5a was reduced by NaBH4 in the presence of NiCl2 to produce cholest-3β,6β-diol (6a). The reaction of 6a with the triethylamine-sulfur trioxide complex generated diammonium 3β,6β-dihydroxy-5α-cholestane disulfate (7a) and the treatment of 7a by cation exchange resin 732 (sodium form)(Na+) yielded the target steroid 1. The thio-Michael addition reaction is traditionally considered a base catalyzed reaction which involves high catalyst concentrations and long reaction times. This reaction utilizes potent, simple nucleophiles to catalyze the reaction, decreases the catalyst concentration and greatly increases the reaction times. The free radical mediated thiol-ene click reaction uses light or heat and an initiator to catalyze the rapid and quantitative addition of thiols to most electron rich enes without the formation of side products and in the absence of solvent. The development of an efficient, catalytically active, biomimetic model for cytochrome P-450 enzymes has been an area of intense research activity. Ruthenium porphyrin complexes have been the center of this research and have successfully been utilized, as catalysts, in major oxidation reactions such as the hydroxylation of alkanes, the epoxidation of alkenes and aromatic rings, and the N-oxidation of amines, among others. Heterocyclic organic and organometallic compounds (e.g. polypyrrole), and their derivatives, have been of great interest for conductive polymers due to their novel properties and environmental stability as compared to non-aromatic analogs (e.g. polyacetylene). Our current interests focus upon the potential role of pyridazines in next generation electronic devices that utilize organics as the semiconducting material. Pyridazines, 6-membered aromatic rings with two adjacent nitrogens, are promising candidates for a variety of materials and commercial applications. A hydrogel microdome that can swell in response to a stimuli or target molecule is formed by polymerizing a mixture comprising a monomer capable of forming a hydrogel with a biopolymer. An array of hydrogel microdomes can be formed on a substrate by microspotting the mixture and polymerizing. The array can be used for high-throughput screening of analytes as well as for use as an actuator and biosensor using the swelling property of the hydrogel.

The Peaks of Otter Salamander, Plethodon hubrichti, is found along a 19 km length of the Blue Ridge Mountains, Virginia, USA, often in sympatry with the Eastern Red-backed Salamander, P. cinereus. In a sympatric area of Bedford County, Virginia, we conducted a mark-recapture study on a 10 × 10 m site. Surface densities of salamanders increased as the number of days without precipitation prior to a collection event increased. This suggests vertical movements in response to surface moisture. When salamanders returned to the surface after rain, individuals appeared to “shuffle” between rocks and likely, leaf litter. That is, we were …

A series of COX-2 selective inhibitor nimesulide derivatives were synthesized. Their anti-cell proliferation activities were evaluated with a long-term estrogen deprived MCF-7aro (LTEDaro) breast cancer cell line, which is the biological model of aromatase inhibitor resistance for hormone-dependent breast cancer. Compared to nimesulide which inhibited LTEDaro cell proliferation with an IC50 at 170.30 μM, several new compounds showed IC50 close to 1.0 μM.

We show that a quantum-mechanics/molecular-mechanics strategy based on ab initio (i.e., first principle) multiconfigurational perturbation theory can reproduce the spectral properties of a tryptophan residue embedded in the contrasting hydrophobic and hydrophilic environments of parvalbumin and monellin, respectively. We show that the observed absorption and emission energies can be reproduced with a less than 3 kcal mol(-1) error. The analysis of the computed emission energies based on a protein disassembly scheme and protein electrostatic potential mapping allows for a detailed understanding of the factors modulating the tryptophan emission. This study was designed to achieve better understanding of (1) how carboxysome genes are regulated and expressed to yield with precise relative ratios and (2) the in vivo roles of two sets of conserved bacterial microcompartment genes, namely the three csoSl and two csoS4 genes of H. neapolitanus, in the biogenesis and function of the carboxysome. For the first goal, a detailed transcriptional profile of carboxysomal genes in H. neapolitanus was established using absolute quantification real-time RT-PCR and transcript ends analysis. This transcriptional profile revealed that a single promoter, denoted cso promoter, was located upstream from the clustered carboxysomal genes. …
