Characterization Of The Gas-Sensing Protein Cooa, Teryn Gehred, Jessica Lyza

This research aims to characterize the binding specificity of heme proteins to various gas signal molecules. Heme proteins in the gas-sensing family bind to specific gas signal molecules, causing a certain biological function to be activated or deactivated. In this study, variants of the carbon monoxide sensing heme protein CooA from the bacteria R.rubrum and C. hydrogenoformans are compared. Key differences in signal binding specificity and heme environment configuration will be investigated. This is done by using site-directed mutagenesis to alter the heme environment, isolating the CooA proteins through protein purification, and analyzing the isolated products through DNA binding assays. …

Porphyrins are molecular complexes that are critical for the proper functioning of many important biological macromolecules such as hemoglobin and myoglobin. The goal of this research is to monitor the transformations and differences that occur with the addition of metals and ligands to a model porphyrin compound called tetraphenylporphyrin (TPP). During this work, a series of metalloporphyrin complexes were prepared, with changes to both metal centers and ligand attachments at those centers. Specifically, iron, zinc, cobalt and manganese metalloporphyrins have been synthesized and identities verified via electronic absorption spectroscopy. The synthesis and crystal structure of a diiron(II) complex containing a bridging semiquinonate radical are presented. The unique electronic structure of this S = 7/2 complex is examined with spectroscopic (absorption, EPR, resonance Raman) and computational methods.

Peran sistem informasi manajemen (SIM) berbasis teknologi informasi sangat penting dalam pengelolaan sekolah untuk meningkatkan kualitas pelayanan, proses, dan prestasi belajar siswa. Oleh karena itu, penelitian ini bertujuan untuk mengungkap dan mendeskripsikan persepsi pengelola terhadap peran SIM berbasis TI dalam pengelolaan administrasi SMK di Kota Mojokerto. Penelitian ini menggunakan rancangan deskriptif kuantitatif dan dilaksanakan pada sembilan SMK di Kota Mojokerto dengan 45 pengelola administrasi akademik sebagai populasi. Penelitian ini bertujuan untuk mendeskripsikan keterlaksanaan pembelajaran inkuiri terbimbing berbasis kegiatan laboratorium dalam meningkatkan keterampilan proses sains dan hasil belajar kognitif IPA (biologi) siswa. Deskripsi keterlaksanaan pembelajaran diperoleh melalui identifikasi permasalahan yang muncul selama proses pembelajaran di kelas, yang selanjutnya dianalisis untuk menemukan solusi berdasarkan data empiris di lapangan. Penelitian ini juga berupaya mengungkap permasalahan pembelajaran secara menyeluruh dan kontekstual dengan menempatkan peneliti sebagai instrumen kunci. Salah satu penyebab rendahnya produktivitas kedelai di Indonesia adalah serangan virus belang samar kacang tunggak, Cowpea mild mottle virus (CPMMV). Penelitian ini merupakan penelitian survei yang bertujuan untuk menggambarkan dan mengumpulkan informasi mengenai kajian morfologi, anatomi, dan agronomi terhadap infeksi CPMMV pada varietas yang terinfeksi dan yang tidak terinfeksi. Data dianalisis secara deskriptif dan inferensial menggunakan ANAVA melalui program SPSS for Windows versi 16. Hasil penelitian menunjukkan bahwa varietas Gumitir sehat, Anjosmoro sehat, Mahameru sehat, MLGG 0021, dan MLGG 0268 memiliki ukuran daun, panjang petiol, serta nisbah daun (L/W dan L/PL) lebih besar dibandingkan varietas yang sakit. Penelitian ini mendeskripsikan rancangan dan pelaksanaan pembelajaran kooperatif Team Assisted Individualized (TAI) dan game puzzle dalam meningkatkan motivasi belajar serta pemahaman konsep fungsi komposisi dan fungsi invers di SMA Negeri 3 Malang. Penelitian ini merupakan penelitian tindakan kelas dengan pendekatan mix research, yang dilaksanakan dalam tiga siklus. Subjek penelitian adalah siswa kelas XI IPA 1 semester genap tahun pelajaran 2011/2012 sebanyak 33 siswa, terdiri atas 13 siswa laki-laki dan 20 siswa perempuan. Berdasarkan teori beban kognitif, pembelajaran efektif terjadi apabila beban intrinsic dikelola dengan baik, beban germane ditingkatkan, dan beban extraneous dikurangi. Penerapan multimedia merupakan salah satu upaya untuk mencapai pembelajaran yang efektif. Penelitian ini mengkaji penerapan pembelajaran matematika berbantuan multimedia dalam meningkatkan kemampuan siswa menyelesaikan masalah program linear. Penelitian ini menggunakan pendekatan penelitian tindakan kelas yang dilaksanakan dalam dua siklus. Hasil penelitian menunjukkan bahwa penggunaan multimedia dapat mengelola beban intrinsic, meningkatkan beban germane, dan mengurangi beban extraneous, sehingga kemampuan siswa dalam menyelesaikan masalah program linear meningkat.

Penelitian ini akan mendeskripsikan penggunaan pita melalui pembelajaran Missouri Mathematics Project untuk memahamkan siswa tentang luas permukaan bola. Pembelajaran yang dilaksanakan berdasarkan pada tahapan pembelajaran Missouri Mathematics Project yang dipadukan dengan bahan manipulatif. Penelitian ini merupakan penelitian tindakan kelas. Setiap siklus terdiri dari tiga kali pertemuan. Dalam penelitian ini juga dilakukan kajian dari jawaban siswa. Hasil penelitian ini menunjukkan bahwa penerapan pembelajaran menggunakan bahan manipulatif bola dan pita dapat memahamkan siswa tentang luas permukaan bola.

Penelitian ini bertujuan untuk menguji pengaruh model siklus belajar berbantuan mind map terhadap prestasi belajar fisika ditinjau dari kinerja laboratorium siswa kelas VIII pada materi cahaya di SMP Negeri 1 Rejoso, Kabupaten Pasuruan. Penelitian ini merupakan penelitian eksperimen semu dengan rancangan faktorial 2 × 2. Subjek penelitian berjumlah 204 siswa. Penelitian menggunakan rancangan deskriptif kuantitatif dengan subjek guru kejuruan bersertifikat pendidik di wilayah Malang Raya. Instrumen penelitian meliputi kuesioner (angket) dan pedoman wawancara. Pembelajaran inkuiri memungkinkan siswa terlibat secara aktif dalam proses berpikir dan menemukan konsep yang dipelajarinya. Pengetahuan dan keterampilan yang diperoleh siswa diharapkan bukan sekadar hasil menghafal fakta, melainkan hasil dari proses penemuan secara mandiri. DNA polymerases must accurately replicate DNA to maintain genome integrity. Carcinogenic adducts, such as 2-aminofluorene (AF) and N-acetyl-2-aminofluorene (AAF), covalently bind DNA bases and promote mutagenesis near the adduct site. The mechanism by which carcinogenic adducts inhibit DNA synthesis and cause mutagenesis remains unclear. Here, we measure interactions between a DNA polymerase and carcinogenic DNA adducts in real-time by single-molecule fluorescence. We find the degree to which an adduct affects polymerase binding to the DNA depends on the adduct location with respect to the primer terminus, the adduct structure and the nucleotides present in the solution. The fabrication resolution of electrochemical wet stamping (E-WETS) was enhanced to several hundred nanometers from several ten micrometers by using polycarylamide gel (PAG) as soft stamps and through optimized processes. The PAG stamp was cured on a nanopatterned soft mold and soaked with 0.2 mol·L-1 KCl solution, then contacted with a silicon wafer with gold film and applied with electric field. And the gold nanopatterns were achieved by the selective anodic dissolving, the diameter of fabricated gold spot was around 200 nm. The parameters which affected the fabrication processes have been discussed in detail.

The first portion of this research explores the role of water vapor on the kinetics of important atmospheric reactions. Work is presented on the self-reaction of β-hydroxyethyl peroxy radical (β-HEP) and the catalytic increase in reaction rate by water vapor. β-HEP serves as a model system for investigating the possible role of water vapor in perturbing the kinetics and product branching ratio of atmospheric reactions of other alkyl peroxy radicals. We present a hardware-based method that can improve single molecule fluorophore observation time by up to 1500% and super-localization by 47% for the experimental conditions used. The excitation was modulated using an acousto-optic modulator (AOM) synchronized to the data acquisition and inherent data conversion time of the detector. The observation time and precision in super-localization of four commonly used fluorophores were compared under modulated and traditional continuous excitation, including direct total internal reflectance excitation of Alexa 555 and Cy3, non-radiative Förster resonance energy transfer (FRET) excited Cy5, and direct epi-fluorescence wide field excitation of Rhodamine 6G. Removal of chromium(III) or (VI) from aqueous solution was achieved using Fe3O4, and MnFe2O4 nanomaterials. The nanomaterials were synthesized using a precipitation method and characterized using XRD. The size of the nanomaterials was determined to be 22.4±0.9 nm (Fe3O4) and 15.5±0.5 nm (MnFe2O4). The optimal binding pH for chromium(III) and chromium(VI) were pH 6 and pH 3. Isotherm studies were performed, under light and dark conditions, to determine the capacity of the nanomaterials. The capacities for the light studies with MnFe2O4 and Fe3O4 were determined to be 7.189 and 10.63 mg/g, respectively, for chromium(III). The capacities for the light studies …

Ensemble and single molecule methods were used to study model systems of 11 base pair and 16 base pair oligonucleotides. From these experiments it was possible to determine the best ensemble analysis method to use, in conjunction with single molecule experiments, to investigate the structure of lgG in solution. To study the interactions within the antigen-binding fragments (Fab), the binding sites were labeled with a FRET pair. Single molecule fluorescence measurements on the antibody indicate minimal FRET occurring between the dye pair suggesting the distance is too large to be measured using single molecule FRET. The electrostatic ion beam trap (EIBT) was designed by D. Zajfman during the previous decade. This ion trap combines many properties of the Fourier-transform ion cyclotron resonance (FTICR) mass analyzer and time-of-flight (TOF) mass analyzer. There are several advantages for the electrostatic ion beam trap. First, large mass-to-charge particles in an electrostatic field could be easier to analyze. Second, there is a folded flight path, which could make the mass analyzer smaller compared to conventional TOF mass analyzer. Dying-back degeneration of motor neuron axons represents an established feature of familial amyotrophic lateral sclerosis (FALS) associated with superoxide dismutase 1 (SOD1) mutations, but axon-autonomous effects of pathogenic SOD1 remained undefined. Characteristics of motor neurons affected in FALS include abnormal kinase activation, aberrant neurofilament phosphorylation, and fast axonal transport (FAT) deficits, but functional relationships among these pathogenic events were unclear. Experiments in isolated squid axoplasm reveal that FALS-related SOD1 mutant polypeptides inhibit FAT through a mechanism involving a p38 mitogen activated protein kinase pathway. Carbon nanotube (CNT) membranes allow the mimicking of natural ion channels for applications in drug delivery and chemical separation. Double-walled carbon nanotube membranes were simply functionalized with dye in a single step instead of the previous two-step functionalization. Non-faradic electrochemical impedance spectra indicated that the functionalized gatekeeper by single-step modification can be actuated to mimic the protein channel under bias. This functional chemistry was proven by a highly efficient ion rectification, wherein the highest experimental rectification factor of ferricyanide was up to 14.4. A simple method was developed for detection and differentiation of five Tier 1 bacterial agents, including Bacillus anthracis, Francisella tularensis, Yersinia pestis, Burkholderia pseudomallei and Burkholderia mallei as well as their closely related near neighbors by gas chromatography-mass spectrometry (GC-MS). Generally, different classes of compounds can be used as biomarkers for biowarfare agent detection, including nucleic acids (i.e., DNA or RNA), proteins (i.e., antibodies), carbohydrates (i.e., sugars), lipopolysaccharides, lipids (i.e., fatty acids) and small molecules. Bacterial Spores Remain Viable After Electrospray Charging And Desolvation, Sara Nielson Pratt

The electrospray survivability of B. subtilis spores and E. coli was tested in atmospheric mobility experiments. E. coli did not survive electrospray charging and desolvation, but B. subtilis did. Experimental conditions ensured that any surviving bacteria were charged, desolvated, and de-agglomerated. B. subtilis was also found to survive both positive and negative electrospray and subsequent introduction into vacuum conditions. Attempts were made to measure the charge distribution of viable B. subtilis spores using electrostatic deflection. Cisplatin is widely used as an antineoplastic drug, but its ototoxic and nephrotoxic side-effects, as well as the inherent or acquired resistance of some cancers to cisplatin, remain significant clinical problems. Cisplatin’s selectivity in killing rapidly proliferating cancer cells is largely dependent on covalent binding to DNA via cisplatin’s chloride sites that had been aquated. We hypothesized that cisplatin’s toxicity in slowly proliferating or terminally differentiated cells is primarily due to drug-protein interactions, instead of drug-DNA binding.
