Alcohol use disorder (AUD) costs the U.S. billions of dollars each year and is a leading cause of preventable death. AUD leads to many health complications, and those who suffer from AUD will often have stress and anxiety disorder comorbidities. To better understand this connection between AUD and stress and anxiety disorders, restraint stress (RS) and chronic-intermittent ethanol exposure (CIE) procedures were used on rats to analyze neuroinflammation via ELISA in the infralimbic and prelimbic cortices, and the bed nucleus of the stria terminalis (BNST). Iterons are short, repeated DNA sequences that are important for the replication of circular single-stranded DNA viruses. No tools that can reliably predict iterons are currently available. The CRUcivirus Iteron SEarch (CRUISE) tool is a computational tool that identifies iteron candidates near stem-loop structures in viral genomes.

When deep-sea hydrothermal fluids mix with cold oxygenated fluids, minerals precipitate out of solution and form hydrothermal deposits. These actively venting deep-sea hydrothermal deposits support a rich diversity of thermophilic microorganisms which are involved in a range of carbon, sulfur, nitrogen, and hydrogen metabolisms. Global patterns of thermophilic microbial diversity in deep-sea hydrothermal ecosystems have illustrated the strong connectivity between geological processes and microbial colonization, but little is known about the genomic diversity and physiological potential of these novel taxa. “Richness, relative abundance and activity of medium and large mammals of a reserve under restoration in Costa Rica”. Introduction: Private protected areas are a valuable complement to national systems of protected areas. The Sierra Zapote Reserve in Abangares, Costa Rica, established in 2000, protects 70 ha of advanced secondary forest, regenerating secondary forest and primary riparian forest. “Human-wildlife conflict in a buffer zone in San Ramón, Costa Rica”. Introduction: Currently, biodiversity is mainly managed by protected areas, but proximity with human activities results in conflicts, which are poorly studied in the tropics. Objective: To estimate human-wildlife conflict in the buffer zone of the Alberto Manuel Brenes Biological Reserve, San Ramón, Costa Rica. Methods: We interviewed the owners or managers of 59 farms. Results: We recorded 540 incidents, mostly with mammals (N=479); coyotes (Canis latrans) killed 1 074 animals in 183 attacks at 12 farms. Dengue is considered one of the most important human arboviral diseases worldwide. Dengue cases have increased in the last three decades in the Americas, especially along the United States (US)- Mexico (MX) border region, where the main vector of dengue, Aedes aegypti mosquito, is widely distributed. The binding of C-C Chemokine Ligand 19 (CCL19) to C-C chemokine receptor 7 (CCR7) receptor has been linked to T-cell activation, tolerance, and inflammation. CCR7 is a member of the chemotactic cytokine family, essential in the development of T cells and regulatory processes such as immune-cell trafficking. This is accompanied by chemokine ligand concentration and time-dependent internalization, which is efficiently promoted by CCL19. A novel N-terminal truncation of CCL19 has been previously developed as an antagonist observed to impede induced chemotaxis and calcium mobilization in mouse primary immune cells. Two libraries of chemical compounds were analyzed for their cytotoxic activity against cancer and non-cancerous cell lines. In chapter 1, fourteen novel piperidones were tested on a panel of different cell lines to identify their cytotoxic activity. These compounds were found to have cytotoxic activity toward cancer cells with a cytotoxic concentration 50 (CC50) in the micromolar range. Furthermore, these fourteen compounds have selectivity preferentially towards cancer cells. Based on selectivity, and cytotoxicity, four compounds were selected for further characterization in the HL-60 cell line. Anticancer drug discovery is a time and resource-consuming process for which exceedingly reliable and efficient modern approaches are needed. Phenotypic drug screenings can generate highly potent and innovative drug candidates; however, deconvolution of the drugâ??s target often presents significant barriers to drug development. To overcome this hurdle, we have originally combined in vitro and in silico analyses to uncover the molecular mechanism(s) driving the anticancer activity of the uniquely structured small molecule drug candidate, Tpz-1. Liver cancer is one of the most prevalent malignancies, in which hepatocellular carcinoma (HCC) takes up more than 80% of liver cancer cases. Early diagnosis and treatment are practical approaches to impede HCC progression. Identifying and classifying autoantibodies against tumor-associated antigens (TAAs) have long been interested in cancer diagnosis, especially for HCC. Circulating anti-TAA autoantibodies amplify the antigen expression and are more stable in blood, guaranteeing anti-TAA autoantibodies as valid diagnostic biomarkers in clinical application. Ferruginous Hawks are a bird of prey species that nest in sagebrush steppe and grassland habitat. These birds are threatened by habitat loss and fragmentation from urbanization, rural development, oil and natural gas extraction, habitat treatment projects, and wildfires, and experiencing widespread population declines across their breeding range. Leishmaniasis is an understudied disease found predominantly in high heat and humidity areas. The disease is transmitted by sandflies which are blood-feeding, biting insects. There is currently no known vaccine for any form of leishmaniasis and treatment imposes a significant economic impact on already poor and marginalized populations, as well as severe, lingering side effects for afflicted individuals. Investing resources into reproduction can limit energy available to other competing demands, such as fighting off an infection; yet, both processes are necessary for organisms to survive and pass on their genes to the next generation. These strategies often follow patterns associated with lifespan, such that shorter-lived animals are more likely to invest more resources into reproduction over survival, and vice versa in long-lived animals. However, environmental change caused by urbanization can disrupt these relationships, and the within- and transgenerational costs of urbanization on females and offspring are unknown. Fish gills are sensitive to a variety of environmental factors because they regulate the exchange of oxygen and ions between the fish and the external environment. This study examined the effects of polyacrylic acid (PAA) coated titanium dioxide nanoparticle injections (TiO2 NPs) on goldfish (Carassius auratus) over 14 days. Following the experimental period, we examined gill histological samples and looked for signs of an inflammatory response through blood glucose levels and relevant gene expression. TiO2 injected fish developed a more robust ILCM (interlamellar cell mass) compared to the non-injected controls, which remained elevated over 14 days post injection. …

I am interested in the development of the nervous system, especially since during development nerves grow and extend, but in adults, they do not regenerate if damaged. We are specifically interested in the molecules that guide nerves to the correct target during their development. Lysophosphatidic acid (LPA) is a bioactive molecule that has been shown to play a role in neural development. BACKGROUND AND OBJECTIVES: The management of intraoperative blood loss in the surgical treatment of paediatric hip dysplasia is resource intensive. There are numerous clinical factors that impact the need for intraoperative transfusion. Identification of patient and surgical factors associated with increased blood loss may reduce the unnecessary use of resources. This study aimed to identify factors predictive of intraoperative transfusion in children undergoing hip dysplasia surgery.

Proteasome inhibitors have become the standard of care for multiple myeloma (MM). Blocking protein degradation particularly perturbs the homeostasis of short-lived polypeptides such as transcription factors and epigenetic regulators. To determine how proteasome inhibitors directly impact gene regulation, we performed an integrative genomics study in MM cells. We discovered that proteasome inhibitors reduce the turnover of DNA-associated proteins and repress genes necessary for proliferation through epigenetic silencing. Specifically, proteasome inhibition results in the localized accumulation of histone deacetylase 3 (HDAC3) at defined genomic sites, which reduces H3K27 acetylation and increases chromatin condensation. Assessment For Deeper Understanding Using Concept Maps: Lessons Learned From Flipped Teaching Of Pharmacology, Munder Zagaar, Weichao Chen

To foster a deeper understanding of pharmacology concepts among physician assistant students, we integrated concept mapping into our flipped teaching to provide assessment for learning. Different mapping-based assessment strategies were adopted based on learner feedback, including in-person collaborative mapping, an individual computerized mapping-based quiz with automated feedback, and a collaborative computerized mapping-based quiz enhanced by the jigsaw strategy. Each mapping activity also leveraged the strength of a specific technology platform. Based on the findings from comparing learner ratings of these mapping activities and thematic analysis of learner feedback, we engaged in critical reflection and share our lessons learned.

Given the critical role that temperature plays in avian reproductive biology, rising temperatures as a result of global climate change will likely impact reproductive success of many bird populations. Secondary cavity nesting birds, many of which rely on artificial nest boxes to maintain population levels, may be particularly at risk because increased temperatures inside nest boxes can determine whether nest boxes are selected, reduce nest success, and/or negatively affect the growth and survival of offspring. Turtles are one the most threatened vertebrate groups in the world due to anthropogenic threats such as habitat loss and overexploitation. In addition to occupying a range that has been vulnerable to major habitat loss, the Florida box turtle (Terrapene bauri) is particularly at risk of overexploitation due to its popularity in the pet trade. Sanibel Island is a barrier island in southwest Florida that has experienced major habitat loss and is the site of a recent poaching event. Introduction
The response of enterobacteria to oxidative stress is usually considered to be regulated by transcription factors such as OxyR and SoxR. Nevertheless, several reports have shown that under oxidative stress the levels, modification and aminoacylation of tRNAs may be altered suggesting a role of codon bias in regulation of gene expression under this condition.
Methods
In order to characterize the effects of oxidative stress on translation elongation we constructed a library of 61 plasmids, each coding for the green fluorescent protein (GFP) translationally fused to a different set of four identical codons.
