Blue marlin (Makaira nigricans) and sailfish (Istiophorus platypterus) are ecologically important predators and valuable species throughout the world’s recreational, commercial, and subsistence fisheries. Comparing multi-species vertical habitat use can inform ecological uncertainties such as inter-species competition, as well as relative vulnerabilities to fishing activities. In this study, we identified key differences in both depth use and which environmental variables drive these selections, which highlights the variability in the catchability both as target species in recreational fisheries and bycatch in commercial fisheries. To understand these two species’ vertical habitat use, we examined depth profiles from 26 sailfish …

Atomistic Assessment Of Drug-Phospholipid Interactions Consequent To Cancer Treatment: A Study Of Anthracycline Cardiotoxicity, Yara Elsayed Ahmed Jun 2023

Atomistic Assessment Of Drug-Phospholipid Interactions Consequent To Cancer Treatment: A Study Of Anthracycline Cardiotoxicity, Yara Elsayed Ahmed

Theses and Dissertations

Despite being one of the most effective chemotherapeutic agents developed to date, Anthracyclines are notorious for their cardiotoxicity. Their clinical use is frequently limited both in dosage and in prescription due to the severe cardiac damage they cause. The mechanism of anthracycline-induced cardiotoxicity is not yet fully understood. However, it is hypothesized that interactions with the myocardial membrane play an important role in imparting cardiotoxicity. In this study, we use molecular dynamics simulations and density functional theory calculations to study the anthracycline drug molecules and the interactions that they have with the myocardial membrane. We construct a myocardial membrane model …

Cry1 Expression During Postnatal Development Is Critical For The Establishment Of Normal Circadian Period, Aaron Schirmer, Vivek Kumar, Andrew Schook, Eun Joo Song, Michael S. Marshall, Joseph S. Takahashi

Biology Faculty Publications

The mammalian circadian system generates an approximate 24-h rhythm through a complex autoregulatory feedback loop. Four genes, Period1 (Per1), Period2 (Per2), Cryptochrome1 (Cry1), and Cryptochrome2 (Cry2), regulate the negative feedback within this loop. Although these proteins have distinct roles within the core circadian mechanism, their individual functions are poorly understood. Here, we used a tetracycline trans-activator system (tTA) to examine the role of transcriptional oscillations in Cry1 and Cry2 in the persistence of circadian activity rhythms. We demonstrate that rhythmic Cry1 expression is an important regulator of circadian period. We then define a critical period from birth to postnatal day …

The Polycomb Complex 2 Contributes To Developmental Delay When Histone Methylation Is Inappropriately Inherited, Sydney Morgan- Benitez Jun 2023

The Polycomb Complex 2 Contributes To Developmental Delay When Histone Methylation Is Inappropriately Inherited, Sydney Morgan- Benitez

Master of Science in Integrative Biology Theses

Histone methylation is a post-transcriptional modification to the N-terminal tails of histone core proteins that regulates DNA accessibility, and consequently, gene expression. Like DNA, histone methylation can be inherited between generations, and is highly regulated during embryonic development. At fertilization, histone methylation must undergo maternal reprogramming to reset the epigenetic landscape in the new zygote. During maternal reprogramming of histone methylation in C. elegans, H3K4me is removed by the H3K4 demethylase, SPR-5, and H3K9me is subsequently added by the histone methyltransferase, MET-2. Recently, it was demonstrated that SPR- 5 and MET-2 maternal reprogramming antagonizes the H3K36 methyltransferase, MES-4, which …

Measuring The Physiological Effects Of Methylobacterium Oryzae Foliar Inoculation Onto Salt-Stressed Tomatoes Grown In A Greenhouse Semi-Hydroponic System, Collin Dice

Master of Science in Integrative Biology Theses

Salinity stress has been shown to improve hydroponic produce quality such as that of tomato but at the trade-off of produce yield. In order to improve demand for hydroponic produce, techniques that improve salinity tolerance could offset declines in yield without altering the improved quality of produce. In two separate trials, we tested whether foliar inoculation of the ACC deaminase-producing bacteria Methylobacterium oryzae CBMB20 improved salt tolerance in tomatoes grown in a semi-hydroponic system using an indeterminate variety exposed to 26mM NaCl (Trial 1) and a determinate variety exposed to 17mM NaCl (Trial 2). We also tested how salinity and …

Nogo-66 receptor 1 (NgR1) binds a variety of structurally dissimilar ligands in the adult central nervous system to inhibit axon extension. Disruption of ligand binding to NgR1 and subsequent signaling can improve neuron outgrowth, making NgR1 an important therapeutic target for diverse neurological conditions such as spinal crush injuries and Alzheimer's disease. Human NgR1 serves as a receptor for mammalian orthoreovirus (reovirus), but the mechanism of virus-receptor engagement is unknown. To elucidate how NgR1 mediates cell binding and entry of reovirus, we defined the affinity of interaction between virus and receptor, determined the structure of the virus-receptor complex, and identified …

Harmful algal blooms (HABs) in coastal British Columbia (BC), Canada, negatively impact the salmon aquaculture industry. One disease of interest to salmon aquaculture is Net Pen Liver Disease (NPLD), which induces severe liver damage and is believed to be caused by the exposure to microcystins (MCs). To address the lack of information about algal toxins in BC marine environments and the risk they pose, this study investigated the presence of MCs and other toxins at aquaculture sites. Sampling was carried out using discrete water samples and Solid Phase Adsorption Toxin Tracking (SPATT) samplers from 2017–2019. All 283 SPATT samples and …

Spatiotemporal Dynamics Of Ticks And Tick-Borne Disease At Neon Sites Across A Sub-Continental Scale, Ana Sofia Rivera

College of Science and Health Theses and Dissertations

Tick-borne diseases in humans such as Lyme disease cases in the United States have doubled between 2004 and 2016. Understanding the dynamics of infectious diseases has long been of interest for ecologists. Tick and tick-borne diseases are influenced by temperature and precipitation at local scales, indirectly through mast seeding in forest trees which increases the abundance of tick hosts (e.g., small mammals), as well as direct effects on survival. Most tick studies occur at local scales that comprise only a small part of their range. The aim of my thesis is to characterize spatiotemporal dynamics of ticks and tick-borne diseases …

Vertebrate Animal Behaviors And Abundances On Estuarine Shorelines Stabilized With Biodegradable Materials Utilizing Wildlife Cameras, Katherine Guise

The Pegasus Review: UCF Undergraduate Research Journal

Living shoreline stabilization is a restoration technique that utilizes natural materials as breakwaters, plus vegetation landward of the breakwaters, to protect coastlines. This research does not provide information about how new, biodegradable restoration materials affect vertebrates that utilize these shorelines. For this project, I monitored 18 restoration sites along Canaveral National Seashore with wildlife trail cameras: 3 made with cement-infused jute breakwaters, 3 with metal gabion oyster shell breakwaters, and 4 with previously used breakwaters manufactured from plastic mesh oyster shell bags. This project used 4 sites as positive controls (intact vegetation) and 4 as negative controls (highly eroded, no …

The Lake Tana Labeobarbus species flock represents one of the world’s most famous examples of lacustrine species radiations. Previous studies of this group have resulted in the description of at least 15 species based on their differences in functional morphology and definition of two clades (lacustrine and riverine spawning clades) based on life history traits. A total of 166 fish representing 14 Labeobarbus species were genotyped using 10 lineage-specific hexaploid microsatellite loci. Six of these loci were developed for this study based on DNA sequence contigs derived from a microsatellite-enriched genomic library of Labeobarbus intermedius from Lake Tana; the remaining …

The City Blues Of An Iridescent Canary: Physiological, Behavioral, And Developmental Impacts Of Lead (Pb) On Songbirds Along An Urban-To-Rural Gradient, Michelle Ross

Master of Science in Integrative Biology Theses

Urbanization is rapidly changing the environment and creating new challenges in the lives of animals across the globe. Anthropogenic contaminants found commonly in old construction materials—including heavy metals like lead— persist within the environment for prolonged periods of time and present a widespread problem for all who live near contaminated areas. Despite the phase-out of lead usage, it continues to threaten the health of all organisms but especially those from urban areas where historical lead use was more common. Currently, the sub-lethal effects of lead on wildlife are not well understood, though lead is known to affect physiology and behavior …

Meiotic drive loci distort the normally equal segregation of alleles, which benefits their own transmission even in the face of severe fitness costs to their host organism. However, relatively little is known about the molecular identity of meiotic drivers, their strategies of action, and mechanisms that can suppress their activity. Here, we present data from the fruitfly Drosophila simulans that address these questions. We show that a family of de novo, protamine- derived X-linked selfish genes (the Dox gene family) is silenced by a pair of newly emerged hairpin RNA (hpRNA) small interfering RNA (siRNA)-class loci, Nmy and Tmy. …

Utilizing Few-Shot Meta Learning Algorithms For Medical Image Segmentation, Nick Littlefield

Thinking Matters Symposium

Deep learning models can be difficult to train because they require large amounts of data, which we usually do not have or are too expensive to get or annotate. To overcome this problem, we can use few-shot meta-learning, which allows us to train deep learning models with little data. Using a few examples, meta-learning, or learning-to-learn, aims to use the experience learned during training to generalize to unknown tasks. Medical imaging is an industry where it is particularly useful, as there is limited publicly available data due to patient privacy concerns and annotating costs.

This project examines how meta-learning performs …

Pelagic predators must contend with low prey densities that are irregularly distributed and dynamic in space and time. Based on satellite imagery and telemetry data, many pelagic predators will concentrate horizontal movements on ephemeral surface fronts—gradients between water masses—because of enhanced local productivity and increased forage fish densities.
Vertical fronts (e.g. thermoclines, oxyclines) can be spatially and temporally persistent, and aggregate lower trophic level and diel vertically migrating organisms due to sharp changes in temperature, water density or available oxygen. Thus, vertical fronts represent a stable and potentially energy rich habitat feature for diving pelagic predators but remain little explored …

All Faculty Scholarship (Archived)

Belugas (Delphinapterus leucas) engage in many forms of play (e.g., object, water, locomotor), but no play is quite as curious as the unusual form of cooperative social play involving mouth-to-mouth interactions. These playful interactions are characterized by two belugas approaching each other head-to-head and interlocking their jaws, clasping one another, as if they were shaking hands. Observed in belugas both in the wild and in managed care, it is seemingly an important type of social play that offers a unique way of socializing with conspecifics. To describe this unusual behavior, a group of belugas in managed care was …

Obstructive Sleep Apnea-Induced Hypertension Is Associated With Increased Gut And Neuroinflammation, Sriram Ayyaswamy, Huanan Shi, Bojun Zhang, Robert M Bryan, David J Durgan

Faculty, Staff and Students Publications

Background

Obstructive sleep apnea (OSA) is an independent risk factor for the development of hypertension. We have demonstrated that OSA induces gut dysbiosis, and this dysbiotic microbiota contributes to hypertension. However, the mechanisms linking gut dysbiosis to blood pressure regulation remain unclear. Recent studies demonstrate that gut dysbiosis can induce a proinflammatory response of the host resulting in peripheral and neuroinflammation, key factors in the development of hypertension. We hypothesized that OSA induces inflammation in the gut that contributes to neuroinflammation and hypertension.

Methods and Results

OSA was induced in 8‐week‐old male rats. After 2 weeks of apneas, lymphocytes were …

Batrachochytrium salamandrivorans (Bsal) is a fungal pathogen of amphibians that is emerging in Europe and could be introduced to North America through international trade or other pathways. To evaluate the risk of Bsal invasion to amphibian biodiversity, we performed dose-response experiments on 35 North American species from 10 families, including larvae from five species. We discovered that Bsal caused infection in 74% and mortality in 35% of species tested. Both salamanders and frogs became infected and developed Bsal chytridiomycosis. Based on our host susceptibility results, environmental suitability conditions for Bsal, and geographic ranges of salamanders in the …

Reintroducing Hemp (Rongony) In The Material Palette Of Madagascar: A Study On The Potential Of Hemp Clay Components And Its Impact On Social And Ecological Communities., Henintsoa Thierry Andrianambinina

Masters Theses

When mentioning the word hemp, especially in the local language of Madagascar, the literal translation does not set it apart from marijuana, as they are both called “rongony” - creating the stigma around hemp as the negative stereotype of marijuana. However, the material has been used by the ancestors of Madagascar, as well as across cultures, in its fibrous form to produce fabrication like textile goods and packaging. During colonization, the prohibition of hemp intensified, and since then, any activity related to either of these plants is prohibited and will end in severe punitive measures. This thesis explores the strengths …

The second wave of COVID-19 is an unprecedented condition in India and began in mid February 2021. Individuals who were already suffering from other comorbidities were found with lung infection, and hence, the number of disease induced deaths were rising faster during the second wave in relation to the first wave. This paper has proposed a mathematical model with fractional order derivatives by correlating the model based number of infectives with the real number of infectives in India. For the system of fractional differential equations, a disease-free state has been computed and proved to be locally asymptotically stable with certain …

Core concepts provide a framework for organizing facts and understanding in neuroscience higher education curricula. Core concepts are overarching principles that identify patterns in neuroscience processes and phenomena and can be used as a foundational scaffold for neuroscience knowledge. The need for community-derived core concepts is pressing, because both the pace of research and number of neuroscience programs are rapidly expanding. While general biology and many subdisciplines within biology have identified core concepts, neuroscience has yet to establish a community-derived set of core concepts for neuroscience higher education. We used an empirical approach involving more than 100 neuroscience educators to …

Nodulation in Leguminous spp. is induced by common environmental cues, such as low nitrogen availability conditions, in the presence of the specific Rhizobium spp. in the rhizosphere. Medicago sativa (alfalfa) is an important nitrogen-fixing forage crop that is widely cultivated around the world and relied upon as a staple source of forage in livestock feed. Although alfalfa’s relationship with these bacteria is one of the most efficient between rhizobia and legume plants, breeding for nitrogen-related traits in this crop has received little attention. In this report, we investigate the role of Squamosa-Promoter Binding Protein-Like 9 (SPL9), a target of miR156, …

Lactobacillus crispatus, Lactobacillus gasseri, Lactobacillus iners, and Lactobacillus jensenii are dominant species of the urogenital microbiota. Prior studies suggest that these Lactobacillus species play a significant role in the urobiome of healthy females. In our prior genomic analysis of all publicly available L. jensenii and Lactobacillus mulieris genomes at the time (n = 43), we identified genes unique to these two closely related species. This motivated our further exploration here into their genotypic differences as well as into their phenotypic differences. First, we expanded genome sequence representatives of both species to 61 strains, including publicly available …

Solanum, a genus of ~1500 global species, is one of the more interesting plant groups in which to study reproductive biology and ecology. Overwhelmingly, species in this group express full cosexuality, where individual plants have flowers containing both fully-functioning “male” (staminate) and “female” (carpellate) organs. However, there have been multiple and widespread evolutionary transitions within the genus to non-normative variations on this ancestral condition. Australian bush tomatoes (ca. 40 species) are especially diverse in this regard, with uncommon variation and combinations of unisexuality and cosexuality -- including, most notably, two sexual systems known as dioecy (unisexual male or female …

DNA methylation, the addition of a methyl (CH3) group to a cytosine residue, is an evolutionarily conserved epigenetic mark involved in a number of different biological functions in eukaryotes, including transcriptional regulation, chromatin structural organization, cellular differentiation and development. In the social amoeba Dictyostelium, previous studies have shown the existence of a DNA methyltransferase (DNMA) belonging to the DNMT2 family, but the extent and function of 5-methylcytosine in the genome are unclear. Here, we present the whole genome DNA methylation profile of Dictyostelium discoideum using deep coverage replicate sequencing of bisulfite-converted gDNA extracted from post-starvation cells. We find an overall …
