Investigating The Mass Mortality Induced Factors Of Pearl Oyster (Pinctada Radiata) At Abu Dhabi Pearls Farm, Uae, Hamad Ahmed Almansoori
Theses
The aquaculture of shellfish plays a crucial role in enhancing maritime environments by mitigating algal blooms and improving water quality through the filtration of organic matter, nutrients, and bacteria. This study investigated the environmental factors influencing oyster (Pinctada radiata) mortalities through a comprehensive analysis of water quality in Al Mirfa and Al Mughirah farming sites of Abu Dhabi (UAE), from February 2021 to February 2022. To reach such understanding of the main factors led to mass mortalities of cultivated oysters, different monthly sampling and both chemical and biological analyses of the farm water, sediments as well as for both living …

Modulation of the heart’s immune microenvironment is crucial for recovery after ischemic events such as myocardial infarction (MI). Endothelial cells (ECs) can have immune regulatory functions; however, interactions between ECs and the immune environment in the heart after MI remain poorly understood. We identified an EC-specific IFN responsive and immune regulatory gene signature in adult and pediatric heart failure (HF) tissues. Single-cell transcriptomic analysis of murine hearts subjected to MI uncovered an EC population (IFN-ECs) with immunologic gene signatures similar to those in human HF. IFN-ECs were enriched in regenerative-stage mouse hearts and expressed genes encoding immune responsive transcription factors …
Studying The Genes And Conditions That Influence Root Development, Tessa Holtkamp, Hannah Ordonez Webb
Studying The Genes And Conditions That Influence Root Development, Tessa Holtkamp, Hannah Ordonez Webb
Undergraduate Research Symposium
Root development in plants is essential for their survival and understanding how hormones influence their development can explain how plants grow under different circumstances. Researching how Indole-3-butyric acid (IBA), a hormone that induces root production, affects the plant model Arabidopsis thaliana helps explain the hormone's effect in agricultural crop systems. To understand root pathways, we performed assays on mutant lines of Arabidopsis by growing plants on varying concentrations of IBA. For wild-type and mutant lines, phenotyping experiments like branching of roots, lengths of stems, and root length were conducted along with PCR and restriction digest genotyping experiments to compare their …

Pesticides are widely used to protect against unwanted insects, but risk assessments of the effects on pollinators have traditionally only been applied on an individual level, not a population level. This has allowed many pesticides to be approved without the knowledge of sub-lethal effects, such as life history characteristics and population structure to name a few. The biopesticide BotaniGard, containing the entomopathogenic fungus Beauveria bassiana, has previously been thought to be safe for non-targeted insects, but recent studies have found it to have significant negative effects on pollinators. Such negative effects can include decrease in longevity and odorant responsiveness with …

Recovering Ancient Dna Using The Polymerase Chain Reaction, Rose Jennings
Undergraduate Research Symposium
Investigations into aDNA offer a window into the past that modern DNA and paleontological studies alone cannot provide and help address the evolution and connections between hominids, domestication timelines, the analysis of populations over time, and general diversity. Progress in aDNA research has been inherently technology-driven, with modern molecular biology methods, such as the inventions of Polymerase Chain Reaction (PCR) and Next Generation Sequencing (NGS), substantially increasing the analysis possibilities of aDNA. My research this semester has taken me along two parallel paths of investigation: literary research into aDNA and practical exposure to the laboratory techniques used in its analysis. …

Food waste is a major issue that is increasingly affecting our environment. More than one-third of food is wasted, resulting in over $400 billion in losses to the U.S. economy. While composting and other small recycling practices are encouraged from person-to-person, it is not enough to balance the net loss of 80 million tons per year. Currently, one of the most promising routes for reducing food waste is through microbial fermentation, which can convert the waste into valuable bioproducts. Among the compounds produced from fermentation, 2,3-butanediol (2,3-BDO) has gained interest recently due to its molecular structure as a building block …
Dissecting Mechanisms Underlying The Cap1 Functions In The Matrix Adhesion And Proliferation Of Cancer Cells, Auburn Ramsey
Dissecting Mechanisms Underlying The Cap1 Functions In The Matrix Adhesion And Proliferation Of Cancer Cells, Auburn Ramsey
Student Theses and Dissertations
Cancer is a significant health burden as the second leading cause of death worldwide. Metastasis-driven fatality accentuates the need for novel strategies to impede cancer dissemination. The cytoskeletal protein CAP1 plays a pivotal role in linking cell signals to control the actin cytoskeleton, cell adhesion, and proliferation – critical aspects of both metastasis and proliferative transformation. While the role of CAP1 in the actin cytoskeleton is well elucidated, the CAP1 functions in adhesion and proliferation are cell type-specific with poorly understood molecular mechanisms. This study seeks to unravel how CAP1 executes functions in these processes, aiming to contribute to our …

Thermal performance curves (TPCs) depict variation in vital rates in response to temperature and have been an important tool to understand ecological and evolutionary constraints on the thermal sensitivity of ectotherms. TPCs allow for the calculation of indicators of thermal tolerance, such as minimum, optimum, and maximum temperatures that allow for a given metabolic function. However, these indicators are computed using only responses from surviving individuals, which can lead to underestimation of deleterious effects of thermal stress, particularly at high temperatures. Here, we advocate for an integrative frame- work for assessing thermal sensitivity, which combines both vital rates and survival …

Genomic instability is one of the well-established hallmarks of cancer. The homologous recombination repair (HRR) pathway plays a critical role in correcting the double-stranded breaks (DSB) due to DNA damage in human cells. Traditionally, the BRCA1/2 genes in the HRR pathway have been tested for their association with breast cancer. However, defects in the HRR pathway (HRD, also termed ‘BRCAness’), which has up to 50 genes, have been shown to be involved in tumorigenesis and treatment susceptibility to poly-ADP ribose polymerase inhibitors (PARPis), platinum-based chemotherapy, and immune checkpoint inhibitors (ICIs). A reliable consensus on HRD scores is yet to be …
Dynamics Of Post-Mating Responses In Drosophila, Gagneet Kaur
Dynamics Of Post-Mating Responses In Drosophila, Gagneet Kaur
Theses - ALL
Mating induces crucial changes in Drosophila females, influencing vital aspects of behavior and physiology essential for reproductive success. These changes primarily result from gene expression shifts in the female reproductive tract. DNA sequencing and bioinformatics advances have deepened our understanding of postmating responses in Drosophila at the transcriptional level. However, the majority of these studies are confined to D. melanogaster, lack tissue-specific investigations, and are extensively focused on the composition of the ejaculate and the role of male reproductive proteins transferred during mating. This study delves into the dynamic lower reproductive tract of D. virilis subgroup species, exploring the postmating …

The Role Of Stressors In The Behavioral Responses Of Lesser Sirens To Chemical Cues, Mackenzie Hanna Nichols
Student Theses and Dissertations
Organisms must make choices to improve their survival, and often perform behaviors in response to stimuli in the environment. I examined behavioral responses under conditions of stress in a species of fully aquatic salamander, Western Lesser Sirens (Siren intermedia nettingi Goin 1942). First, I presented individuals with chemical cues from a variety of known prey items to determine a potential prey preference. Sirens did not exhibit a preference by prey type. I then exposed individuals to chemical cues from prey, predator, and insecticide sources and evaluated behavioral responses, while also administering a nonlethal physical injury to a subset of subjects …

Soybean (Glycine max) produces a class of phenylalanine (Phe) derived specialized metabolites, isoflavonoids. Isoflavonoids are unique to legumes and are involved in defense responses in planta, and they are also necessary for nodule formation with nitrogen-fixing bacteria. Since Phe is a precursor of isoflavonoids, it stands to reason that the synthesis of Phe is coordinated with isoflavonoid production. Two putative AROGENATE DEHYDRATASE (ADT) isoforms were previously co-purified with the soybean isoflavonoid metabolon anchor ISOFLAVONE SYNTHASE2 (GmIFS2), however the GmADT family had not been characterized. Here, we present the identification of the nine member GmADT family. We determined that the GmADTs …

The Combined And Individual Effects Of Azadirachta Indica L. Oil And Nahco3 On The Inhibition Of F. Oxysporum, Neya Murugesan
Journal of the South Carolina Academy of Science
F. oxysporum is the causal agent of Fusarium wilt. This disease is known to penetrate the roots of plants and wilt the vascular system. The most common method for control is through the use of synthetic fungicides, which are known to cause adverse effects on the environment and human health. Thus, it is necessary that more methods of biocontrol be explored. Current literature shows that various salts and essential oils are effective for the biocontrol of F. oxysporum. However, the two types of substances have not yet been tested together, causing a gap in current research. Due to the antifungal …

The Effect Of Exposure To Lentinula Edodes And Flammulina Velutipes On The Biodegradation Of Crystallized Polylactic Acid, Iraine Taine
Journal of the South Carolina Academy of Science
Crystallized polylactic acid (CPLA) is a prevalent bioplastic which, despite being biodegradable, is extremely resistant to decomposition in natural environments. As a result, CPLA waste contamination is rapidly becoming a prevalent issue. Lentinula edodes and Flammulina velutipes are two widespread white-rot fungi whose capabilities for bioplastic degradation have not been examined. This study aimed to determine a natural approach for reducing plastic waste by evaluating the ability of L. edodes and F. velutipes to degrade CPLA bioplastic. It was hypothesized that CPLA samples would decrease in mass when individually treated with L. edodes and F. velutipes due to their similarities …
