Molecular Basis Of Loss-Of-Function Mutations In The Glp-1 Gene Of Caenorhabitis Elegans, Voula Kodoyianni, Eleanor M. Maine, Judith Kimble

The glp-1 gene encodes a membrane protein required for inductive cell interactions during development of the nematode Caenorhabditis elegans. Here we report the molecular characterization of 15 loss-of-function (lf) mutations of glp-1. Two nonsense mutations appear to eliminate glp-1 activity; both truncate the glp-1 protein in its extracellular domain and have a strong loss-of-function phenotype. Twelve missense mutations and one in-frame deletion map to sites within the repeated motifs of the glp-1 protein (10 epidermal growth factor [EFG]-like and 3 LNG repeats extracellularly and 6 cdc10/SW16, or ankyrin, repeats intracellularly). During June and July, 1991, a search for western prairie fringed orchid was conducted in the Platte River Valley in central and west-central Nebraska. No orchids were found in the 370 sites surveyed. Factors which may contribute to the scarcity of this orchid in the area surveyed include drought conditions in the previous three years; habitat alteration by draining, ditching, herbicide application, mowing for hay, crop cultivation and planting of non-native species for hay fields; invasion of the area by trees; and the presence of alkaline soils in some areas

Effect Of Temperature And Humidity On Evaporative Water Loss In Anna's Hummingbird (Calypte Anna), Donald R. Powers

Evaporative water loss (EWL), oxygen concumption (V0 ), and body temperature (Tb) of Anna's 2 Hummingbirds (Calypte anna; ca. 4.5 g) were measured at combinations of ambient temperature (T J and water vapor density (Qva) ranging from 20 to 37 °C and 2 to 27 g · m-3 , respectively. The EWL decreased linearly with increasing eva at all temperatures. The slopes ofleast squares regression lines relating EWL to Qva at different temperatures were not significantly different and averaged -0.50mg H20·m-3 ·g-2 ·h-1 (range: -0.39 to -0.61). Increased Qva restricted EWL in C. anna more than has been reported for …

The role of microorganisms in the degradation of xenobiotics in the environment is well established. Bacteria from the genus Pseudomonas are particularly well adapted to the degradation of hydrocarbons, aromatics, and numerous other natural and introduced substrates. We have isolated a strain of Pseudomonas putida, designated PC2P15, that uses toluene, phenol, benzene, and a number of other substrates as its sole sources of carbon and energy.

Modulation By Camp Of A Slowly Activating Potassium Channel Expressed In Xenopus Oocytes, Edward M. Blumenthal, Leonard K. Kaczmarek

When expressed in the Xenopus oocyte, the minK protein induces a slowly activating voltage-dependent potassium current (Isk). We studied the modulation of this current by altering intracellular cAMP levels and found that the amplitude of Isk is dramatically increased by treatments that raise cAMP levels and decreased by agents that lower cAMP levels. Preinjection of a protein inhibitor of the cAMP-dependent protein kinase blocked the effects of increased cAMP levels. There were no changes in the voltage dependence or kinetics of Isk. The purpose of this work was to examine the tRNATrp-encoding genes (tRNATrp) of Saccharomyces cerevisiae to gain insight as to why tRNATrp amber suppressors, isolated by conventional genetic techniques, have not been reported. The results herein indicate that the haploid yeast genome contains six tRNATrp genes which map to five or six chromosomes. Not only do the six genes have identical coding sequences but their introns are also identical. Gene replacement experiments indicate that five copies of tRNATrp are sufficient for cell viability. We investigated the costs of active female choice in sage grouse, Centrocercus urophasianus, a lekking species in which females make repeated, lengthy visits to leks to assess males before mating. Several potential costs were measured by monitoring changes in hens' ranging behavior, time budgets, and encounter rates with predators when they visited leks. Two costs were identified: hens moved further per day and encountered golden eagles, Aquila chrysaetos, more frequently when visiting leks. However, extra travel due to visiting leks increased predicted daily energetic expenditure by only about 1%, and the risk of predation by golden eagles over …

In a phase I trial, 12 patients with GD2 antigen-positive metastatic melanoma received the murine anti-GD2 monoclonal antibody 14G2a. The monoclonal antibody was administered in four doses over an 8-day period with total dose ranging from 10 to 120 mg. All patients receiving >10 mg of 14G2a experienced transient abdominal/pelvic pain during the antibody infusion. Five patients had a delayed extremity pain syndrome following the third and fourth antibody infusion. Four of the five patients developed neurological toxicity, including two patients with significant although reversible motor neuropathy. Two of the patients developed hyponatremia secondary to a syndrome of …

The entomopathogenic fungi produce a wide assortment of metabolites, some of which are important to the host-specialization of this group of fungi. Very few entomopathogenic fungi have been examined in detail for their metabolites in the disease process. Nevertheless, it is clear that certain proteases and lipases are crucial to the invasion of insect cuticle by the fungus. These fungi have the unusual ability to hydrolyze branched alkanes. Also, they normally produce very active, broad-spectrum proteases. These are needed for penetration of the hydrophobic epicuticle and the highly proteinaceous cuticle of insects. Chitinases also are involved, but usually have only …

Our previous work showed that pap pilin gene transcription is subject to a thermoregulatory control mechanism under which pap pilin is not transcribed at a low temperature (23°C) (L. B. Blyn, B. A. Braaten, C. A. White-Ziegler, D. H. Rolfson, and D. A. Low, EMBO J. 8:613-620, 1989). In order to isolate genes involved in this temperature regulation of gene expression, chromosomal mini-TnlO (mTnlO) mutations that allowed transcription of the pap pilin gene at 23°C were identified, and the locus was designated tcp, for "thermoregulatory control of pap" (C. The purpose of this study was to examine the effect of consumption of carbonated beverages on maximal aerobic capacity, as measured by maximal oxygen consumption (VO2MAX), and respiratory physiology, as measured by ventilation frequency (f), ventilatory equivalent for oxygen and carbon dioxide (O2VE, co2VE), expired minute ventilation (VE), carbon dioxide output (VCO2), respiratory exchange ratio (RER), and tidal volume (VT).
Thirteen male high school, collegiate and post collegiate distance runners from the local community performed two maximal treadmill tests exactly one week apart, following two different fluid ingestion treatments. The subjects were randomly assigned to a carbonated beverage or noncarbonated …

V79 Chinese hamster cells were collected by colcemid addition to study the effect of heat on mitosis. When they were heated at 42°C and 45°C in the presence of 0.06 μg/mL colcemid, cell survival increased over the control samples, which were heated in ordinary medium. Scanning electron microscopy showed that cells heated to 45°C in the presence or absence of colcemid had fewer microvilli on the surface, but they did not have increased bleb formation. Transmission electron microscopy showed that the chromatin was diffuse in the heated cells and the kinetochores were indistinct. The mitochondria in the heated cells were …

Determining the biological effects of low doses of radiation with high linear energy transfer (LET) is complicated by the stochastic nature of charged-particle interactions. Populations of cells exposed to very low radiation doses contain a few cells which have been hit by a charged particle, while the majority of the cells receive no radiation damage. At somewhat higher doses, a few cells receive two or more events. Because the effects of damage produced by separate events can interact in the cell, we have had to make assumptions about the nature of these interactions in order to interpret the results of …

The application of the scanning tunneling microscope (STM) and the atomic force microscope (AFM) to the study of small biological molecules, such as DNA and smaller molecules, has received considerable attention in the literature. This paper reviews STM and AFM studies of larger biological structures such as bacterial membranes, bacteriophages, viruses, antibodies, etc. The problems encountered in these applications are emphasized, with particular reference to the unknown conduction mechanism, tip-sample interaction forces, and tip-sample convolution artifacts in the images. The latter problem is illustrated by new results from IgG antibody complexes attached to a bacterial sheath layer. A new conduction …

The microvasculature of human tongue papillae originating from 9 males and 6 females, aged 0.5 to 2 years was studied by scanning electron microscopy (SEM) of vascular corrosion casts and by light microscopy (LM) of India ink injected specimens. All papillae showed a microvasculature characterized by primary, secondary and tertiary capillary loops. In the filiform papillae the loops were generally arranged in a corolla-like pattern with the tertiary loops demonstrating a hair-pin shape. The fungiform papillae showed basically a similar architectural pattern although the loops were somewhat more compact and complex in structure. A small, shallow depression of the tertiary …

Studies were undertaken to expand the versatility and the resolution of low temperature conventional and field emission scanning electron microscopy (SEM). The results indicated that simple modified specimen holders, which could be used in conjunction with the commercial cryosystems, allowed one to store specimens for several weeks in liquid nitrogen, either before or after observation in a conventional SEM, without incurring degradation of the surface features. Other modified holders permitted one to move the specimen closer to the final lens or to use the upper secondary electron detector, which is available with some SEMs. Both of these procedures increased the …

The general principles of the low-temperature spectroscopic cathodoluminescence (LTSCL) technique in the scanning electron microscope (SEM) are outlined. CL microscopy and spectroscopy are now being extended in new directions. Analysis of the intrinsic CL from quantum well materials by LTSCL is proving valuable for microcharacterization and such work can be expected to expand rapidly. The main, early CL results in this exciting field are summarized here. In geology, CL microscopy, without spectroscopy, is widely used via electron-beam attachments to light microscopes and the addition of spectral analysis to these studies is overdue. Much CL from minerals and ceramics comes from …

A comparative odontologic study was conducted on teeth and their supporting tissues from extant sharks, fossil sharks, and living lungfishes using scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) X-ray microprobe to investigate evolutionary changes in tooth structure of the fishes.
No histological differences between the extant and fossil teeth of lamnoid sharks were observed. The weight percent of calcium in osteodentin of the fossil shark was higher than that in the extant shark. The weight percents of both P and Ca of tooth tissues in the giant extinct shark showed comparatively low values.

I have investigated the possibility that specific conalbumin (ovotransferrin) iron saturation levels enable less virulent strains of Salmonella to become more virulent. Iron starved cells of two pathogenic Salmonella strains, S. paratyphi B var. java and S. thompson, were cultured in iron limited media at 3 different iron conalbumin saturation levels. Results indicate that strains differ significantly at both low and high iron saturation conalbumin. These differences depict a growth advantage for S. paratyphi B which correlates with reports by the Centers for Disease Control that S. paratyphi B was 3 times more frequent in blood isolates than S. …

Using interfacially-controlled crystallization and gel diffusion crystallization methods, calcium oxalate monohydrate (COM), dihydrate (COD) and trihydrate (COT) crystals were grown by the slow diffusion of reacting ions in the presence of mucin. It was demonstrated that mucin in the growth media dramatically affected the size, habit, surface structure, thermodynamic stability and phase transition kinetics of hydrated calcium oxalate crystals. The results obtained revealed that mucin as a glycoprotein model controlled the growth of COT and COD single crystals as well as cluster formation. Growth inhibition of specific crystal faces and phase transition retardation occurred in its presence. The data confirmed …

Panchromatic (integral) and spectrally resolved cathodoluminescence characterization was used to investigate the near surface gettering properties of Cu in liquid-encapsulated, Czochralski-grown, undoped semi-insulating (SI) GaAs. Samples from two sources were investigated to determine if gettering treatments applied to GaAs result in improvements in uniformity similar to those observed in gettered Si. Before Cu contamination, typical cellular structure is observed for all samples. Experimentally, it is found that the panchromatic CL images change significantly after Cu doping and subsequent gettering processing for all samples. A contrast reversal is generally observed after Cu contamination. After gettering, the image of the samples from …

An Ohio County, Kentucky, population of Tree Swallows utilizing nest boxes located over a strip pit lake on reclaimed Peabody Coal Company land was studied on a weekly basis throughout its breeding season. The season lasted from late April to late July.
