  • In ecology, desert rock pools form an ideal model ecosystem. These typically isolated pools a small enough that a researcher can observe the entire system and record everything going into and out of this aquatic system. This removes one of the most important confounding factors for observational studies in ecology, making rock pools an important system to understand.
  • The rock pools of the Colorado Plateau in the western United States consist of three main types: 1) scoured pools in temporary stream channels that violently flood during rainstorms (locally called Tinajas), 2) shorthydroperiod pools which are shallow depressions (< 0.6 m deep) that thaw in March and dry by the end of April , and 3) long hydroperiod pools which are deep pools (1-3 m) that thaw in March and persist until June.

Synovial sarcomas (SS) are rare soft tissue cancers that are found beside bone in children and adolescents. Currently, the prognosis for these diseases are low, as diagnostic and treatment options are limited due to the rarity of the disease. With high rates of metastasis, treatment options for these diseases become further limited to primarily radiation and chemotherapies. These therapies have limited specificity in their mechanisms, causing severe side effects in patients. This work examined alternative options for synovial sarcoma theranostic approaches, specifically using immunoconjugates for Oncostatin M Receptors (OSMR). Recent phylogenetic work within Hetaerininae based on Sanger data highlighted classification issues (e.g., non-monophyly between the genera), however no changes were proposed at that time. We explore the phylogeny of Hetaerininae using a targeted enrichment approach (AHE) of 20KB (18.5KB of nuclear and 1.5 of mitochondrial DNA) in an attempt to resolve any discrepancies.

Diabetes is a chronic metabolic disease characterized by an inability of beta cells to produce or secrete insulin due to decreasing beta cell mass, a condition induced by beta cell death or overuse. Current treatment consists of daily administration of insulin to diabetic individuals. We have shown that Sirtuin 7 (Sirt7), a deacetylase located in the nucleus, directly interacts with Nkx6.1, a transcription factor essential for beta cell function and proliferation. Background: Although the human bladder is reported to harbor unique microbiota, our understanding of how these microbial communities interact with their human hosts is limited, mostly owing to the lack of isolates to test mechanistic hypotheses. Niche-specific bacterial collections and associated reference genome databases have been instrumental in expanding knowledge of the microbiota of other anatomical sites, such as the gut and oral cavity. Results: To facilitate genomic, functional, and experimental analyses of the human bladder microbiota, we present a bladder-specific bacterial isolate reference collection comprising 1134 genomes, primarily from adult females. BACKGROUND: Acute exacerbations of idiopathic pulmonary fibrosis (AE-IPF) affect a significant proportion of patients with IPF. There are limited data to inform therapeutic strategies for AE-IPF, despite its high mortality. We discuss the rationale and design of STRIVE-IPF, a randomized, multi-center, open-label Phase IIb clinical trial to determine the efficacy of combined therapeutic plasma exchange (TPE), rituximab, and intravenous immunoglobulin (IVIG), in comparison to treatment as usual (TAU), among patients with acute IPF exacerbations.

METHODS: The STRIVE-IPF trial will randomize 51 patients among five sites in the United States. The inclusion criteria have been designed to select a study population …

Anaplastic thyroid cancer is a rare, fatal cancer with a five-year survival of 4%. Universally diagnosed at stage IV, anaplastic thyroid cancer is characterized by its lack of differentiation, rapid proliferative rate, highly inflammatory tumor microenvironment, and metabolic dysregulation. Refractory to all established therapies, anaplastic thyroid cancer requires a novel therapeutic approach that targets all of these drivers of anaplastic thyroid cancer carcinogenesis. Soil pH is a foundational element of agronomy, profoundly influencing biological, chemical and physical soil processes. Adjusting soil pH is a paramount factor for enhancing soil health and productivity with far-reaching environmental benefits. Over time soil naturally tends towards acidity, a process expedited by intensive agriculture practices. To determine the extent of necessary pH corrections, precise soil pH testing is imperative. Mycobacterium tuberculosis (Mtb)-specific γ9δ2 T cells secrete granzyme A (GzmA) protective against intracellular Mtb growth. However, GzmA-enzymatic activity is unnecessary for pathogen inhibition, and the mechanisms of GzmA-mediated protection remain unknown. We show that GzmA homodimerization is essential for opsonization of mycobacteria, altered uptake into human monocytes, and subsequent pathogen clearance within the phagolysosome. Although monomeric and homodimeric GzmA bind mycobacteria, only homodimers also bind cluster of differentiation 14 (CD14) and Toll-like receptor 4 (TLR4). Without access to surface-expressed CD14 and TLR4, GzmA fails to inhibit intracellular Mtb. Upregulation of Rab11FIP1 was associated with inhibitory activity. Furthermore, GzmA colocalized with …

Early life deprivation and stress can contribute to life-long, problematic consequences, including epigenetic variations related to behavior and health. Domestic dogs share human environments and social–cognitive traits, making them a promising comparative model to examine developmental plasticity. We examined 47 owner–dog dyads, including dogs rescued from abusive or neglectful environments, and matched control dogs for changes in DNA methylation of glucocorticoid (NR3C1) and oxytocin (OXTR) receptor genes previously shown to be affected by early life stress in other species including humans. Evolution over billions of years has led to unique animals of all types. The tiny sea butterfly is one that remains mostly anonymous because of its size and low place on the food chain, but it is a swimming creature that when examined closely reveals a surprise. Shockingly, it has a swimming motion nearly identical to small insects flying at intermediate Reynolds numbers, referred to as the clap-and-fling. The centimeter long pteropod flapping briskly at 5Hz escapes attention with its size and speed. Triple-negative breast cancer (TNBC) confers considerable interventional challenge owing to its massive heterogeneity, aggression and metastatic prowess. These facets are attributed to chromosomal instability (CIN), a feature consequent to TNBC cell’s ability to harbor more than two centrosomes (supernumerary centrosomes; SNCs).

In principle, if a normal cell possesses SNCs, uneven pull forces are generated consequent to multipolar spindle, which activates cellular surveillance. This ensures error-free mitosis by either repairing the error or, in an irreparable condition, driving cells to death. This averts the possibility of aneuploidy (fundamental to tumorigenesis).

Seasonal coronaviruses (HCoVs) are known to contribute to cross-reactive antibody (Ab) responses against SARS-CoV-2. While these responses are predictable due to the high homology between SARS-CoV-2 and other CoVs, the impact of these responses on susceptibility to SARS-CoV-2 infection in cancer patients is unclear. Urbanization poses significant challenges to wildlife, including amphibians. The present study uses a multi-parameter approach that incorporates veterinary techniques to investigate the current health status of the adult southern leopard frogs (Lithobates sphenocephalus) in Tampa Bay, Florida, amidst urban stressors including nutrient loading, fragmented habitats, and light and noise pollution. Novel values for fluctuating asymmetry (e.g., variation between left and right bilateral traits; FA), body condition (CF), stress (e.g., neutrophil:lymphocyte (N:L) ratio), and inflammation (e.g., neutrophil morphologic abnormalities, parasite load) are reported. Two species of clonal Typha [T. latifolia (native) and T. angustifolia (exotic)] hybridize to form the highly invasive, heterotic (high vigor) T. × glauca in North American wetlands leading to increased primary production, litter accumulation, and biodiversity loss. Conservation of T. latifolia has become critical as invasive Typha has overwhelmed wetlands. In the field, Typha taxa identification is difficult due to subtle differences in morphology, and molecular identification is often unfeasible for managers. Furthermore, improved methods to non-destructively estimate Typha biomass is imperative to enhance ecological impact assessments. Causes of colony failure in interior least terns (Sternula antillarum athalassos) have been a prevalent topic for researchers investigating this previously endangered species. In Arkansas, these colonies are primarily located on sandbars formed along the McClellan-Kerr Arkansas River Navigation System. Reduction in nesting site quality may put this sensitive species at a greater risk for predator encounters. Mediterranean forest ecosystems will be increasingly affected by hotter drought and more frequent and severe wildfire events in the future. However, little is known about the longer-term responses of these forests to multiple disturbances and the forests' capacity to maintain ecosystem function. This is particularly so for below-ground organisms, which have received less attention than those above-ground, despite their essential contributions to forest function. Ecologists and conservation biologists have become quite familiar with the concept of tipping points: abrupt changes in an ecosystem's state that occur after a period of relative stasis. Most of the familiar ecological examples of tipping points occur either because a once-stable state has lost stability, or the system has been subjected to a particularly large perturbation and transitions to an alternative stable state, distinct from the pre-perturbed state.
