Embryo Culture In Barley (Hordeum Vulgare L.), Betül Bürün, Esi̇n Çoban Poyrazoğlu

The effects of medium and sterilization on the mature embryo culture of barley (Hordeum vulgare L.) were studied. Among the different media used, the highest plantlet development from embryos was obtained in Randolph and Cox medium (92.1%), followed by Murashige and Skoog (MS) (91.3%), half-strength MS (1/2 MS) (87.7%) and B5 (85.7%). Among the sterilization methods studied, sodium hypochlorite sterilization followed by treatment of antibiotic solution and sterilization HgCl_2 application was found to be more effective than the other methods. However, no statistically significant negative effect of HgCl_2 on the plantlet development from embryos was observed.

Alkaline environments are typical extreme environments which include naturally occurring soda lakes, deserts, soils and artificially occurring industrial-derived waters. Micro-organisms that occupy extreme pH environments have resulted in the definition of an unusual group, termed alkaliphiles. In this review, the current status of the biodiversity of alkaliphilic micro-organisms in various environments and aspects of their biotechnological potential are summarised briefly.

Purification And Determination Of The Molecular Structure Of Hemolymph Lectin Of Agrotis Segetum (Denis And Schiff.), Nursel Gül, Cevat Ayvali

A soluble lectin was purified from the hemolymph of Agrotis segetum by two methods, via affinity chromatography on a Sepharose-4B column and gel filtration on a Superdex-200 column. Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE) showed a single protein band with a molecular weight of 69,000 in both the presence and absence of 2-Mercaptoethanol. Alternatively, the molecular weight of this lectin was estimated to be 69,000 by gel filtration on Superdex-200. Electron microscopic observation of the purified lectin was reticular in structure and consisted of tandem aligned spherical basic units.

The effects of a sublethal concentration of three plant growth regulators (PRGs) on serum enzymes in rats were investigated under laboratory conditions. 100-ppm of PRGs, indoleacetic acid (IAA), indolebutiric acid (IBA) and kinetin were administered orally to 8 rats ad libitum for 3 weeks. The hormon treatments caused different effects on the level of serum alanin aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), amylase and creatine phosphokinase (CPK) in comparison to those of control rats. The effects of chronic treatment with small sublethal doses (20 mg/kg and 50 mg/kg) of Teucrium polium ethanolic extract were investigated on the hematological and biochemical composition of the blood, histopathology of the liver and kidney, and fertility in the rat. Hematological and biochemical parameters of the blood, as well as sperm count, morphology and motility were normal after 6 weeks of herbal treatment. However, blood urea (P < 0.05) and cholesterol (P < 0.005) were significantly increased, with marked cytoplasmic vacuolation of the liver and kidney cells after chronic treatment with 50 mg/kg of the plant. These results demonstrate some histopathological effects of T. polium on the liver and kidney under long-term administration conditions.

Rates of growth and sexual maturation of Mongolian gerbils vary in response to photoperiod. Previous work has shown that the responses of the testes and body weight of juvenile and adult gerbils are influenced not only by the photoperiods but also by other factors. The present work sought to determine if Mongolian gerbils alter body mass, body composition and/or reproductive organ mass in response to different photoperiods and temperatures. In this study, it was aimed to increase the frequency of shoots, which would be nominated to be transgenic after Agrobacterium inoculation, through expanding the wounding area by peeling the epidermis layer of hypocotyl explants of flax plants. Callus weight, percentage of hypocotyls producing shoots, shoot number per explant and total shoot number per petri dish were recorded as 0.8 g, 40%, 4.24 and 16.96, respectively in non-peeled explants. The conjugation of glutathione with herbicides and insecticides is considered to be one of the major detoxification mechanisms of plants. This conjugation is catalysed by a family of multifunctional proteins known as glutathione-s-transferases. Treatment of wheat (Triticum aestivum L. cv. Cumhuriyet-75), barley (Hordeum vulgare L. cv. Kaya), chickpea (Cicer arietinum L. cv. Ispanyol), and lentil (Lens culinaris Medik. cv. Kışlık-kırmızı-51) with the selective herbicides 2,4-D (2,4-Dichloro phenoxyacetic acid) and linuron (N-(3,4-dichlorophenyl)-N´-methoxy-N´-methyl urea), and insecticides parathion methyl (O,O-dimethyl O-(4-nitrophenyl) phosphorothioate) and malathion (S-1,2-bis(ethoxycarbonyl)ethyl O,O-dimethyl phosphorodithioate) caused a progressive increase in glutathione-s-transferase activity in roots and shoots. This study was carried out to determine the effects of heavy metals (Ni, Fe, Pb, Co, Cd, Hg, Al, Zn and Cu) on pollen germination and pollen tube length in the tobacco plant (Nicotiana tabacum L.) cv. Karabağlar. The results showed that enhanced concentrations of heavy metals, except Fe, decreased the pollen germination rates and the pollen tube lengths. With Fe concentrations, on the other hand, first a positive, and then a negative relation was determined between the pollen characteristics examined. Peptone from ram horn was compared with a casein and other peptones for bacterial growth. First, horns were ground and 35 g of horn flour was hydrolyzed chemically (acid hydrolysis). As a result of this process, 30 g of the 35 g horn flour (85.7%) could be hydrolyzed. Hydrolyzed material was completed to 400 ml with deionized water, and this resulting solution was termed ram horn hydrolysate (RHH). The contents of protein, nitrogen, ash, some minerals, total sugars, total lipids and amino acids of RHH were determined. Simple sequence repeat (SSR) DNA polymorphism is a PCR-based marker system, also known as the microsatellite marker system. SSRs are tandemly repeated 1 to 5 nucleotide units dispersed throughout eukaryotic genomes. They are considered one of the most powerful molecular markers for many diverse applications in genome related studies, e.g., DNA fingerprinting for discrimination of genotypes and genetic mapping. In this study, ribotyping (restriction polymorphism ribosomal RNA analyses) was applied as a taxonomic tool for bacteria belonging to the genus Nocardia. A total of 21 Nocardia strains were clearly distinguished using a combination of EcoRV gene restriction endonuclease patterns together with a digoxigenin-labelled Streptomyces violaceoruber TK21 rDNA probe. Five distinct ribotypes were identified, and each ribotype contained 4 to 8 restriction fragments ranging in size from 1.2 to 20.7 kb. The evaluation of the results showed that N. brasiliensis, N. farcinica, N. otitidiscaviarum and N. seriolea are taxonomically well-established species, except N. pseudobrasiliensis. Three experimental diets containing 14.9% (low carbohydrate; LC), 31.4% (medium carbohydrate; MC) and 42.3% (high carbohydrate; HC) of extruded wheat meal were fed to rainbow trout, Oncorhynchus mykiss, to investigate the significance of carbohydrate level on the rates of gastric evacuation and return of appetite (feed intake). Gastric evacuation measurements were performed by serial slaughter and recovery of digesta whilst X-radiography was used to estimate the rates of return of appetite. Hypocotyl and stem explants excised from in vitro and greenhouse-grown seedlings of Madaras, 1886 Sel. and Omega flax cultivars were compared in terms of percentage of explants producing shoots, shoot number per explant and total shoot number per petri dish. Hypocotyl explants resulted in better shoot regeneration. In vitro-grown seedlings were found to be more suitable than greenhouse-grown seedlings as an explant source.

Forty strains were isolated from six different soil samples. The soil samples were taken from the grasslands of Elmadağ, Çubuk, Gölbaşı, Haymana, Polatlı and Kazan near Ankara. As a result of the identification tests, 30 strains were identified as Bacillus subtilis, Bacillus megaterium, Bacillus firmus, Bacillus sphaericus, Bacillus thuringiensis, Bacillus pumilus and other Bacillus spp. The inhibitory effect of Bacillus strains against the test bacteria Escherichia coli, Staphylococcus aureus, Yersinia enterocolitica and Micrococcus flavus was examined. It was found that B. thuringiensis D1, B. thuringiensis D3 and B. megaterium Y6 strains showed an inhibitory effect on all of the test …

We present data on freshwater mussel (Mollusca: Bivalvia: Unionidae) distributions for Tinkers Creek, a small Ohio stream that previously had not been surveyed for its unionid fauna. The distribution of these mussels was mapped against the changing habitat of the stream, with special attention paid to two zones of human manipulation, a wastewater treatment plant and a stretch of the river where the bed was relocated to enable development. At least five unionid species live in Tinkers Creek. These are Pyganodon grandis, Lampsilis radiata luteola, Fusconaiajlava, Lasmigona costata, and Lasmigona compressa. We provide new information from archaeological samples on the historical freshwater mussel fauna (Mollusca: Bivalvia: Unionoidea) of the Cuyahoga River (South Park site: occupied between ca. A.D. 950 and 1650) and Black River (White Fort site, occupation centered at ca. A.D. 1350), northeast Ohio. Data from these prehi storic sites are compared with information on extant mussel populations of the Cuyahoga River published between 1890 and 2000. The high representation at both archaeological sites of the species Actinonaias ligamentina, Elliptio dilatata, and Ptychobranchus fasciolaris suggests that these were· among the important clean water species in northeast Ohio prior to European …

An unusually well preserved skull, mandible, and indisputably associated post-cranial elements of new sloth, Acratocnus (Miocnus), were recovered from a cave in Jaragua National Park, Dominican Republic. The animal died lying in a rimstone pool and was rapidly coated with a thin calcite patina. We have documented a late Wisconsinan age for this specimen by inorganic 14C radiometric dating of the patina, with supporting data on the carbon systematics of the speleothems in this cave. The extinct oryzomyine rodent Megalomys curazensis has been known from abundant but fragmentary remains on the island of Curaçao since 1959. Here we demonstrate an age of 130 000 to 400 000 years before present based on geomorphological context, and propose a biogeographical model for the genus.

A Late Quaternary Paleoecological Record From Caves Of Southern Jamaica, West Indies, Donald A. Mcfarlane, Joyce Lundberg, A.G. Fincham

Studies of an unusual and diverse system of caves in coastal southern Jamaica have yielded a paleoclimatic record associated with a fossil vertebrate record that provides useful insights into the poorly documented paleoecology of latest Wisconsinan and Holocene Jamaica. Episodes of significantly increased precipitation during the Holocene have left characteristic deposits of speleothems, and have supported both faunal and archaeological communities that were dependent on these mesic conditions. Deposits of fossil bat guano preserved in the caves provide a δ¹³C record of alternating mesic and xeric climatic episodes that supports the interpretation of the faunal and archaeological record.

The seed coat of Stylistna pickeringii (Torr.) Gray var. pattersoni (Patterson bindweed), an endangered species of Illinois sand prairies, inhibits water uptake and seed germination. The purpose of this research was to find an effective and efficient way to scarify seeds of S. pickeringii to aid reintroduction into its natural habitat. Seeds were collected from sandy areas close to the Illinois River near Snicarte (Mason Co.), Illinois during the summers of 1998 and 1999. Urinary tract infections are one of the most common causes of ambulatory care visits, physician-office visits and hospitalization each year in the US. Sexual activity primarily contributes to acquisition of UTI in women; nearly half of adult women have had a UTI at least once. The most common causative agent is E. coli, followed by Enterobacter spp., Klebsiella spp., Pseudomonas aeruginosa, Proteus mirabilis, other Gram-negative rods and Group B streptococci, depending on geographical location. 1 Until the late 1990's, trimethoprim-sulfamethoxazole (TMP-SMX) was the most commonly prescribed and effective treatment option, but the rampant development of resistance to this drug has …

Diagnosis of far lateral disc herniations has become more common in recent years. This study describes the surgical procedure used for L5-S1 far lateral disc herniations, shows the benefits derived from a far lateral approach and retrospectively evaluates the outcome of the surgery. Eight patients were included in the study, five females and three males. The mean age was 62 years (range, 45-77 years). VAS pain scale, OSW index and SF-36 forms were used to evaluate the surgical outcome. Both the VAS pain scale and OSW index were improved postoperatively. The SF-36 scores were significantly lower (p<0.05) for the far lateral patients compared to both the low back pain population and the U.S. aged 55-64 year population.

Hyperbaric oxygen therapy promotes faster tissue healing by increasing the amount of oxygen carried to the tissues. HBO is currently used to treat carbon monoxide poisoning as well as smoke inhalation. Based on this information, it was believed that HBO would also promote faster healing for hydrogen cyanide gas or hydrogen chloride gas inhalation, which are encountered in burning material. To test this theory, mice were exposed to HCN or HCl gas. The mice were then placed in hyperbaric oxygen or supplemental oxygen treatments for five days. Aspergillus flavus Link ex Gray and Cladosporium cladosporioides (Fresen.) de Vries, isolated from the soil of Florya Atatürk Forest, were extracted and some toxicity tests were carried out in order to determine their toxic effects on mice for the first time in Turkey. The method of extraction complied with the one described in the literature. During the process, Coca's solution was used as an extractive agent. Changes in developmental patterns are some of the most important effects that may be observed at radioactively contaminated sites like those at Chornobyl. Developmental instability may arise from the interactions between an organism's genotype and its environment and be manifested as deviant morphology. Fluctuating asymmetry (FA) is a measure of deviations from the expected bilateral symmetry of the body. Our objective was to test for differences in FA in two rodent species (Apodemus flavicollis and Clethrionomys glareolus) that live in habitats suirounding the failed Chornobyl reactor. Nesting male gulf toadfish Opsanus beta produce a boatwhistle advertisement call used in male–male competition and to attract females and an agonistic grunt call. The grunt is a short-duration pulsatile call, and the boatwhistle is a complex call typically consisting of zero to three introductory grunts, a long tonal boop note, and zero to three shorter boops. The beginning of the boop note is also gruntlike. Anomalous boatwhistles contain a short-duration grunt embedded in the tonal portion of the boop or between an introductory grunt and the boop. Cohesion between sister chromatids is a prerequisite for accurate chromosome segregation during mitosis and meiosis. To allow chromosome condensation during prophase, the connections that hold sister chromatids together must be maintained but still permit extensive chromatin compaction. In Drosophila, null mutations in the orientation disruptor (ord) gene lead to meiotic nondisjunction in males and females because cohesion is absent by the time that sister kinetochores make stable microtubule attachments. We provide evidence that ORD is concentrated within the extrachromosomal domains of the nuclei of Drosophila primary spermatocytes during early G2, but accumulates on the meiotic chromosomes by mid to late …
