Characterizing The Quorum Sensing (Qs) System In The Model Eukaryote Chlamydomonas Reinhardtii, Kirstin Donnette Cutshaw
Characterizing The Quorum Sensing (Qs) System In The Model Eukaryote Chlamydomonas Reinhardtii, Kirstin Donnette Cutshaw
Theses and Dissertations
Group behaviors in microorganisms are often regulated by low-molecular weight molecules that enable them to sense population density, a signaling system known as quorum sensing (QS). QS in prokaryotes has provided insight into the role ‘social’ behaviors play in the bacterial world and refined our models of virulence, bacterial motility, and more. While less common, similar behaviors are observed in certain fungi, primarily yeasts, in which QS controls cell morphology, biofilm production, and other phenotypes. We have recently pursued the potential for QS to be more broadly distributed among unicellular eukaryotes, which could significantly alter our understanding of social behaviors …
Investigating Microbe-Plant Symbioses In Space, David Handy
Investigating Microbe-Plant Symbioses In Space, David Handy
Theses and Dissertations
As life has evolved on Earth, gravity has been a constant around which biological processes have developed, rather than a pressure that has been adapted to. As humanity ventures into space, we remove this constant and can observe a variety of changes in biological systems, such as bone loss in humans, stunted plant growth, or increased virulence in pathogens. The growing body of data shows that life adapts to spaceflight and microgravity in unique ways that, due to our limited understanding, can be difficult to predict. A particularly under-studied area is that of host-microbe interactions. Despite efforts to limit their …

The Influence Of Recycled Water On Microbial Source Tracking Techniques That Aim To Identify Untreated Sewage In Surface Waters, Aldo E. Lobos
The Influence Of Recycled Water On Microbial Source Tracking Techniques That Aim To Identify Untreated Sewage In Surface Waters, Aldo E. Lobos
USF Tampa Graduate Theses and Dissertations
Fecal pollution in recreational waters can introduce pathogens that cause increased human health risk, where exposure can occur during recreational activities such as swimming, diving, surfing, and fishing. Furthermore, human fecal pollution (i.e., sewage) can introduce nutrients and other harmful chemicals that may negatively affect the environment and aquatic species. Generally, sewage is considered the highest risk to human health compared to animal fecal pollution because it typically contains a high diversity of pathogens (including human specific viruses), antibiotic resistant bacteria and genes associated with antibiotic resistance. However, pathogens are difficult to detect due to their low concentrations in the …

Roles Of Mitochondrial Chchd2 And Gpcr-Associated Β-Arrestin2 In The Pathogenesis Of Lewy Body Disorders, Teresa R. Kee
USF Tampa Graduate Theses and Dissertations
Coiled-coil-helix-coiled-coil-helix domain containing 2 (CHCHD2) is a mitochondrial protein that plays important roles in cristae structure, oxidative phosphorylation, and apoptosis. Multiple mutations in CHCHD2 have been associated with Lewy body disorders (LBDs), such as Parkinson’s disease (PD) and dementia with Lewy bodies (DLB), with the CHCHD2T61I mutation being the most widely studied. We generated the first transgenic mouse models expressing human CHCHD2WT and the PD-linked CHCHD2T61I mutation driven by the mPrP promoter. CHCHD2T61I Tg, but not CHCHD2WT Tg mice exhibit perinuclear mitochondrial aggregates, neuroinflammation, and have impaired long-term synaptic plasticity associated with synaptic dysfunction. Dopaminergic neurodegeneration, a hallmark of PD, …

The Influence Of Childhood Trauma On Dna Methylation Patterns And Sex-Specific Cortisol Stress Reactivity: A Secondary Analysis, Isabella F. Correia
USF Tampa Graduate Theses and Dissertations
Childhood trauma has been shown to significantly impact physical and mental health outcomes, leading to conditions such as heart disease, diabetes, depression, and difficulties in decision-making and maintaining relationships. Exposure to such trauma has the potential to dysregulate an individual’s stress response system, resulting in abnormal cortisol levels. Many studies suggest a lasting impact of early-life stress on DNA methylation patterns and, separately, cortisol stress reactivity. In addition, there are known sex differences in exposure to childhood trauma and cortisol reactivity; yet studies to date have not investigated whether there is an association to DNA methylation. This study utilized a …
Feather Mites Through Genomic And Ecological Lenses: Host Specificity And Symbiosis At Multiple Levels Of Organization, Alix Elizabeth Matthews
Feather Mites Through Genomic And Ecological Lenses: Host Specificity And Symbiosis At Multiple Levels Of Organization, Alix Elizabeth Matthews
Student Theses and Dissertations
Symbionts, or organisms that form close and persistent associations with hosts, are ubiquitous across the tree of life, and symbioses are among the most common interspecific interactions in the natural world. Symbionts play a fundamental ecological and evolutionary role across multiple levels of biological organization – from individuals to ecosystems – and can have strong effects on global biodiversity. However, many aspects of these relationships are poorly understood including symbiont genetic diversity (inter- and intraspecific), host specificity, and the mechanisms underlying their functional nature or any context-dependent patterns. This dissertation uses a model symbiosis between permanent, obligate, ectosymbiotic vane-dwelling feather …

Salt Pollution Reduces Turbidity, Dissolved Organic Matter, And Cyanobacteria In Experimental Vernal Pool Communities, Jared P. Vigil, Matthew S. Schuler
Department of Biology Faculty Scholarship and Creative Works
Anthropogenic activities such as the over-application of road deicers are causing an increase in the concentration of salts in historically fresh waters. Experimental and field investigations demonstrate that freshwater salinization disrupts ecosystem functions and services, causing the death of freshwater organisms and changes to nutrient conditions. Wetland habitats are one system negatively affected by salt pollution, including ephemeral wetlands (vernal pools) that fill with salt-polluted water after snowmelt. In urbanized areas, the degradation of these ecosystems could result in irreversible ecological damage including reduced water quality and a reduction in biodiversity. To investigate the effects of freshwater salinization on vernal …

A Model Of Time-Dependent Macromolecular And Elemental Composition Of Phytoplankton, Anne Willem Omta
Faculty Scholarship
Phytoplankton Chl:C:N:P ratios are important from both an ecological and a biogeochemical perspective. We show that these elemental ratios can be represented by a phytoplankton physiological model of low complexity that includes major cellular macromolecular pools. In particular, our model resolves time-dependent intracellular pools of chlorophyll, proteins, nucleic acids, carbohydrates/lipids, and N and P storage. Batch culture data for two diatom and two prasinophyte species are used to constrain parameters that represent specific allocation traits and strategies. A key novelty is the simultaneous estimation of physiological parameters for two phytoplankton groups of such different sizes. The number of free parameters …

Environmental variation commonly drives functional trait diversity within species, among species, and across communities. Climate and shared evolutionary history can both influence trait–environment relationships. We studied variation in plant functional traits among closely related Costus species occurring across environmental gradients, the extent to which this variation occurs within single species, and how that variation may be influenced by shared evolutionary history. We measured leaf, aboveground stem, rhizome, and fine root traits of 17 species of Costus in eight sites in Costa Rica and Panama, which varied in elevation, temperature, and precipitation. We then assessed the relationships among traits and environmental …

Metals and metalloids (hereafter, metal(loid)s) in plant-based foods are a source of exposure to humans, but not all metal(loid)-food interactions are the same. Differences exist between metal(loid)s in terms of their behavior in soils and in how they are taken up by plants and stored in the edible plant tissue/food. Thus, there cannot be one consistent solution to reducing toxic metal(loid)s exposure to humans from foods. In addition, how metal(loid)s are absorbed, distributed, metabolized, and excreted by the human body differs based on both the metal(loid), other elements and nutrients in the food, and the nutritional status of the human. …

Prochlorococcus Death By A Ubiquitous Zooplankton Filter Feeder, Avery Harman
University Honors Theses
Prochlorococcus is a globally abundant cyanobacterium that can help reveal the ecology of microorganisms in the ocean. Specifically, mortality by zooplankton such as appendicularians is globally important and Prochlorococcus can help reveal some of the choices made around feeding for this ubiquitous filter feeder. Two ecotypes of Prochlorococcus were introduced to appendicularians to determine if appendicularian feed selectively between these ecotypes. Further, a single ecotype of Prochlorococcus was fed to appendicularians over a variety of life stages to determine if retention rate changed with appendicularian life stage. In this paper, we introduce the numerical strategy for mixed uncertainty propagation based on probability and Dempster–Shafer theories, and apply it to the computational model of peristalsis in a heart-pumping system. Specifically, the stochastic uncertainty in the system is represented with random variables while epistemic uncertainty is represented using non-probabilistic uncertain variables with belief functions. The mixed uncertainty is propagated through the system, resulting in the uncertainty in the chosen quantities of interest (QoI, such as flow volume, cost of transport and work). With the introduced numerical method, the uncertainty in the statistics of QoIs will be represented using belief …

The red-eared slider is considered one of the most ubiquitous freshwater turtles globally. Several key ecological advantages and life history traits of this species jointly impact growth rates and survivorship of native freshwater turtles where sympatry occurs. Evaluation of current management actions for red-eared slider turtles is limited, resulting in a paucity of information to guide management for the species and conservation of declining native freshwater turtles. We used previously collected mark-recapture and removal sampling data from sites with sympatric populations of red-eared slider and western painted turtles. We examined the effects of control (trapping and removal) of red-eared sliders …
