Clinical presentation of congenital heart disease is heterogeneous, making identification of the disease-causing genes and their genetic pathways and mechanisms of action challenging. By using in vivo electrocardiography, transthoracic echocardiography and microcomputed tomography imaging to screen 3,894 single-gene-null mouse lines for structural and functional cardiac abnormalities, here we identify 705 lines with cardiac arrhythmia, myocardial hypertrophy and/or ventricular dilation. Among these 705 genes, 486 have not been previously associated with cardiac dysfunction in humans, and some of them represent variants of unknown relevance (VUR). Mice with mutations in Casz1, Dnajc18, Pde4dip, Rnf38 or Tmem161b genes show developmental cardiac structural abnormalities, …

Karbala International Journal of Modern Science

A simple green chemistry approach was used to synthesize β-carotene from edible carrot without using solvent and heat. In this study, beta-carotene was extracted from edible carrots through a natural extraction method with several physical technical steps. The extracted sample was isolated using high-performance liquid chromatography (HPLC). A β-carotene peak was detected from HPLC with an absorbance maximum (2.135 mAU). The absorption of UV-Visible spectrum indicated that beta-carotene absorbs most strongly between 230-300 and 400-500 nm. The functional groups, namely, C=C (1649 cm-1), =C-H (900-900 cm-1) vibrations of antisymmetric deformation of CH3 groups and CH …

Recent studies have described a direct relationship between the illegal wildlife trade (IWT) and the prevalence of zoonotic pathogens in human populations. In the Philippines, the Philippine Integrated Disease Surveillance and Response (PIDSR) framework outlines the monitoring, response, and management of disease outbreaks, but needs to be updated in the wake of zoonoses from IWT. Here, we identified zoonotic pathogens that may be introduced to human populations through the IWT, pinpointed potential outbreak hotspots, and provided recommendations on how to improve the Philippines’ public health response while considering One Health. Using seizure data from the Biodiversity Management Bureau (DENR-BMB) covering …

Maternal methyl donor supplementation during pregnancy has demonstrated lasting influence on offspring DNA methylation. However, it is unknown whether an adverse postnatal environment, such as high-fat (HF) feeding, overrides the influence of prenatal methyl donor supplementation on offspring epigenome. In this study, we examined whether maternal supplementation of choline (CS), a methyl donor, interacts with prenatal and postnatal HF feeding to alter global and site-specific DNA methylation in offspring. We fed wild-type C57BL/6J mouse dams a HF diet with or without CS throughout gestation. After weaning, the offspring were exposed to HF feeding for 6 weeks resembling a continued obesogenic …

Variation In American Bison (Bison Bison) Ecological Functionality Across Management Sectors: Implications For Conducting Conservation Assessments, Luke Rollie Rogers Jan 2022

Variation In American Bison (Bison Bison) Ecological Functionality Across Management Sectors: Implications For Conducting Conservation Assessments, Luke Rollie Rogers

Biology Theses, Dissertations, and Student Creative Activity

The American bison (Bison bison subspp.), once on the verge of extinction, now number in the hundreds of thousands. Despite an understanding of their numerical recovery, a comprehensive survey of bison herds and how their management varies between sectors has never been completed, and neither has a critical evaluation of the level of ecological recovery of the species. I surveyed bison managers from all major sectors, collecting extensive information about each bison situation. I identified significant proportional differences between management sectors among my survey questions using Freeman-Halton-Fisher exact tests of independence. I visualized and grouped individual herds based on their …

Characterizing The Impact Of Postharvest Temperature Stress On Polyphenol Profiles Of Red And White-Fruited Strawberry Cultivars, Alyssa N. Smith Jan 2022

Characterizing The Impact Of Postharvest Temperature Stress On Polyphenol Profiles Of Red And White-Fruited Strawberry Cultivars, Alyssa N. Smith

USF Tampa Graduate Theses and Dissertations

All living organisms have developed mechanisms that help them prevent internal stress and survive under harsh conditions. For fruits, specifically strawberries, stress is fought against primarily by increasing the synthesis of polyphenols, which are secondary plant metabolites with bioactive activity. Characterizing these bioactive compounds and the differences between strawberry cultivars can be vital for strawberry breeders. Furthermore, understanding the mechanisms that trigger the synthesis of polyphenols and their levels within different strawberry cultivars will provide breeders with tools to successfully identify cultivars with higher resistance to pre- and postharvest stressors. To gain that understanding, this study aimed to characterize the …

Ecology Of Periphyton In A Subtropical River Floodplain, Kamela De Gallardo Jan 2022

Ecology Of Periphyton In A Subtropical River Floodplain, Kamela De Gallardo

LSU Doctoral Dissertations

The Atchafalaya River Basin (ARB) in southcentral Louisiana, USA, is a large and biologically diverse floodplain surrounding the Atchafalaya River (AR), which is the largest distributary of the Mississippi River, receiving 30% of the combined daily discharge of the Mississippi and Red Rivers. Annual flooding facilitates exchange between the AR and its floodplain and is thought to give rise to the high productivity of the river-floodplain system. Primary production within the aquatic ARB is driven by periphytic algae, phytoplankton, and aquatic macrophytes, however, very little is known about periphytic algal assemblages in floodplain systems. In this study, artificial substrates were …

Characterizing Human Gut Bacterial Tir Domains In Vitro, In Gnotobiotic Mice And Malnourished Children, James Stephen Weagley Jan 2022

Characterizing Human Gut Bacterial Tir Domains In Vitro, In Gnotobiotic Mice And Malnourished Children, James Stephen Weagley

Arts & Sciences Graduate Student Theses and Dissertations

Worldwide, malnutrition (undernutrition) is the leading cause of death in children under 5. Studies of healthy members of birth cohorts from low- and middle-income countries have identified a program of gut microbial community development that is disrupted in children with severe acute malnutrition (SAM) and moderate acute malnutrition (MAM). This perturbed gut microbiome development has been linked to the pathogenesis of malnutrition. Repair of this gut microbial dysbiosis in children with MAM with microbiota-directed complementary foods (MDCFs) designed to increase the fitness and expressed beneficial functions of underrepresented bacterial community members restores healthy growth, including improvements in mediators of metabolic …

Maternal Provisioning Of Alkaloid Defenses Are Present In Obligate But Not Facultative Egg Feeding Dendrobatids, Emily D. Villanueva, Olivia L. Brooks, Sarah K. Bolton, Nina Savastano, Lisa M. Schulte, Ralph Saporito

2022 Faculty Bibliography

Poison frogs sequester alkaloid defenses from a diet of largely mites and ants. As a result, frogs are defended against certain predators and microbial infections. Frogs in the genus Oophaga exhibit complex maternal care, wherein mothers transport recently hatched tadpoles to nursery pools and return regularly to supply developing tadpoles with unfertilized (nutritive) eggs. Developing tadpoles are obligate egg feeders. Further, female O. pumilio and O. sylvatica maternally provision their nutritive eggs with alkaloid defenses, providing protection to their developing tadpoles at a vulnerable life-stage. In another genus of poison frog, Ranitomeya, tadpoles only receive and consume eggs facultatively, and …

Alkaloid-Based Chemical Defenses In 11 Species Of Bufonid Poison Frogs From Brazil (Bufonidae: Melanophryniscus), Emily Staufer

Masters Theses

The term ‘poison frog’ describes members of five anuran families that are capable of sequestering lipophilic, alkaloid-based chemical defenses from their prey. Alkaloid defenses can vary widely within and among poison frog species, mostly linked to differences in availability of arthropod prey at different scales. While research has mainly focused on alkaloid variation within the dendrobatid and mantellid poison frogs, bufonid poison frogs in the genus Melanophryniscus demonstrate high inter- and intra-species alkaloid variation, and some members are capable of both sequestering dietary alkaloids and synthesizing alkaloids metabolically. However, of the 31 species of Melanophryniscus, only eight have ever had …

Textile dyes in wastewater can be harmful pollutants when released into the environment without treatment. Biodegradation of textile dye effluents by different microbes, including fungi, has become popular as an alternative to physicochemical methods. The mangrove Avicennia marina is known to harbor endophytic fungi which have the potential to carry out dye degradation. Therefore, this study assessed the ability to decolorize synthetic dyes of endophytic fungi isolated from the leaves of A. marina. Of the nine fungal endophytes, Aspergillus niger, Syncephalastrum racemosum and Penicillium citrinum exhibited the highest mycelial growths in solid media, while all endophytes adsorbed Congo red. Blueberries (Vaccinium spp.) are valued for their high nutritional quality and flavor. There are five species in the Vaccinium genus native to the state of Florida, but only three are palatable to humans: V. myrsinites, V. darrowii, and V. corymbosum. While V. darrowii has been studied extensively due to its successful hybridization with northern highbush blueberry cultivars, V. myrsinites and wild V. corymbosum have been understudied. This study aims to 1) quantify the abundance of various phytochemicals in fruits of these wild blueberry species and compare fruit quality both among the wild taxa and two varieties of …

The Nutritional Condition Of Moose Co-Varies With Climate, But Not With Density, Predation Risk, Or Diet Composition, Jennifer S. Forbey, Daniel P. Melody Jan 2022

The Nutritional Condition Of Moose Co-Varies With Climate, But Not With Density, Predation Risk, Or Diet Composition, Jennifer S. Forbey, Daniel P. Melody

Biology Faculty Publications and Presentations

A fundamental question about the ecology of herbivore populations pertains to the relative influence of biotic and abiotic processes on nutritional condition. Nutritional condition is influenced in important, yet poorly understood, ways by plant secondary metabolites (PSMs) which can adversely affect a herbivore’s physiology and energetics. Here we assess the relative influence of various abiotic (weather) and biotic (intraspecific competition, predation risk and diet composition) factors on indicators of nutritional condition and the energetic costs of detoxifying PSMs for the moose population in Isle Royale National Park (U.S.A.). Specifically, we observed interannual variation in the ratio of urea nitrogen to …

Effects Of Clipping Of Flight Feathers On Resource Use In Gallus Gallus Domesticus, Renee Garant, Bret W. Tobalske, Neila Bensassi, Nienke Van Staaveren, Dan Tulpan, Tina Widowski, Donald R. Powers, Alexandra Harlander-Matauschek

Faculty Publications - Department of Biological & Molecular Science

Ground-dwelling species of birds, such as domestic chickens (Gallus gallus domesticus), experience difficulties sustaining flight due to high wing loading. This limited flight ability may be exacerbated by loss of flight feathers that is prevalent among egg-laying chickens. Despite this, chickens housed in aviary style systems need to use flight to access essential resources stacked in vertical tiers. To understand the impact of flight feather loss on chickens’ ability to access elevated resources, we clipped primary and secondary flight feathers for two hen strains (brown-feathered and white-feathered, n = 120), and recorded the time hens spent at elevated resources (feeders, …

Transitions In Bacterial Communities Across Two Fermentation-Based Virgin Coconut Oil (Vco) Production Processes, Zomesh A N Maini, Crisanto M. Lopez

Biology Faculty Publications

Despite being one of the most used methods of virgin coconut oil (VCO) production, there is no metagenomic study that details the bacterial community shifts during fermentation-based VCO production. The identification and quantification of bacteria associated with coconut milk fermentation is useful for detecting the dominant microbial genera actively involved in VCO production which remains largely undescribed. Describing the constitutive microbial genera involved in this traditional fermentation practice can be used as a preliminary basis for improving industrial practices and developing better fermentation procedures. In this study, we utilized 16S rRNA metagenomic sequencing to trace the transitions in microbial community …

Multiomic Hypotheses Underlying Behavioral Manipulation Of Camponotus Floridanus Ants By Ophiocordyceps Camponoti-Floridani Fungi, Ian Will

Electronic Theses and Dissertations, 2020-2023

Parasitic manipulation of host behavior lies at the intersection of disease, animal behavior, and coevolutionary processes. In many of these interactions, the underpinning biology is brought into sharp focus as they are obligate relationships, under strong selection to bring about specific changes in host behavior that determine if the parasite will transmit or die. However, experimental and molecular techniques to understand these interactions are still developing and identification of the mechanisms of manipulation is a primary goal in the field. As such, we investigated host-parasite interactions between Camponotus floridanus (Florida carpenter ant) and Ophiocordyceps camponoti-floridani (Florida zombie ant fungus) from …

Effect Of Metal-Containing Nanoparticles On Bacterial Biofilms And On The Microbiome Of Girardia Tigrina, Azar Saikali Jan 2022

Effect Of Metal-Containing Nanoparticles On Bacterial Biofilms And On The Microbiome Of Girardia Tigrina, Azar Saikali

Modeling The Flow And Creep Compliance Properties Of Ice-Cream Mixes, Hiran Ranaweera

Electronic Theses and Dissertations

This work documented the influence of the protein source (MPC80 and WPC80), protein content (4-12%), and temperature (5-35ºC) on the rheological behavior (flow curve, frequency sweep, and creep-recovery) of ice-cream mixes (ICM). For each protein source, the viscosity of the ICM was satisfactorily modeled (𝑅2>0.98, 𝑅2 𝑎𝑑𝑗>0.98, and E0.99), while the recovery phase was represented by an empirical model. The percentage of recovery (%R) of the ICM significantly decreased with the protein content.

Turkish Journal of Biology

Arabinofuranosyl cytidine (AraC) is an analog of deoxycytidine used as an anticancer drug for leukemic patients. The effective dose always produces severe complications. The present study investigated the modulation of autophagy and its impact on the cytotoxicity of AraC toward murine myelogenous leukemia cells (M-NFS-60). Autophagy was inhibited by NH4Cl or Bafilomycin A1 or enhanced by amino acid starvation, glucose starvation, mild hyperthermia (41 °C), or rapamycin (Rap). Cells were treated with different concentrations, 0 to 2 µM, of AraC in the presence or absence of autophagy modulators. AraC-induced apoptosis is combined with autophagy, especially at lower concentrations. This autophagy …

Turkish Journal of Biology

Gemcitabine is a broad-spectrum antimetabolite and a deoxycytidine analog recognized as a standard therapy alone or in combination with other antineoplastic agents in the therapy of pancreas cancer. Drug resistance following gemcitabine treatment is a common phenomenon; therefore, combinational therapy models are usually preferred. Pancreatic ductal adenocarcinoma, or pancreas cancer, is the fourth leading cause of cancer-related deaths worldwide. With the increasing incidence of pancreatic cancer every year, the mortality rate is also rising significantly because of late diagnosis, and limited chemotherapy options. Adjuvant chemotherapy after surgical resection is the typical option for the treatment of early pancreatic cancer. Mostly, …

Whilst modern medicine has increased longevity, the rise in life expectancy has brought about new struggles, namely that of aging and age related disease. Thus, humanity has been presented with a new problem: at what point is death preferable to aggressive life-prolonging treatment in the face of inevitable death? And if so, what right do individuals have to control the circumstances of their death? In the West, traditionally, individuals who seek to end their own lives in the face of terminal illness opt for forgoing artificial hydration and nutrition. Driven by an increase in desire for autonomous dignified death, a …

Effects Of Drought On Habitat Quality For Native Bees In Residential Gardens Of Claremont, Ca, Max Proctor Jan 2022

Effects Of Drought On Habitat Quality For Native Bees In Residential Gardens Of Claremont, Ca, Max Proctor
