The Tuckahoe Creek has been classified impaired by the Virginia Department of Environmental Quality (DEQ) indicating that one of the following parameters exceed the accepted limits: pH, temperature, dissolved oxygen (DO), phosphates, nitrates, and concentration of fecal coliforms. Throughout the course of this study, two selected sites have been monitored monthly in order to measure these parameters. Route 288 is located near a construction site, and Route 621 is near Sycamore Creek Golf Course in a more rural area. The results from this study have shown that the fecal coliforms have been uniformly within the set legal limit of 1,000 …

The six main objectives of this project were: 1) test for levels of West Nile Virus in mosquitoes collected at a Fulton County Recreational Facility, 2) assay various mosquito oviposition attractants on West Nile virus positive female mosquitoes, 3) test effects of distance between mosquito infusion attractants, 4) determine effects of study site location, treatment, and competition distances on mosquito collections, 5) test for any correlation between environmental factors and mosquito abundance, and 6) measure levels of enzymatic insecticide resistance present in species of female mosquitoes that tested positive for the presence of West Nile Virus. We present accounts for 40 species of mammals collected from 15 localities in the Mongolian People's Republic. Accounts include taxonomic, morphometric, reproductive and ecological information, as well as trap effort and success. In addition, we include a brief history of mammalogical work within Mongolia, a taxonomically updated species list for the country, and a list of institutions with holdings of Mongolian mammals.

Objective: To determine the prevalence and site of isolation of different serotypes of group B streptococcus (GBS) colonization or infection at a small community hospital.
Methods: GBS isolates were obtained from a small community hospital and were then serotyped as la, Ib, II, III, IV, V or non-typeable. Hospital records were reviewed for patient sex, age and pregnancy status as well as the site of GBS isolation.
Results: GBS serotypes Ia, III and V were most common and accounted for over 60% of the total number of isolates. Serotype Ia was most prevalent in reproductive-age females, while serotypes V and …

Gompertz and Weibull functions imply contrasting biological causes of demographic aging. The terms describing increasing mortality with age are multiplicative and additive, respectively, which could result from an increase in the vulnerability of individuals to extrinsic causes in the Gompertz model and the predominance of intrinsic causes at older ages in the Weibull model. Experiments that manipulate extrinsic mortality can distinguish these biological models. To facilitate analyses of experimental data, we defined a single index for the rate of aging (ω) for the Weibull and Gompertz functions. Despite a growing need, oceanographers are limited by existing technological constrains and are unable to observe aquatic microbes in their natural setting. In order to provide a simple and easy to implement solution for such studies, a new Thin Light Sheet Microscope (TLSM) has been developed. The TLSM utilizes a well-defined sheet of laser light, which has a narrow (23 micron) axial dimension over a 1 mm x 1 mm field of view. This light sheet is positioned precisely within the depth of field of the microscope’s objective lens. The technique thus utilizes conventional microscope optics but replaces the illumination …

Urinary tract infections are one of the most common causes of ambulatory care visits, physician-office visits and hospitalization each year in the US. Sexual activity primarily contributes to acquisition of UTI in women; nearly half of adult women have had a UTI at least once. The most common causative agent is E. coli, followed by Enterobacter spp., Klebsiella spp., Pseudomonas aeruginosa, Proteus mirabilis, other Gram-negative rods and Group B streptococci, depending on geographical location. 1 Until the late 1990's, trimethoprim-sulfamethoxazole (TMP-SMX) was the most commonly prescribed and effective treatment option, but the rampant development of resistance to this drug has …

We assessed the status of furbearing mammals on Fort A.P. Hill, Caroline County, Virginia during the 1998-1999 trapping season with the cooperation of local licensed trappers. Our analyses were based on 345 captures representing of 10 mammal species, ranging from one bobcat (Lynx rufus) to 157 beavers (Castor canadensis). Mean number of captures per 100 trap nights was 17.0. Captures varied from 11.9 to 17.9 per 100 trap nights for conibear traps and 9.7 to 18.3 per 100 trap nights for leg-hold traps. Fish losses from infectious diseases are a significant problem in aquaculture worldwide. However, peptide antibiotics show potent activity against a broad range of pathogens including fish pathogens. Therefore, the ability of antimicrobial peptides to protect fish against infections caused by fish pathogens clearly shows the potential for utilisation of antimicrobial peptides for the treatment of fish diseases. The strategy of overexpressing the antimicrobial peptide genes in transgenic fish may provide a method of decreasing fish disease problems in aquaculture.

We evaluated an artificial capillary feeding method to infect nymphal Ixodes scapularis (Say) ticks with Borrelia burgdorferi, the causative agent of Lyme disease. Thirty to 70% of the nymphs were infected after feeding for 2.5 h from glass capillary tubes filled with a solution of spirochetes. Capillary infection was stable and persisted in the nymphs for at least 10 d after feeding. Capillary feeding also maintained natural vector competence patterns because I. scapularis ticks acquired infection unlike Dermacentor variablis (Say), which did not become infected. Capillary infected I. scapularis nymphs were capable of transmitting the infection to naive mice …

The intracellular parasite Trichinella is a genus in phylum Nematoda that contains six named species including Trichinella spiralis and Trichinella pseudospiralis. These parasites infect a large variety of wild and domestic animals, human beings and a few species of birds. The parasitic strategies and the pathological effects on the host between trichinella spiralis and Trichinella pseudospiralis are quite different.

We sought to test the hypothesis that bacterial species richness and composition vary at the millimeter scale in the marine pelagic environment, in response to the heterogeneous distribution of organic matter. To test this hypothesis, it was necessary to design and test a protocol to sample, lyse and analyze (by polymerase chain reaction/denaturing gradient gel electrophoresis [PCR-DGGE]) bacterioplankton species richness in a single microliter of seawater. Significant variation in bacterial species richness was found amongst 1 μl samples. Greater species richness was seen when seawater was enriched with diatom detritus. Antimicrobial susceptibility profiles were assembled for isolates of Moraxella catarrhalis collected from the Mountain Home Veteran's Affairs Medical Center (VAMC) clinical laboratory in Johnson City, Tennessee. The goal of the study was to identify isolates for genetic characterization using comparisons of susceptibility profiles. Isolates of Moraxella catarrhalis collected from July 1984 through 1994 were analyzed for β-lactamase production using a Cefinase disk assay.
A multi-drug profile consisting of 11 β-lactam antibiotics was performed on the 41 M. catarrhalis isolates. Kirby Bauer disk assays were performed for 7 cephalosporin and 4 non-cephalosporin antibiotics.

We previously described a bacteriophage of the Lyme disease agent Borrelia burgdorferi designated phi BB-1. This phage packages the host complement of the 32-kb circular plasmids (cp32s), a group of homologous molecules found throughout the genus Borrelia. To demonstrate the ability of phi BB-1 to package and transduce DNA, a kanamycin resistance cassette was inserted into a cloned fragment of phage DNA, and the resulting construct was transformed into B. burgdorferi CA-11.2A cells. The kan cassette recombined into a resident cp32 and was stably maintained. The cp32 containing the kan cassette was packaged by phi BB-1 released from this B. …

We investigated how constant temperatures of 22, 24, and 26 degreesC experienced across the full life cycle affected the dynamics of caged populations of Aedes albopictus (Skuse). All cages were equipped with plastic beakers that served as sites for oviposition and larval development. We measured the per capita daily mortality and emergence rates of the adults and size of adult females, and estimated the intrinsic rate of increase (r) and asymptotic density (K) for each adult population. populations at 26 degreesC had greater intrinsic rates of increase and lower asymptotic densities than populations at 22 and 24 degreesC. The Mediterranean fruit fly (Ceratitis capitata) is an invasive agricultural pest with a wide host range and a nearly global distribution. Efforts to forgo the medfly''s spread into the United States are dependent on an understanding of population dynamics in newly established populations elsewhere. To explore the potential influence of demographic and historical parameters in six medfly populations distributed from Mexico to Peru, we created population genetic null models using Monte Carlo simulations. In an effort to identify the sources of the viruses that emerge after discontinuation of therapy, analyses of human immunodeficiency virus (HIV) quasi species were done for 3 patients with sustained levels of HIV RNA of <50 copies/mL for 1-3 years. The sequences found in the rebounding plasma virus were closely related to those of the actively replicating form of viruses present before the initiation of combination therapy. All quasi species found in the rebounding plasma virus were also present in proviral DNA, cell-associated RNA in peripheral blood mononuclear cells (PBMC), and virion RNA derived from PBMC coculture during periods when plasma HIV RNA levels were <50 copies/mL. These findings suggest that the rapid resurgence of plasma viremia observed after discontinuation of therapy and the viruses cocultured from PBMC are derived from a relatively stable pool of the replicating form of virus rather than from activation of a previously latent pool.

The blacklegged tick, Ixodes scapularis Say, transmits the Lyme disease spirochete (Borrelia burgdorferi), whereas the American dog tick, Dermacentor variabilis (Say), is unable to transmit the bacterium. We compared the innate immune response of these ticks against spirochetes directly inoculated into the hemocoel cavity of ticks. In I. scapularis, some Borrelia were found associated with hemocytes, while numerous other spiral-shaped, intact bacteria remained free in the hemolymph. In contrast, in D. variabilis only remnants of the bacteria were evident in the hemolymph, indicating lysis; intact spirochetes were rare. Spirochetes were observed bound to or within the organs …

In the past few decades there has been growing evidence that species of Agrobacterium can be opportunistic animal pathogens. One of these species is Agrobacterium tumefaciens, which for years has been studied as a plant pathogen. The results of one published experiment suggested that an Agrobacterium toxin could kill mice (12). Other strong evidence for Agrobacterium as an opportunistic human pathogen is the over fifty published clinical cases in which humans were infected by Agrobacterium species (3, 13-25). Human granulocytic ehrlichiosis (HGE) is a newly emerging, tick-borne zoonosis in North America. The agent that causes HGE is very similar to Ehrlichia phagocytophilia and Ehrlichia equi, but has yet to be classified as a separate species.
This study was carried out in order to determine the prevalence rate of HGE infection in blacklegged ticks (Ixodes scapularis) in insular and mainland sites in southeastern Georgia using nested PCR techniques. Identification Of A Defensin From The Hemolymph Of The American Dog Tick, Dermacentor Variabilis, Robert Johns, Daniel E. Sonenshine, Wayne L. Hynes

Hemolymph from partially fed virgin Dermacentor variabilis females was collected following Borrelia burgdorferi challenge and assayed for antimicrobial activity against Bacillus subtilis and B. burgdorferi. A small inducible cationic peptide was identified by SDS-PAGE in the hemolymph of these ticks as early as 1h post challenge. Following purification by a three-step procedure involving sequential SepPak elution, reversed phase high performance liquid chromatography (RP-HPLC) and gel electrophoresis, the yield of the active peptide was approximately 0.1% of the total protein in the hemolymph plasma. Bartonella quintana, the agent of trench fever and a cause of endocarditis and bacillary angiomatosis in humans, has the highest reported in vitro hemin requirement for any bacterium. We determined that eight membrane-associated proteins from B. quintana bind hemin and that a similar to 25-kDa protein (HbpA) was the dominant hemin binding protein. Like many outer membrane proteins, HbpA partitions to the detergent phase of a Triton X-114 extract of the cell and is heat modifiable, displaying an apparent molecular mass shift from approximately 25 to 30 kDa when solubilized at 100 degreesC. The nuclear factor-КB (NF-КB)/Rel family of inducible dimeric transcription factors regulates numerous genes associated with the immune, mitogenic, and apoptotic responses. Investigations were made into the role of NF-КB in breast carcinoma (MCF-7, T47D, MDA-MB-231) and glioblastoma (U87MG, U373MG, Ul 18MG) cell lines. NF-КB binding to DNA was quantified using both gel mobility shift assays and transient transfections of a vector containing a luciferase gene under the control of 4 tandem copies of the NF-КB binding sequence. Bartonella bacilliformis and Bartonella henselae, the respective agents of Oroya fever and cat-scratch disease in humans, are known to produce bacteriophage-like particles (BLPs) that package 14 kbp segments of the host chromosome. Data from this study suggest that other Bartonella species including Bartonella quintana, Bartonella doshiae and Bartonella grahamii also contain similar BLPs, as evidenced by the presence of a 14 kbp extrachromosomal DNA element in their genomes, whereas Bartonella elizabethae and Bartonella clarridgeiae do not. A purification scheme utilizing chloroform, DNase I and centrifugation was devised to isolate BLPs from B. bacilliformis. Intact BLPs were observed by transmission electron …
