Functional Influence Of 14-3-3 (Ywha) Proteins In Mammals, Elizabeth Barley, Santanu De
Mako: NSU Undergraduate Student Journal
The 14-3-3 (YWHA) proteins are homologous, ubiquitous, and conserved in most organisms ranging from plants to animals and play important roles in regulating key cellular events such as cell signaling, development, apoptosis, etc. These proteins consist of seven isoforms in mammals, termed under Greek alphabetization: beta (β), gamma (γ), epsilon (ε), eta (η), tau/theta (τ), sigma (σ), and zeta (ζ). Each of these isoforms can interact with a plethora of binding partners and has been shown to serve a distinct role in molecular crosstalk, biological processes, and disease susceptibility. Protein 14-3-3 isoforms are scaffolding proteins capable of forming homodimers and …

Environmental DNA (eDNA) metabarcoding has demonstrated its applicability as a highly sensitive biomonitoring tool across small spatial and temporal scales in marine ecosystems. However, it has rarely been tested across large spatial scales or biogeographical barriers. Here, we scale up marine eDNA metabarcoding, test its ability to detect a major marine biogeographic break and evaluate its use as a regional biomonitoring tool in Australia.

Bison from Yellowstone National Park (YNP) have an important genetic history. As one of the few wild herds of bison with no evidence of cattle DNA introgression and a large enough population to maintain genetic diversity, they are considered a conservation priority for the species. Unfortunately, there is a high prevalence of the zoonotic disease brucellosis in the herd. Part of the management strategy for controlling the disease and herd size in YNP is to remove bison from the population during the winter migration out of the park. This interagency management cull provides an opportunity to collect a large number …

The Brown Vine Snake, Oxybelis aeneus, was until recently considered a single species, distributed from southern Arizona through the Neotropics into southeastern Brazil. However, newly conducted research restructured the species with a substantial taxonomic revision, recognizing five additional taxa (i.e. O. koehleri, O. microphthalmus, O. potosiensis, O. rutherfordi, O. vittatus) in this species complex. This revision focused on populations in North America, Central America, and northern South America while neglecting the southern portion of its distribution. Here, we examine the taxonomic history of the complex and use it along with specimen data to resurrect O. acuminatus from southeastern Brazil. Finally, …

We investigated the landscape epidemiology of a globally distributed mammal, the wild pig (Sus scrofa), in Florida (U.S.), where it is considered an invasive species and reservoir to pathogens that impact the health of people, domestic animals, and wildlife. Specifically, we tested the hypothesis that two commonly cited factors in disease transmission, connectivity among populations and abundant resources, would increase the likelihood of exposure to both pseudorabies virus (PrV) and Brucella spp. (bacterial agent of brucellosis) in wild pigs across the Kissimmee Valley of Florida. Using DNA from 348 wild pigs and sera from 320 individuals at 24 sites, we …

Prenatal And Postnatal Nutrition Influence Pancreatic And Intestinal Carbohydrase Activities Of Ruminants, Ronald J. Trotta, Kendall C. Swanson
Animal and Food Sciences Graduate Research
In ruminant livestock species, nutrition can play an important role in the long-term programming of gastrointestinal function. Pancreatic and small intestinal digestive enzymes are important for postruminal digestion of carbohydrates and protein. Carbohydrases have been shown to respond to changes in the level of feed intake and the dietary inclusion of specific nutrients, including arginine, butyrate, folic acid, fructose, and leucine. Understanding how diet influences enzyme development and activity during prenatal and postnatal life could lead to the development of dietary strategies to optimize offspring growth and development to increase digestive efficiency of ruminant livestock species. More research is needed …

Single-Locus Species Delimitation And Ecological Niche Modeling Provide Insights Into The Evolution, Historical Distribution, And Taxonomy Of The Pacific Chorus Frogs, Robert C. Jadin, Sarah A. Orlofske, Tereza Jezkova, Christopher Blair
Publications and Research
The Pacific chorus frogs are a complex of three wide-ranging species (i.e. Hyliola hypochondriaca, Hyliola regilla, Hyliola sierra) whose current taxonomy remains unresolved. We conducted species delimitation analyses of these taxa using fragments of the cytochrome b and 12S–16S mtDNA genes to assess the species diversity. Importantly, we included samples from new locations throughout the range to better understand species distributions and identify potential contact zones among clades. Our analyses revealed three slightly parapatric but distinct species-level clades. Molecular dating revealed that these species began diverging in the Pleistocene c. 1.4 Mya with H. hypochondriaca and …

Accuracy Of Gebv Of Sires Based On Pooled Allele Frequency Of Their Progeny, Napoleón Vargas Jurado, Larry A. Kuehn, John W. Keele, Ronald M. Lewis
Roman L. Hruska U.S. Meat Animal Research Center: Reports
Despite decreasing genotyping costs, in some cases individually genotyping animals is not economically feasible (e.g., in small ruminants). An alternative is to pool DNA, using the pooled allele frequency (PAF) to garner information on performance. Still, the use of PAF for prediction (estimation of genomic breeding values; GEBVs) has been limited. Two potential sources of error on accuracy of GEBV of sires, obtained from PAF of their progeny themselves lacking pedigree information, were tested: (i) pool construction error (unequal contribution of DNA from animals in pools), and (ii) technical error (variability when reading the array). Pooling design (random, extremes, K-means), …

When the number of horn flies that blood feed on cattle exceeds the economic threshold, they can adversely affect the health and wellbeing of their hosts. Excessive horn fly burdens also lead to reduced weight gain and, consequently, diminished profits for livestock producers. Effective management and treatment require reliable surveillance methods for estimating the degree of horn fly burden (i.e., counting the number of flies on cattle). Traditionally, these estimates are obtained through human visual estimation, either in-person or by counting images on a photo; however, these methods are costly both in terms of time and labor and are prone …

The scorpion venom consisting of a pair of gland connected to telson has great importance for their survival, assisting in feeding and self-defense. Scorpion venoms are a complex mixture of inorganic salts, free amino acids, heterocyclic components, peptides and proteins. The present study aimed to identify telson components of four scorpion species Leiurus quinquestriatus, Androctonus amoreuxi, Orthochirus innesi and Buthacus leptochelys. Five telsons separated from four species of scorpions were preserved in 100% ethanol, homogenized in acetonitrile, centrifuged and sonicated as preparation of gas chromatography mass spectrometry (GC-MS) analysis. The present study indicated variations in the number and types of …

Sheep is one of the ungulates and critical for animal husbandry development like on grassland, whereas there are few pieces of research about their heat responses to date. In the present study, we selected two sheep breeds consisting of Mongolian sheep with evolving cold resistibility and Dorper sheep specifically bred for heat tolerance to explore how the different breeds of sheep positively respond to heat conditions. Three Mongolian sheep and 3 Dorper sheep underwent 37 $^{o}$C stress (2 h per day) during 4 weeks. The energy metabolism-associated adipose tissues of the experimental subjects were sampled, prepared and performed to RNA-seq …

The effects of an aminoglycoside antibiotic, streptomycin, on survival rate, development time, and adult longevity of Drosophila melanogaster (Diptera: Drosophilidae) were examined by adding on the artificial diet at different concentrations in laboratory condition. The effect of streptomycin on malondialdehyde (MDA) and protein carbonyl (PCO) contents, superoxide dismutase (SOD), and catalase (CAT) activities in all developmental stages of D. melanogaster was also investigated. First-instar larvae were reared into diets with 600 mg/L, 1200 mg/L, and 1800 mg/L of streptomycin concentrations and were fed until the adult emergence. The highest concentration of streptomycin significantly reduced the survival rates of all development …

This study was designed to record some morphometric and haematological parameters of common snipe, Gallinago gallinago. For this purpose, 20 individuals (10 male, 10 female) were collected from the wetlands of District Bahawalnagar, Punjab, Pakistan. Significant differences between the sexes were recorded in tail length, length of longest primary feather, and chest circumference. . No significant difference was recorded in haematological parameters between the sexes. Gut analysis revealed that common snipe feed mostly on Chironomidae and Ceratopogonidae larvae and male and female snipes have the same food preference.
