Genome targeting with CRISPR/Cas9 is a popular method for introducing mutations and creating knock-out effects. However, limited information is currently available on the mutagenesis of essential genes. This study investigated the efficiency of CRISPR/Cas9 in targeting rice essential genes: the singleton TARGET OF RAPAMYCIN (OsTOR) and the three paralogs of the Sucrose non-fermenting-1 (SNF1)-related kinase 1 (OsSnRK1α), OsSnRK1αA, OsSnRK1αB and OsSnRK1αC. Strong activity of constitutively expressed CRISPR/Cas9 was effective in creating mutations in OsTOR and OsSnRK1α genes, but inducible CRISPR/Cas9 failed to generate detectable mutations. Resistance to protoporphyrinogen IX oxidase (PPO)-inhibitors in Amaranthus palmeri and Amaranthus tuberculatus is mainly contributed by mutations in the PPO enzyme, which renders herbicide molecules ineffective. The deletion of glycine210 (ΔG210) is the most predominant PPO mutation. ΔG210-ppo2 is overexpressed in rice (Oryza sativa c. ‘Nipponbare’) and Arabidopsis thaliana (Col-0). A foliar assay was conducted on transgenic T1 rice plants with 2× dose of fomesafen (780 g ha−1), showing less injury than the non-transgenic (WT) plants. Climate change related drought is projected to harm maize production. Water use strategies can help mitigate the impact of drought on crop yield. However, little is known about maize metabolic response to drought at different developmental stages. To shed light on this, drought conditions were applied to maize at the six-leaf stage (V6), twelve leaf stage (V12), and tassel stage (VT). V6 and VT took eight days to achieve a low stomatal conductance threshold, but V12 took 16 days. Differential gene expression analysis of the transcriptome indicates that V6 showed the most response with 53 impacted metabolic pathways, many of …

Nitrogen (N) is a crucial nutrient for proper plant growth and development. N deficiency results in poor tillering and thin, short internode stem production for sugarcane production, thus resulting in low cane tonnage. Therefore, proper N fertilizer management is essential for a sustainable sugarcane production system. This series of experiments is designed to understand the development and chemistry of sickle gloss. Sickles are common in the archaeological record and have long been studied for their eponymous “sickle gloss”. There is debate as to how this gloss is formed. Five experimental sickles containing flint and novaculite blades were used to harvest high and low moisture content rye and common fescue and associated field weeds. The differences in the development of sickle gloss were examined. High moisture content plants create thick, smooth fields of polish with undulating, billowing margins at a higher developmental speed. Low moisture content plants …

Cost-effective weed suppression is an important consideration for tomato growers. Growers often choose methods which minimize hand labor, as hand weeding can be prohibitively expensive. This project determined economic viability of high tunnel tomatoes treated with several methods of weed control, both organic and chemical. These methods included: 2-week hand weeding, 1-week hand weeding, preemergent, straw, landscape fabric, and untreated weedy control plots. These treatments were applied to randomized blocks in a high-tunnel. Weeding, planting, and harvest were all timed to determine labor and material costs of weed management strategy implementation. After harvest, marketable yield was weighed to determine revenue. …

Rice is utilized as an adjunct grain by the beer manufacturing industry. Before utilization, the rice has to be dried to a desired moisture content. The drying process employed may have an impact on the rice’s physicochemical properties which influence the rice’s performance in the brewing process. This study focused on investigating the impact of microwave drying on rice’s physicochemical properties and utilization in the brewing process. The introduction of dicamba-tolerant (DT) soybeans (Glycine max L. Merr) and cotton (Gossypium hirsutum L) in 2017 provided an additional tool for herbicide resistant weeds management. In the subsequent years, off-target movement of dicamba allegedly caused damage to sensitive crops and vegetation.
Possible causes of off-target movement include tank contamination, physical drift, and volatility. Additional products, such as herbicides to control grass, are often added to tank with dicamba, which is used to control broadleaf weeds, to increase the spectrum of control and application efficiency. Sweetpotato (Ipomoea batatas L.) is a low-technology, subsistence crop that enhances food and nutrition security particularly in developing nations. Weed interference with the crop can reduce sweetpotato storage root yields and product quality. Current weed management practices in sweetpotato include PRE or POST herbicides application, cultivation, mowing, or handweeding. Cover cropping can improve soil health, increase plant-available nitrogen (N), provide surface residue to prevent erosional soil loss, increase water infiltration, and increase weed suppression. The use of warm-season putting greens in the transition zone has increased in recent years. Ultradwarf bermudagrass (Cynodon dactylon (L.) Pers x C. transvaalensis Burtt-Davy) is the most prevalent warm-season putting green selection in the transition zone, however, newly developed greens-type zoysiagrass (Zoysia spp.) cultivars represent another potential selection for golf courses. The two major limitations of warm-season grasses in the transition zone are a general lack of cold- and shade-tolerance. Reducing tillage in cropping systems causes weed management to be dependent on chemical and cultural methods for weed control. Over time, herbicide-resistant weeds have developed due to the continuous selection pressures from herbicides, particularly in the Midwest Corn Belt. The implementation and future success of hybrid wheat (Triticum aestivum L.) is impacted by breeders’ inability to create consistent high yielding, high heterosis hybrids. This research addresses this problem by conducting an exploration of transcriptomes from hybrids and parent lines to determine what genes are active in heterotic or non-heterotic hybrids and how their level of expression can explain the phenotype of grain yield heterosis. Thlaspi arvense (field pennycress) is being domesticated as a winter annual oilseed crop capable of improving ecosystems and intensifying agricultural productivity without increasing land use. It is a selfing diploid with a short life cycle and is amenable to genetic manipulations, making it an accessible field-based model species for genetics and epigenetics. Soybean production in the U.S. Mid-South relies heavily on irrigation with 85% of soybean surfaces irrigated in Arkansas. Reduction in irrigation due to water quantity restrictions will significantly affect soybean seed yield, making variety selection increasingly important. The objective of the study was to assess if irrigation onsets at different reproductive stages affect wilting, seed yield, and key agronomic traits on determinate maturity group 5 (MG 5) soybean. The Amazon basin includes 550 M ha covered with rainforests, with 60% of this area being in Brazil. Conversion of rainforest for soybean production raises concerns about the degree to which Brazil can reconcile production and environmental goals. Here we investigated the degree to which intensification could help Brazil produce more soybean without further encroachment of the Amazon Forest. Our analysis shows that continuation of current trends in soybean yield and area would lead to conversion of additional 5.7 M ha of forests and savannas during the next 15 years, with an associated 2550 Mt of CO2eq released into the …

Limited knowledge about how nitrogen (N) dynamics are affected by climate change, weather variability, and crop management is a major barrier to improving the productivity and environmental performance of soybean-based cropping systems. To fill this knowledge gap, we created a systems understanding of agroecosystem N dynamics and quantified the impact of controllable (management) and uncontrollable (weather, climate) factors on N fluxes and soybean yields. Kangkung (Ipomoea reptans Poir) adalah sayuran potensial untuk dikembangkan pada sistem akuaponik. Kelemahan akuaponik adalah nutrisi yang hanya bertumpu pada sisa pakan dan kotoran ikan, sehingga tanaman memerlukan pasokan hara dari sumber lain. Pemberian pupuk foliar merupakan salah satu upaya yang dilakukan untuk meningkatkan hasil kangkung pada sistem akuaponik. Penelitian ini bertujuan untuk mengetahui pengaruh jenis pupuk foliar dan padat tebar lele terhadap hasil kangkung pada sistem akuaponik menggunakan budidaya ikan dalam ember (budikdamber). Ubi jalar memiliki keragaman genetik yang luas sehingga potensi genetiknya dapat dikembangkan lebih lanjut. Ubi jalar berdaging putih memiliki kandungan pati paling tinggi dibandingkan dengan warna yang lain. Tujuan dari penelitian ini adalah untuk mengetahui variasi karakter hasil dan komponen hasil klon ubi jalar berdaging putih. Percobaan dilakukan di tiga lokasi sentra produksi ubi jalar yakni Kabupaten Sumedang, Kabupaten Garut, dan Kabupaten Majalengka, Jawa Barat mulai bulan Desember 2019 hingga Desember 2020. Verification of clonal identity of hop (Humulus lupulus L.) cultivars within breeding programs and germplasm collections is vital to conserving genetic resources. Accurate and economic DNA-based tools are needed in dioecious hop to confirm identity and parentage, neither of which can be reliably determined from morphological observations. In this study, we developed two fingerprinting sets for hop: a 9-SSR fingerprinting set containing high-core repeats that can be run in a single PCR reaction and a kompetitive allele specific PCR (KASP) assay of 25 single nucleotide polymorphisms (SNPs). Soybean, Glycine max (L.) Merr, is considered the most economically important oil and feed crop in the US and is grown on nearly 35,000,000 ha on an annual basis. Factors that limit yield include weed pressure, disease infection, and of particular interest to entomologists, insect herbivory. Soybean is targeted by a range of above and below ground pests which are primarily controlled by insecticides. Neonicotinoids are a recently popularized insecticide class that when applied as a seed treatment, can provide protection from early season pests.
