Maize is highly sensitive to short term flooding and submergence. Early season flooding reduces germination, survival and growth rate of maize seedlings. We aimed to discover genetic variation for submergence tolerance in maize and elucidate the genetic basis of submergence tolerance through transcriptional profiling and linkage analysis of contrasting genotypes. A diverse set of maize nested association mapping (NAM) founder lines were screened, and two highly tolerant (Mo18Wand M162W) and sensitive (B97 and B73) genotypes were identified. Tolerant lines exhibited delayed senescence and lower oxidative stress levels compared to sensitive lines. Transcriptome analysis was performed on these inbreds to provide …

Soybean, rather than nitrogen-containing forages, is the primary source of quality protein in feed formulations for domestic swine, poultry, and dairy industries. As a sole dietary source of protein, soybean is deficient in the amino acids lysine (Lys), threonine (Thr), methionine (Met), and cysteine (Cys). Increasing these amino acids would benefit the feed industry. The objective of the present study was to identify quantitative trait loci (QTL) associated with crude protein (cp) and amino acids in the ‘Benning’ × ‘Danbaekkong’ population. The population was grown in five southern USA environments. Amino acid concentrations as a fraction of cp (Lys/cp, Thr/cp, …

Using A Simple Leaf Color Chart To Estimate Leaf And Canopy Chlorophyll A Content In Maize (Zea Mays), Anthony Nguy-Robertson, Yi Peng, Timothy Arkebauer, David Scoby, James Schepers, Anatoly Gitelson
Department of Agronomy and Horticulture: Faculty Publications
This study utilized a leaf color chart (LCC) to characterize the variation in leaf chlorophyll and estimate canopy chlorophyll in maize (Zea mays). The LCC consisted of four levels of greenness and was used to sort maize leaves in 2011 for three fields near Mead, Nebraska, USA. Leaf chlorophyll content for each color chart class was determined using two leaf-level sensors. The variation within each LCC class was reasonable (CV < 56%). The darkest color class predominated and indicated adequate fertilization rates using a SPAD. Canopy chlorophyll content was estimated using destructively measured leaf area index (LAI) and the LCC. This approach was verified with a method utilizing canopy reflectance collected by both satellite imagery and a four-band radiometer. The error between the two methods was reasonable (RMSE = 0.55-0.88 g m-2; CV = 25.6-50.4%), indicating that both leaf and canopy chlorophyll can be estimated cheaply without a wet lab or field-based sensors.

Proteomic Profiling Of Maize Opaque Endosperm Mutants Reveals Selective Accumulation Of Lysine-Enriched Proteins, Kyla J. Morton, Shangang Jia, Chi Zhang, David R. Holding
Department of Agronomy and Horticulture: Faculty Publications
Reduced prolamin (zein) accumulation and defective endoplasmic reticulum (ER) body formation occurs in maize opaque endosperm mutants opaque2 (o2), floury2 (fl2), defective endosperm*B30 (DeB30), and Mucronate (Mc), whereas other opaque mutants such as opaque1 (o1) and floury1 (fl1) are normal in these regards. This suggests that other factors contribute to kernel texture. A liquid chromatography approach coupled with tandem mass spectrometry (LC-MS/MS) proteomics was used to compare non-zein proteins of nearly isogenic opaque endosperm mutants. In total, 2762 proteins were identified that were enriched for biological processes such as protein transport and folding, amino acid biosynthesis, …

Tonoplast Sugar Transporters (Sbtsts) Putatively Control Sucrose Accumulation In Sweet Sorghum Stems, Saadia Bihmidine, Benjamin T. Julius, Ismail M. Dweikat, David M. Braun
Department of Agronomy and Horticulture: Faculty Publications
Carbohydrates are differentially partitioned in sweet versus grain sorghums. While the latter preferentially accumulate starch in the grain, the former primarily store large amounts of sucrose in the stem. Previous work determined that neither sucrose metabolizing enzymes nor changes in Sucrose transporter (SUT) gene expression accounted for the carbohydrate partitioning differences. Recently, 2 additional classes of sucrose transport proteins, Tonoplast Sugar Transporters (TSTs) and SWEETs, were identified; thus, we examined whether their expression tracked sucrose accumulation in sweet sorghum stems. We determined 2 TSTs were differentially expressed in sweet vs. grain sorghum stems, likely underlying the massive difference in sucrose …

Selection For Silage Yield And Composition Did Not Affect Genomic Diversity Within The Wisconsin Quality Synthetic Maize Population, Aaron Lorenz, Timothy M. Beissinger, Renato Rodrigues Silva, Natalia De Leon
Department of Agronomy and Horticulture: Faculty Publications
Maize silage is forage of high quality and yield, and represents the second most important use of maize in the United States. The Wisconsin Quality Synthetic (WQS) maize population has undergone five cycles of recurrent selection for silage yield and composition, resulting in a genetically improved population. The application of high-density molecular markers allows breeders and geneticists to identify important loci through association analysis and selection mapping, as well as to monitor changes in the distribution of genetic diversity across the genome. The objectives of this study were to identify loci controlling variation for maize silage traits through association analysis …

Zinc deficiency in humans is a serious problem worldwide with an estimated one third of populations at risk for insufficient zinc in diet, which leads to impairment of cognitive abilities and immune system function. The goal of this research was to increase the bioavailable zinc in the edible portion of cassava roots to improve the overall zinc nutrition of populations that rely on cassava as a dietary staple. To increase zinc concentrations, two Arabidopsis thaliana genes coding for ZIP1 and MTP1 were overexpressed with a tuber-specific or constitutive promoter. Eighteen transgenic events from four constructs, out of a total of …

Urea Formulations On The Productivity Of Bermudagrass And Bermudagrass-White Clover Pastures, Caitlin Timberlake
Theses and Dissertations--Plant and Soil Sciences
Bermudagrass (Cynodon dactylon L.) is a perennial warm-season grass that is very responsive to nitrogen (N) fertilization. Excessive N applications have negative environmental consequences and make maintaining mixed swards difficult. This study determined the effects of enhanced efficiency (EE) N fertilizers and fertilizer rate on bermudagrass yields, nutritive values, and white clover persistence. Nitrogen sources included urea, urea formulated with Agrotain® (U+A), urea with Agrotain® and dicyandiamide (SuperU), a polymer-coated urea (ESN), ESN+urea (75% ESN, 25% urea), and methylene urea (MU). In the urea formulation trial, SuperU and U+A maximized forage yields at lower N rates. The …

First Year Biomass Production Following Application Of Various Size Strip Thinnings In A Young Hardwood Stand In West Virginia., Ashlee Martin
Graduate Theses, Dissertations, and Problem Reports (ETD)
Strip thinning can reduce stand density to promote better growth of residual trees and has potential to create a woody bioenergy feedstock in addition to enhancing and maintaining early successional habitat for wildlife. However, little research has been conducted on strip thinning within young hardwood stands to determine if these are viable attributes of strip thinning. The goal of this study was to assess the first year biomass production after strip thinning among three different cut strip widths (8 ft, 12 ft, and 16 ft) in a 22-year-old mixed mesophytic hardwood stand. Woody biomass production from stump sprouts and true …

Journal of Public Affairs and Development
In 2010, the Philippines passed a law (Republic Act No. 10068) to support the expansion of organic agriculture (OA) in the country, with a goal of 5 percent of total production area being devoted to this technology. Available data show that in 2012, only about 0.7 percent of the agricultural land area in the country was devoted to OA production. While several other administrative orders supported the law, there seems to be a need to understand the processes to operationalize the said law.
This paper analyzed the production, marketing, and consumption issues surrounding the organic vegetable industry to come up …

Estimating Desirable Cattle Traits Using Latent Class And Mixed Logit Models: A Choice Modeling Application To The U.S. Grass-Fed Beef Industry, Isaac Sitienei
LSU Doctoral Dissertations
This study examines the preferences for cattle traits using mixed logit and latent class models. Choice experiment data from a 2013 mail survey of 1,052 U.S. grass-fed beef producers were used. The findings indicate that grass-fed beef producers generally preferred lower-priced, heavy animals that were small-to-medium framed and easy to handle for grass-finishing. Black animals that were retained from their own calves were preferred. Relative to intact males, steers and heifers were preferred. Except for the estimated parameter for the weight attribute, the standard deviations for the temperament, body frame, source, color, gender, and price attribute levels were significant at …

Department of Agronomy and Horticulture: Faculty Publications
Seed oils have proved recalcitrant to modification for the production of industrially useful lipids. Here, we demonstrate the successful metabolic engineering and subsequent field production of an oilseed crop with the highest accumulation of unusual oil achieved so far in transgenic plants. Previously, expression of the Euonymus alatus diacylglycerol acetyltransferase (EaDAcT) gene in wild-type Arabidopsis seeds resulted in the accumulation of 45 mol% of unusual 3-acetyl-1,2- diacyl-sn-glycerols (acetyl-TAGs) in the seed oil (Durrett et al., 2010 PNAS 107:9464). Expression of EaDAcT in dgat1 mutants compromised in their ability to synthesize regular triacylglycerols increased acetyl-TAGs to 65 mol%. Camelina and soybean …

Proper Handling And Transportation Of Eggs For Sale At Kentucky Farmers’ Markets, Anthony J. Pescatore, Jacquie Jacob
Proper Handling And Transportation Of Eggs For Sale At Kentucky Farmers’ Markets, Anthony J. Pescatore, Jacquie Jacob
Agriculture and Natural Resources Publications
Regardless of the number of eggs produced, and whether the eggs are for home use or sale, careful egg handling is very important. The United States Food and Drug Administration (FDA) lists eggs as a potentially hazardous food, therefore careful handling is important. In Kentucky, if you sell more than 60 dozen eggs a week you need to purchase a retail license. The same holds true if you want to sell your eggs to a retail store to be re-sold or to a distributor that will sell them for you. If you produce fewer than 60 dozen eggs a week …

Studies To Characterize Heavy Metal Content And Migration From Recycled Polyethyleneterephthalate, Michael John-Ross Whitt
Master's Theses
Packaging Materials account for 31% of the world’s municipal solid waste. Agencies like the Environmental Protection Agency (EPA) and the Agency for Toxic Substances and Disease Registry (ATSDR) are pushing for the increased use of recycled thermoplastic materials. Polyethylene terephthalate (PET) is a commonly recycled thermoplastic which is used to package ready-to-eat fruits and vegetables. Most recycled polyethylene terephthalate (RPET) packaging materials contain heavy metal catalysts, the most common being antimony. The recent increased use of recycled plastic materials has been suspected as the source of increased human heavy metal exposure. In this study, cadmium, chromium, nickel, lead and antimony …
