Using aminoacyl-tRNA synthetase/suppressor tRNA pairs derived from Methanocaldococcus jannaschii, an Escherichia coli cell-free protein production system affords proteins with site-specifically incorporated unnatural amino acids (UAAs) in high yields through the use of optimized amber suppressor tRNACUA opt and optimization of reagent concentrations. The efficiency of the cell-free system allows the incorporation of trifluoromethyl-phenylalanine using a polyspecific synthetase evolved previously for p-cyanophenylalanine, and the incorporation of UAAs at two different sites of the same protein without any re-engineering of the E. coli cells used to make the cell-free extract.

"A novel bath lily-like graphene sheet-wrapped nano-Si composite synthesized via a simple spray drying process exhibits a high reversible capacity of 1525 mAh g(-1) and superior cycling stability, which could be attributed to a synergistic effect between highly conductive graphene sheets and active nanoparticles in the open nano/micro-structure."

We combine CO column measurements from the MOPITT, AIRS, SCIAMACHY, and TES satellite instruments in a full-year (May 2004–April 2005) global inversion of CO sources at 4◦ ×5◦ spatial resolution and monthly temporal resolution. The inversion uses the GEOS-Chem chemical transport model (CTM) and its adjoint applied to MOPITT, AIRS, and SCIAMACHY. Observations from TES, surface sites (NOAA/GMD), and aircraft (MOZAIC) are used for evaluation of the a posteriori solution. Using GEOSChem as a common intercomparison platform shows global consistency between the different satellite datasets and with the in situ data. The Precambrian shield of northern Greenland has been investigated by SHRIMP U-Pb zircon dating of 14 orthogneisses and granitoids plus 5 metasediments, integrated with mapping by the Geological Survey of Denmark and Greenland and whole-rock Nd isotopic studies. The Inglefield Mobile Belt is a tract of Palaeoproterozoic sedimentation, plutonism, polyphase deformation and high-grade metamorphism that underlies Inglefield Land and northern Prudhoe Land. In the southern part of the belt at 78 degrees 30'N, the E-W-trending Sunrise Pynt Straight Belt is a high-grade, but structurally late, shear zone with contrasts in the geology on either side. We have mapped U (238U) and Th (232Th) elemental concentrations as well as U-series isotope distributions in a Neanderthal tooth from the Middle Palaeolithic site of Payre using laser ablation ICP-MS. The U-concentrations in an enamel section varied between 1 and 1500 ppb. The U-concentration maps show that U-migration through the external enamel surface is minute, the bulk of the uranium having migrated internally via the dentine into the enamel. The uranium migration and uptake is critically dependent on the mineralogical structure of the enamel. Molecular dynamics (MD) simulations using empirical force fields are popular for the study of proteins. In this work, we compare anisotropic atomic fluctuations in nanosecond-timescale MD simulations with those observed in an ultra-high-resolution crystal structure of crambin. In order to make our comparisons, we have developed a compact graphical technique for assessing agreement between spatial atomic distributions determined by MD simulations and observed anisotropic temperature factors.

Reef development during past Interglacial periods, when sea level and sea surface temperatures were higher than today, provide unique insights into how reef systems may respond to projected human-induced global warming. Lord Howe Island currently represents the southernmost limit of reef development in the Pacific. Reef growth of Pleistocene age has been inferred to have occurred around the island, and this paper provides the fi rst detailed descriptions on the character of this development. Frequency Modulation AFM (FM-AFM) is commonly operated in ultra-high vacuum, though its inception in liquids for biological samples is relatively new. Here, we highlight the ability of FM-AFM to perform molecular resolution imaging of biomembrane surfaces and to detect individual layers of structured water at similar membrane interfaces. These studies highlight the potential of FM-AFM for studying model membranes and lipid raft systems on the molecular scale.

We have shown previously that a 'soluble' form of PrP (prion protein), not associated with membranous vesicles, exists in the male reproductive fluid [Ecroyd, Sarradin, Dacheux and Gatti (2004) Biol. Reprod. 71, 993-1001]. Attempts to purify this 'soluble' PrP indicated that it behaves like a high-molecular-mass complex of more than 350 kDa and always co-purified with the same set of proteins. The main associated proteins were sequenced by MS and were found to match to clusterin (apolipoprotein J), BPI (bacterial permeability-increasing protein), carboxylesterase-like urinary excreted protein (cauxin), beta-mannosidase and beta-galactosidase.
