Structural Insights Into The Dehydroascorbate Reductase Activity Of Human Omega-Class Glutathione Transferases, Huina Zhou, Joseph Brock, Dan Liu, Philip G. Board, Aaron J. Oakley

The reduction of dehydroascorbate (DHA) to ascorbic acid (AA) is a vital cellular function. The omega-class glutathione transferases (GSTs) catalyze several reductive reactions in cellular biochemistry, including DHA reduction. In humans, two isozymes (GSTO1-1 and GSTO2-2) with significant DHA reductase (DHAR) activity are found, sharing 64% sequence identity. While the activity of GSTO2-2 is higher, it is significantly more unstable in vitro. We report the first crystal structures of human GSTO2-2, stabilized through site-directed mutagenesis and determined at 1.9Å resolution in the presence and absence of glutathione (GSH). The Early Paleozoic Lachlan Fold Belt of eastern Australia is widely regarded as an ancient convergent plate margin beneath which paleo-Pacific (Panthalassic) oceanic lithosphere was continuously subducted. It is cited as the type example of a retreating accretionary orogeny. However, sandstone compositions, the sedimentological nature and timing of chert accumulation and overall stratigraphic architecture are not necessarily consistent with this model. We suggest an alternative explanation for growth of Gondwanan continental margin. Oceanic lithosphere outboard of the passive Gondwana continental margin was subducted beneath an extensive intra-oceanic island arc that now crops out as an allochthonous element (Macquarie arc) within …

The Ikkattoq gneisses of the Archaean gneiss complex in the Nuuk region, southern West Greenland, are the orthogneiss component within the amphibolite facies Tre Brodre terrane. They have mostly granodioritic compositions, with a small amount of quartz diorite. Sm-Nd isotopic data for a quartz diorite and five granodiorite Ikkattoq gneiss samples from within 5 km of the Ikkattoq (fjord) type locality yielded a regression with a slope equivalent to 2005 +/- 52 Ma (MSWD = 0.72). The sources and petrogenetic processes that generated some of the Earth's oldest continental crust have been more tightly constrained via an integrated, in situ (U-Pb, O and Hf) isotopic approach. The minerals analysed were representative zircon from four Eoarchaean TTG tonalites and two felsic volcanic rocks, and olivine from one harzburgite/dunite of the Itsaq Gneiss Complex (IGC), southern West Greenland. The samples were carefully chosen from localities with least migmatisation, metasomatism and strain. Zircon was thoroughly characterized prior to analysis using cathodoluminescence, scanning electron, reflected and transmitted light imaging.
