Barrows and gilts (n=210) were used to test the effects of crystalline amino acid (AA) supplementation of reduced crude protein (RCP) diets formulated on a net energy basis on quality characteristics of the LM from growing-finishing pigs. Pigs were blocked by weight, and pens (3 barrows and 3 gilts/pen) within each block were assigned randomly to either corn-soybean meal diets (C) or 1 of 3 RCP diets and added crystalline AA levels for the dietary treatments during each feeding phase. This thesis evaluates the PromegaTM P450-Glo assay (PromegaTM V9800) as a tool for quantifying ergot alkaloid concentration. Current techniques used for detection of ergot alkaloids are slow and expensive, do not detect all ergot alkaloids, or are not effective on bovine bodily fluids. The first study was conducted to determine effects of commercial ergot alkaloids (n = 6; 0 - 400 μM) on the PromegaTM P450-Glo assay. Cytochrome P450 (CYP450) activity in assay had a differential response to each ergot alkaloid and concentration. The potential of in situ rumen undegradable dry matter (RUDM), indigestible neutral-detergent fiber (INDF), indigestible acid-detergent fiber (IADF), acid-detergent insoluble ash (ADIA), alkaline-peroxide lignin (APL), and acid-detergent lignin (ADL) to predict digestibility (DMD) and fecal output (FO) by cattle fed bermudagrass [Cynodon dactylon (L.) Pers.] hay-diets categorized by their low (L), medium low (ML), medium high (MH), or high (H) CP concentrations (79, 111, 131, and 164 g/kg DM, respectively) was evaluated.
