Trace Dna Detection Using Diamond Dye: A Recovery Technique To Yield More Dna, Leah Davis
Trace Dna Detection Using Diamond Dye: A Recovery Technique To Yield More Dna, Leah Davis
Master's Theses
This study aspires to find a new screening approach to trace DNA recovery techniques to yield a higher quantity of trace DNA from larger items of evidence. It takes the path of visualizing trace DNA on items of evidence with potential DNA so analysts can swab a more localized area rather than attempting to recover trace DNA through the general swabbing technique currently used for trace DNA recovery. The first and second parts consisted of observing trace DNA interaction with Diamond Dye on porous and non-porous surfaces.
The third part involved applying the Diamond Dye solution by spraying it onto …
An Evaluation Of The Effect Of Degradation On Irisplex Snps, Maria Gruber
An Evaluation Of The Effect Of Degradation On Irisplex Snps, Maria Gruber
Master's Theses
The information from short tandem repeats (STRs), the current standard forensic DNA typing technology, does not always result in identification. In these cases, there are other informative markers in DNA that can be used to help generate investigative leads, one of which is single nucleotide polymorphisms (SNPs). One of the uses of SNPs is for phenotype determination. Among the phenotypic characteristics that can be predicted is eye color. A commonly used tool for eye color prediction is the free, online statistical prediction model IrisPlex, which includes 6 SNPs known to control eye color.
While SNPs can be useful, they, like …

Evaluation Of Raman Microspectroscopy And Hyperspectral Imaging As A Screening Tool To Determine Optimal Sampling Sites For Dna Recovery From Human Skeletal Remains, Kathleen M. Smith
Master's Theses
Numerous challenges exist with forensic genetic testing of human skeletal remains due to diagenesis patterns in bone microstructure, DNA degradation, and the presence of PCR inhibitors. Diagenesis is the microscopic breakdown of the bone matrix, which consists primarily of mineralized calcium hydroxyapatite and collagen. The process of diagenesis occurs in a heterogeneous, non-uniform manner along the diaphysis of a long bone, and determining the region with the most intact bone microstructure is not possible with the naked eye. Therefore, taking cuttings from the diaphysis for DNA testing is somewhat of a “blind” process, and decades of research and casework have …
Correlation Between Tooth Root Translucency, Crystallinity, And Dna Preservation In Teeth For Unidentified Human Remains (Uhr) Investigations, Paul Yount
Master's Theses
Endogenous genetic material protected by the rigid, mineralized hard tissues of bones and teeth is often targeted for forensic DNA testing in missing persons and unidentified human remains (UHR) investigations. Although standard approaches have narrowed sample selection to a few optimal skeletal elements (i.e., weight-bearing long bones, teeth), subsampling and decisions beyond this can be challenging due to the non-uniform, heterogeneous pattern of postmortem diagenesis. One particular component of skeletal microstructure — the inorganic mineral matrix [hydroxyapatite (HAp), Ca10(PO4)6(OH)2] — is purported to play a major role in DNA preservation. The potential correlation between molecular changes to skeletal microstructure and …
Multiple Contacts Of Drug Contaminated Fingermarks And Their Analysis With Raman Microspectroscopy, Victoria Deprimo
Multiple Contacts Of Drug Contaminated Fingermarks And Their Analysis With Raman Microspectroscopy, Victoria Deprimo
Master's Theses
This thesis research aimed to determine if substrate, enhancement technique, and multiple contacts affect the detection and identification of drugs in fingermarks using Raman Spectroscopy. It has the potential to be of great importance in forensic science as fingermarks are one of the most important traces left behind at crime scenes and illicit drugs are a significant criminal justice problem. Thus, being able to associate illicit drugs with a specific fingermark has great potential for forensic science, as it can put the drugs in the hands of a specific individual.
The ridges of fingerprints trap trace amounts of material that …
Determining The Interference Of Lactate And Lactate Dehydrogenase In An Ethanol Enzyme Assay, Julia Liebl
Determining The Interference Of Lactate And Lactate Dehydrogenase In An Ethanol Enzyme Assay, Julia Liebl
Master's Theses
Enzymatic ethanol assays are widely used to determine blood alcohol content for individuals admitted to a hospital. In some instances, this hospital data is later used as evidence in a court of law, most commonly in DUI cases. As the enzymatic assay targets, but does not exclusively measure ethanol, it is theoretically possible that interference could occur and produce a falsely elevated result, leading to wrongful convictions or other consequences. This project examined the potential of clinically relevant levels of one potential interferant, lactate and the hepatic enzyme lactate dehydrogenase (LDH) to cause significant and falsely elevated results. Threshold levels …
Dna Methylation Methods For Donor Age Prediction Using Touch Dna, Emily Neverett
Dna Methylation Methods For Donor Age Prediction Using Touch Dna, Emily Neverett
Master's Theses
The International Labor Organization (ILO) estimated over 30 million individuals fall victim to human trafficking each year, of which, 50% are children below the age of 16. In 2012, the ILO reported there to be 168 million child laborers worldwide, with many trafficked into hazardous conditions to manufacture consumer products that are sold in developed countries. This is a modern form of slavery with poor working conditions, no access to education, and low wages. The hidden nature of this crime, however, makes it extremely difficult to identify and locate victims of forced child labor, and thus making it challenging to …
Detection Of Larval Aggregations Using A Drone Mounted Thermal Imaging Camera, Megan Descalzi
Detection Of Larval Aggregations Using A Drone Mounted Thermal Imaging Camera, Megan Descalzi
Master's Theses
When a body’s temperature reaches ambient temperature after death, remote detection of the body can become difficult. At this point, useful search tactics include the use of cadaver dogs, search parties and aerial imaging devices, all of which can be costly and time-consuming for every day law enforcement use. This study investigated the potential of a novel search technique in which a small, unmanned aerial system (drone) mounted with a forward-looking infrared radar (FLIR) was utilized to detect decomposing animal carcasses via the heat generated by associated Diptera larval aggregations. Hot water baths were utilized as analogs for larval aggregations …
Forensic Characterization And Discrimination Of Manila Envelopes, María Isabel Sánchez Melo
Forensic Characterization And Discrimination Of Manila Envelopes, María Isabel Sánchez Melo
Master's Theses
Envelopes can be found at crime scenes when ransom, threat letters or potentially harmful substances are sent to victims. Therefore, they are important probative items of evidence analyzed by forensic document examiners. Manila envelopes are commonly used in the United States to transport or send documents as they are made of thick and durable paper. Although there are many studies on the forensic analysis of office paper and paper-based banknotes, and only a few on white envelopes, there does not exist previously published research on the physical or chemical characterization of manila envelopes. The goal of this research was to …
Investigating The Masking Effects Of Blood On Organic Gunshot Residue Using Gc/Ms, Raman, And Atr-Ftir, Tyler Willits
Investigating The Masking Effects Of Blood On Organic Gunshot Residue Using Gc/Ms, Raman, And Atr-Ftir, Tyler Willits
Master's Theses
Gunshot residue (GSR) can be an indicator for determining the distance of a firearm discharge and linking potential suspects and witnesses to a shooting event. However, the evidentiary value of inorganic gunshot residue (IGSR), commonly used to detect GSR, has been called into question due to the potential for false negatives and false positives. Organic gunshot residue (OGSR) acts as supporting evidence; this increases the overall confidence of the examiner, as well as the value of gunshot residue evidence as a whole.
Current research has focused on three main areas for OGSR. The first is what compounds we should identify …
Small Rna Sequencing And Rt-Qpcr Validation Of Forensically Relevant Body Fluids, Karly Johannsen
Small Rna Sequencing And Rt-Qpcr Validation Of Forensically Relevant Body Fluids, Karly Johannsen
Master's Theses
Body fluid identification is an important aspect of forensic investigations because it assists with the reconstruction of crime scenes and can refute and/or support witness statements. Currently, there is no universal method for body fluid identification. Each body fluid has several tests for its identification, both presumptive and confirmatory. A universal method for body fluid identification that is sensitive, specific, efficient, and minimally destructive is necessary.
In recent years miRNAs have been heralded as novel biomarkers for the identification of body fluids. Several research groups across the US and internationally have been formed to determine which miRNAs are suitable for …
Construction Of A Reference Allelic Ladder For An Odocoileus Str Multiplex, Jolene Strand
Construction Of A Reference Allelic Ladder For An Odocoileus Str Multiplex, Jolene Strand
Master's Theses
Odocoileus is a genus of Cervidae (deer) consisting of white-tailed deer (Odocoileus virginianus, Zimmerman, 1780), mule deer (Odocoileus hemionus, Rafinesque, 1817) and blacktailed deer (Odocoileus hemionus hemionus, Rafinesque, 1817). Hunting Odocoileus is only legal with proper permits according to laws that dictate when, where, how, and what can be hunted. Anything outside the legal limits is considered to be poaching. A useful tool for investigating poaching cases is Short Tandem Repeat (STR) DNA analysis. This is commonly used in human forensic casework to link DNA evidence found at crime scenes to either victims or suspects. In poaching cases, it can …
The Impact Of Environmental Degradation On The Analysis Of Manufactured Fibers, Alexis R. Weber
The Impact Of Environmental Degradation On The Analysis Of Manufactured Fibers, Alexis R. Weber
Master's Theses
The analysis of fiber evidence involves the ability to link a questioned fiber back to its known source. But before the collection and analysis of questioned fibers, they could be potentially exposed to various environmental conditions for an extended period of time. There has been little research on how manufactured fibers are affected by various environments and if this interaction affects the ability to link a questioned fiber back to its known source. Changes to the manufactured fibers are essential to be aware of, in order to avoid the possibility of erroneous exclusions when performing fiber comparisons. A six-month time …

Master's Theses
Regulation standards for dietary supplements lack greatly. Due to this, fraud during the manufacturing of these products might be committed. Previous investigations on herbal dietary supplements have shown that the main ingredient was not present in most of the herbal supplements tested and that these products also contained other substances not stated in the label The purpose of this study was to develop a fluorescent qPCR assay for the identification of Morinda citrifolia herbal species in dietary supplements. Primers for genetic regions rbcL and matK of M. citrifolia were designed. The potential of novel matK primer showed discrimination potential …
High Resolution Melt Analysis Of Samples With Differential Dna Methylation To Identify Tissue Source Of Origin, Stephanie M. Ledgerwood
High Resolution Melt Analysis Of Samples With Differential Dna Methylation To Identify Tissue Source Of Origin, Stephanie M. Ledgerwood
Master's Theses
In Forensic Science casework, identifying the source of a tissue can assist in crime scene reconstruction. Currently, presumptive testing methods to identify tissue type are utilized in crime scene laboratories, but there is a need for a more reliable confirmatory test for tissue type identification. High Resolution Melt (HRM) analysis is an innovative technology that has the potential to determine tissue types through variations in DNA methylation patterns. Recently, DNA methylation patterns have been found to correspond with specific tissue types in particular regions of DNA. Two markers, B_SPTB_03 and DDX4 have been effective in differentiating sperm from other tissue …

Millions of dollars in destruction in the past decade have resulted from the use of Chinese drywall in homes. There are also potential health hazards related to this corrosive material. As such, it is important to find a way to identify Chinese drywall. Drywall can be tested for certain markers, such as strontium, sulfur, and carbonates to identify it as corrosive Chinese drywall. The laboratory preparation and analysis should be efficient and cost effective. The methods previously used, such as an X-ray fluorescence gun have had issues with getting a proper reading due to the layers of other materials found …

Sensitivity And Species Specificity Of Bisulfite Modification And Pyrosequencing Technology Used To Identify Tissue-Specific Dna Methylation Patterns In Common Biofluids Foudn In Crime Scenes, Sitara Christian Shirwani
Master's Theses
In order to convict the guilty or exonerate the innocent in criminal cases, it is crucial to reconstruct the crime scene and/or determine the nature of the crime. Identifying the different biofluids found at a crime scene can help shed light on these aspects of forensic casework. Recently, DNA methylation has been used as a means of identifying biological materials, as opposed to conventional protein/enzyme based methods. DNA methylation is the addition of a methyl group to the 5' carbon on a cytosine base (C), which is directly followed by a guanine base (G) and are called CpG sites. The …
Comparison Of Liquid-Liquid Extraction And Solid Phase Extraction Of Mephedrone, Brittany Simone Love
Comparison Of Liquid-Liquid Extraction And Solid Phase Extraction Of Mephedrone, Brittany Simone Love
Master's Theses
Legal high drugs are modified scheduled drugs. As with any new drug, researchers have to conduct studies to gather information about the drug. The problem with obtaining accurate information on new drugs is that by the time information is gathered, drug abusers and street chemists have developed new ones. Comparing designer drugs to their illegal counterparts is often helpful in that it can provide a starting point. Mephedrone is a new designer drug that has become a problem over the past few years. Often marketed as bath salts and plant food, mephedrone has become the knockoff replacement for amphetamines. This …

Comparison Of Stated Iron Concentrations Between Name Brand And Generic Children's Multivitamins, Ashlea Nicole Pettengill
Master's Theses
The mineral iron is one of the most common minerals on Earth; not only is iron found in the atmosphere, water, and plants, it is an essential mineral needed for the human body. Children need iron at a young age to ensure proper growth and development of myelin, to promote oxygen transportation within red blood cells, and to assist in Deoxyribonucleic acid (DNA) and Ribonucleic acid (RNA). With iron readily available in children's multivitamins, iron toxicity can become an issue when taking too many tablets. With two types of brands, generic and name brand, parents should be aware of the …
Single Liquid-Liquid Extraction Of Delta-9-Tetrahydrocannabinol And 11-Nor-Carboxy-Delta-9-Tetrahydrocannabinol At An Acidic Ph, Meghan Ann Tanner
Single Liquid-Liquid Extraction Of Delta-9-Tetrahydrocannabinol And 11-Nor-Carboxy-Delta-9-Tetrahydrocannabinol At An Acidic Ph, Meghan Ann Tanner
Master's Theses
Because of the high occurrence of marijuana usage in the workplace and in driving under the influence cases, it is important to test for both delta-9-THC and delta-9-THCA in order to determine current and recent usage. The laboratory preparation and analysis of delta-9-THC and delta-9-THCA in urine should be as cost and time efficient as possible. Current extraction of delta-9-THC and delta-9-THCA from urine requires two separate techniques for each. Because delta-9-THC is nonacidic, while delta-9-THCA is acidic, the extractions involve two separate pH levels, while GC/MS analysis requires two separate methods. This separation requires a great deal of time …
Capecitabine Induced Hand And Foot Syndrome And The Reproducibility Of Friction Skin, Rodney Allen Schenk
Capecitabine Induced Hand And Foot Syndrome And The Reproducibility Of Friction Skin, Rodney Allen Schenk
Master's Theses
In 2008, a 62 year old male was detained by United States Customs and Immigration officials when attempting to enter the United States because his fingerprints were not detectable. It was later reported by his medical doctor in Singapore that the individual suffered from Hand and Foot Syndrome (palmar-plantar erythrodysaesthesia) as a result of his cancer treatment of capecitabine (N4-pentyloxycarbonyl-5-deoxy-5- fluorocytidine) which causes interruptions to the normal growth of keratinocytes in the friction skin. Capecitabine is a recently developed, orally administered fluoropyrimidine prodrug designed to generate 5-fluorouracil through a three-step enzymatic process giving it antineoplastic properties to combat cancerous tumor …

Efficiency Of Using Commercial Dna Extraction Kits And The Organic Extraction Method In Removal Of Polymerase Chain Reaction Inhibitors, Brandi Jo Payne
Master's Theses
The first step in the determination of a perpetrator of a crime using DNA profiling is obtaining good quality DNA. The substrate on which the body fluid is located may contain substances that can co-extract with DNA and may inhibit subsequent PCR analysis. In this study, the efficiency of the removal of such contaminants were tested using three different methods, namely the PrepFiler DNA extraction kit, DNA IQ DNA extraction system, and the organic DNA extraction protocol. One, two, five, and ten μl of whole blood were deposited on soil, wood chips, and cotton swatches treated with bleach. DNA was …
