The goal of this study is to internally validate the ANDE 6C Rapid DNA I-Chip reagent for use on disaster victim identification (DVI) samples. The internal validation tested nine samples each of postmortem blood, tissue, and bone. This study tested the instrument’s precision, accuracy, repeatability, and reproducibility. Precision and accuracy were tested by confirming that the same sample run on the ANDE instrument and the Power Plex Fusion amplification system produced a DNA profile that is consistent with the known profile. The goal of this project was to perform a validation and a method development for an Agilent 7890 Gas Chromatograph connected to a Thermo iS50 Fourier Transform Infrared Spectrophotometer in order to put the instrument into functional and operational use. Both instruments are connected via an interface allowing for simultaneous analysis of a sample. A custom-made in-house library was created in order to search and compare to future unknown case samples. Bullets can be broadly differentiated into jacketed and unjacketed groups. Jacketed ammunition can be further broken down based on the bullet’s design, one of which is hollow points. This research examines a representative brand of jacketed hollow point bullets used in a common caliber cartridge, the 9mm Luger. A jacketed hollow point bullet is designed to expand when it encounters a viscous medium such as tissue. This study attempted to correlate individual biological characteristics to the amount of DNA recovered from a person’s hands (representative of shedding propensity). We used adhesive D-Squame tape to collect DNA from the fingers of more than 100 volunteers with and without controlled activity after handwashing. Two no-activity samples were taken after 30 min from the non-dominant hand and two samples from the dominant hand were collected after 60 min of controlled activity. In forensic science, DNA extraction can be a tedious and resource-intensive process. Extraction with Proteinase K is an industry standard but has its drawbacks, such as requiring multiple ionic detergents and washing steps. MicroGEM has developed a new enzyme called forensicGEM that is temperature-dependent and compatible with mesophilic enzymes, offering complete DNA extraction in about 20 minutes in a single tube, limiting contamination, loss of sample, and working time. Previous research has found that cosmetic hair treatments can decrease concentrations of drugs that are already present in the hair shaft, however little is known about the impact of these treatments on the hair when the drugs are deposited externally. This project serves to provide additional information on the prevalence of non-self DNA on the exposed skin locations specifically the inner upper arm, the nape area of the neck and the neck area just below the ear, where current literature is lacking. Skin tape lifts from these locations were collected from 15 male participants and 13 female participants once a week over a period of three weeks. This project suggests improvements to the treatment methods utilized for the sample preparation and evaluation of minerals in soil samples encountered in forensic science casework and research. The evaluation of pre-treated and post-treated soil samples were performed using Fourier-Transform Infrared Spectroscopy with Attenuated Total Reflection (FTIR-ATR) and Scanning Electron Microscopy-Energy Dispersive Spectroscopy (SEM-EDS). Because soil samples contain a significant amount of organic matter and water which interfere with examination of these minerals while using these instruments, pretreatment methods are necessary to isolate the mineral fraction for examination and analysis.

The current understanding is an immediate expulsion of soil gaseous mercury upon water treatment and an upward transport of dissolved soil ionic mercury as the water evaporates. An opioid abuse crisis in recent years has contributed to many overdoses and overdose-related deaths in the United States. In 2017, fentanyl and fentanyl analogs contributed to more than half of NYC’s overdose deaths in 2017. There is known individual variation in DNA left behind during contact, this DNA shedding propensity has an effect on whose DNA is detected. This study evaluated this variability using a cell staining approach. In this research, bovine leg bones were burned, both with and without meat intact, in order to determine if it was possible to extract and amplify DNA to obtain profiles. The meat was burned over an open flame and the DNA was extracted using the Qiagen DNeasy Blood & Tissue Mini Kit, quantified with the NanoDrop, and amplified using the Bovine Genotypes Panel 3.1 Kit, and separated using the 3500 Genetic Analyzer. Decomposed human remains are complex forensic puzzles, escalating in difficulty as the remains’ age obscures evidence, like trauma. Research has shown that scanning electron microscopes with energy dispersive X-ray spectrometers (SEM-EDX) are capable of detecting and identifying gunshot residue (GSR) particles on bones. However, SEM-EDX work is time consuming, expensive, and not accessible to every forensic department. Cocaine overdose deaths are on the rise in the United States. This increase may be due to the co-consumption of cocaine with other drugs. When cocaine is ingested in conjunction with alcohol, a specific metabolite called cocaethylene forms. Information regarding cocaethylene, specifically its interpretive value in post-mortem cases, has not been widely studied. Data from the Office of the Chief Medical Examiner in New York City revealed that there were 195 cases in 2017 positive for cocaethylene. These cases were most likely to involve black (42.1%) males (79.5%) in Manhattan (28.2%) aged 50 - 59 (34.4%). Most of the cases …

In the United States, there has been a significant increase in the presence of methamphetamine in recent years, specifically in the Northeastern region of the country. In this experiment, a method of calculating ballistic coefficients of .22LR (long rifle) caliber ammunition was developed using Doppler radar to track the pre-impact and post-impact velocities of projectiles passing through gypsum board. The method demonstrated has distinct advantages in accuracy and flexibility over conventional chronograph projectile tracking that forensic firearm experts should consider when determining the optimal method to obtain a ballistic coefficient (BC).
