The potential is great for liposome drug delivery systems that provide specific contents release at diseased tissue sites upon activation by upregulated enzymes; however, this potential will only come to fruition with mechanistic knowledge of the contents release process. NAD(P)H:quinone oxidoreductase type 1 (NQO1) is a target for reductively-responsive liposomes, as it is an enzyme upregulated in numerous cancer tissues and is capable of reducing quinone propionic acid (QPA) trigger groups to hydroquinones that self-cleave from dioleolylphosphatidylethanolamine (DOPE) liposome surfaces, thereby initiating contents release. The work presented in this dissertation is a description of novel core-shell nanomaterials derived from a group of uniform materials based on organic salts (GUMBOS) and the synthesis and characterizations of novel liposomal ionogels (LIGs). These GUMBOS are an extension of ionic liquids (ILs) which are organic salts with melting points between 25 °C and 100 °C. Ionic liquids have high thermal stability, low vapor pressure, and tunable physiochemical and functional properties. All of the properties of ILs are controlled by the chosen cation and anion pair. Over the last few decades, liposomes have generated a lot of interest as drug delivery vehicles to address the need for providing both increased therapeutic efficacy and decreased systemic exposure, simultaneously. The challenge of increasing drug accumulation at diseased sites, without compromising the integrity and stability of the liposomal carrier during circulation, has been met with two possible solutions: (1) active targeting and (2) active triggering. Liposomes have had a long and fruitful application of drug delivery devices in the treatment of select tumors and cancer types. Third-generation liposomes possess endogenous triggering methods that hold the ability for site-specific unloading of liposomal contents, thus providing necessary temporal and spatial control over contents release that can be tuned to match the drug efficacy for decreased side effects during treatment. Of these new endogenous triggers, redox-active liposomes that can be triggered to unload contents via two-electron reduction by enzymes or chemical means have demonstrated promise for the encapsulation and delivery of cargo. Self-assembly is scientifically interesting, technologically important, and sometimes life-threatening. A striking example: Alzheimer’s disease is caused by the self-assembly of the â-amyloid fragment of amyloid precursor protein. Such natural amphiphiles normally are very expensive and complicated to isolate. Synthetic mimics that are much cheaper and easier to prepare can facilitate the development of efficient characterization methods, even as they suggest new materials science applications. Two-directional arborols are synthetic bolaform amphiphilic dendrimers. They dissolve in hot water and form reversible gels at room temperature. Bundles of fibrils, which are made by self-assembly of arborols, are found in the gels. The properties …
