Quinones are a class of organic compounds containing a six-membered unsaturated ring with two carbonyl groups. They are biologically relevant mostly due to their ability to participate in redox reactions. Prior experiments in our lab showed that quinones can induce protein modifications that are pH dependent. In an acidic environment the modifications were less significant than in a basic environment. Previous computational studies have also been carried out to model, in neutral solutions, the reaction between various quinones and various amines. Quinones are molecules that are commonly found in the environment, as cofactors for proteins or an electron carrier in the mitochondrial electron transport chain. However, quinones also can come from polycyclic aromatic hydrocarbons as their metabolites, harming cellular integrity. In this study, we investigated protein modification of lysozyme in the presence of substituted benzoquinones and naphthoquinones. To simulate physiological conditions, all reactions used phosphate buffer (50.0 mM). Quinones are the metabolites of a class of chemicals known as polycyclic aromatic hydrocarbons. These chemicals have been found to be toxic in the environment, especially when interacting with certain proteins. In this study, we investigated the modification of Lysozyme and Ribonuclease A by a series of benzoquinones as well as few naphthoquinones. Fluorescence spectroscopy was used to measure the degree of modification of Lysozyme and Ribonuclease A when incubated with quinones at differing concentrations and for various times.
