While superresolution microscopy has opened the doors to insights into biological phenomena we couldn’t have dreamed of in the last century, its methodology is naturally limited. We aim to push the envelope of its capabilities by testing the effect that Ca2+ and H+ ions have on the fluorescent protein Dendra2. Utilizing a newly designed perfusion chamber, we flow separate solutions containing Ca2+ and H+ ions into a cellular environment, in which the cells in question have been tagged with Dendra2. Radiative Transfer Modeling Of Phytoplankton Fluorescence Quenching Processes, Peng Wang Zhai, Emmanuel Boss, Bryan Franz, P. Jeremy Werdell, Yongxiang Hu

We report the first radiative transfer model that is able to simulate phytoplankton fluorescence with both photochemical and non-photochemical quenching included. The fluorescence source term in the inelastic radiative transfer equation is proportional to both the quantum yield and scalar irradiance at excitation wavelengths. The photochemical and nonphotochemical quenching processes change the quantum yield based on the photosynthetic active radiation. A sensitivity study was performed to demonstrate the dependence of the fluorescence signal on chlorophyll a concentration, aerosol optical depths and solar zenith angles. Fluorescence microscopy is popular for its noninvasive properties and its use in imaging multiple species, simultaneously. Furthermore, superresolution fluorescence microscopy (SRFLM) utilizes photoswitchable proteins to improve the lateral resolution of conventional fluorescence microscopy by an order of magnitude. There is little work conducted on the study of excitation laser polarizations and their effect on the number of localizations as well as the brightness of molecules. This thesis attempts to study the effect of excitation wavelength polarization on the number of localizations and the brightness of molecules by comparing two orientations of circularly-polarized, excitation lasers. Pharmaceuticals and Personal Care Products (PPCPs) pollute our water systems regularly by entering the environment through inefficient methods of water treatment, ineffective sanitation and unregulated industrial and agricultural runoff. Currently, the most common methods of identifying PCPPs are expensive, time consuming and often require the aid of a trained expert. This project aims to further the methodology of utilizing fluorescence spectroscopy as a means of pollutant detection with a focus on contaminant mixtures. Four non-regulated EPA contaminants were fluoresced at compound specific EEM parameters and then again in two component mixtures to determine their limits of detection (LOD). The results …

Chlorophyll a fluorometry has long been used as a method to study phytoplankton in the ocean. In situ fluorometry is used frequently in oceanography to provide depth-resolved estimates of phytoplankton biomass. However, the high price of commercially manufactured in situ fluorometers has made them unavailable to some individuals and institutions. Presented here is an investigation into building an in situ fluorometer using low cost electronics. The goal was to construct an easily reproducible in situ fluorometer from simple and widely available electronic components.
