Mitochondria Imaging And Targeted Cancer Treatment, Tinghan Zhao Dec 2019

Mitochondria Imaging And Targeted Cancer Treatment, Tinghan Zhao

Dissertations

Mitochondria are essential organelles as the site of respiration in eukaryotic cells and are involved in many crucial functions in cell life. Dysfunction of mitochondrial metabolism and irregular morphology have been frequently found in human cancers. The capability of imaging mitochondria as well as regulating their microenvironment is important both scientifically and clinically. Mitochondria penetrating peptides (MPPs), certain peptides that are composed of cationic and hydrophobic amino acids, are good candidates for mitochondria targeting. Herein, a novel MPP, D-argine-phenylalanine-D-argine-phenylalanine-D-argine-phenylalanine-NH2 (rFrFrF), is conjugated with a rhodamine-based fluorescent chromophore (TAMRA). The TAMRA-rFrFrF probe exhibits advantageous properties for long-term mitochondria tracking of …

Cancer Risk Prediction With Whole Exome Sequencing And Machine Learning, Abdulrhman Fahad M Aljouie Dec 2019

Cancer Risk Prediction With Whole Exome Sequencing And Machine Learning, Abdulrhman Fahad M Aljouie

Dissertations

Accurate cancer risk and survival time prediction are important problems in personalized medicine, where disease diagnosis and prognosis are tuned to individuals based on their genetic material. Cancer risk prediction provides an informed decision about making regular screening that helps to detect disease at the early stage and therefore increases the probability of successful treatments. Cancer risk prediction is a challenging problem. Lifestyle, environment, family history, and genetic predisposition are some factors that influence the disease onset. Cancer risk prediction based on predisposing genetic variants has been studied extensively. Most studies have examined the predictive ability of variants in known …

Non-Standard Finite Difference Schemes For Investigating Stability Of A Mathematical Model Of Virus Therapy For Cancer, A. R. Yaghoubi, H. S. Najafi

Applications and Applied Mathematics: An International Journal (AAM)

In this paper, a special case of finite difference method called non-standard finite difference (NSFD) method was studied to compute the numerical solutions of the nonlinear mathematical model of the interaction between tumor cells and oncolytic viruses. The global stability of the equilibrium points of the discrete model is investigated by using the Lyapunov stability theorem. Some conditions were gained for the local asymptotical stability of the equilibrium points of the system. Finally, numerical simulations are carried out to illustrate the main theoretical results. The discrete system is dynamically consistent with its continuous model, it preserves essential properties, such as …

Preparation Of Folic Acid-Carbon Dots-Doxorubicin Nanoparticles As Targeting Tumor Theranostics, Samson Dada

Electronic Theses and Dissertations

Carbon dots (CDs) have attracted much attention as an excellent gene/drug delivery and biological imaging agent for early cancer theranostics. In this study, we prepared two series of nanoparticles (NPs), which are composed of (CDs) with a targeting agent, folic acid (FA), and a chemotherapeutic agent Doxorubicin (Dox). All the NPs and their intermediates were characterized using ultraviolet-visible spectroscopy (UV-vis), fluorescence spectroscopy, and Fourier transform-infrared spectroscopy (FT-IR). The drug loading capacity (DLC) and drug loading efficiency (DLE) of two series of FA-CDs-Dox were assessed using UV-vis absorption spectroscopy at the wavelength of 485 nm. Both showed good DLE and DLC …

Vimentin Protects Cells Against Nuclear Rupture And Dna Damage During Migration, Alison E. Patteson, Amir Vahabikashi, Katarzyna Pogoda, Stephen A. Adam, Kalpana Mandal, Mark Kittisopikul, Suganya Sivagurunathan, Anne Goldman, Robert D. Goldman, Paul A. Janmey

Physics - All Scholarship

Mammalian cells frequently migrate through tight spaces during normal embryogenesis, wound healing, diapedesis, or in pathological situations such as metastasis. Nuclear size and shape are important factors in regulating the mechanical properties of cells during their migration through such tight spaces. At the onset of migratory behavior, cells often initiate the expression of vimentin, an intermediate filament protein that polymerizes into networks extending from a juxtanuclear cage to the cell periphery. However, the role of vimentin intermediate filaments (VIFs) in regulating nuclear shape and mechanics remains unknown. Here, we use wild-type and vimentin-null mouse embryonic fibroblasts to show that VIFs …

Elucidating The Mechanism Of Bone Morphogenetic Protein (Bmp) -Mediated Sox2 Downregulation In Ovarian Cancer, Zainab Motolani Shonibare

Theses and Dissertations

Sex determining region Y-box 2 (Sox2) is a transcription factor essential for maintaining self-renewal and pluripotency of undifferentiated embryonic stem cells. Sox2 is involved in multiple processes of cancer cells, however, regulation of Sox2 expression and the consequences of that regulation in cancer remains elusive. Previously, we demonstrated that BMP9/GDF2 is significantly reduced in expression in ovarian cancer (OVCA) cells, methylated in patient tumors and, promoted anoikis resistance in both breast and OVCA cell lines. In an attempt to identify genes downstream of BMP9 that may provide anoikis resistance, transcriptomics was performed leading to the identification of Sox2, a developmental …

Cancerous Male And Female Gene Expression, Clarissa Farmer, E. Shannon Tass Jun 2019

Cancerous Male And Female Gene Expression, Clarissa Farmer, E. Shannon Tass

Journal of Undergraduate Research

Genetic diagnosing is becoming more popular, as well as more and more accurate. However, many genetic diseases have complex genetic effects and are still not fully understood. Transthyretin Amyloidosis (ATTR; also known as familial or hereditary amyloidosis) is a terminal genetic disease. It is caused by unstable transthyretin proteins that fold improperly, and then deteriorate. The fragmented proteins are deposited outside of the cell and build up in the tissues over time, forming insoluble oligomers. The oligomers continue to grow into Amyloid fibrils, which adversely affect many organs in the body, eventually causing their failure. In order to accurately diagnose, …

Development Of Novel Tumor-Targeted Compounds For Boron Neutron Capture Therapy, Micah John Luderer May 2019

Development Of Novel Tumor-Targeted Compounds For Boron Neutron Capture Therapy, Micah John Luderer

Arts & Sciences Electronic Theses and Dissertations

Glioblastoma multiforme (GBM) represents the most common primary brain tumor among adults. Despite surgical resection and aggressive chemoradiotherapy regimens, the current 2- and 5-year survival rates are only 27% and 9.8%, respectively. The low survival stems from the poor response to conventional therapy and underscores the critical need to develop new therapeutic approaches for GBM treatment. The high recurrence rate observed in GBM is in part attributed to the hypoxic (poorly oxygenated) tumor microenvironment. Hypoxic tumor conditions have been shown to increase metastasis, promote angiogenesis, and confer resistance to chemotherapy and radiation.

Hypoxic tissues are inherently radiation resistant due to …

Synthesis Of Novel Reiii And Rev Metallointercalator Complexes For Use As Dna Damage Probes, Eli Schwartz May 2019

Synthesis Of Novel Reiii And Rev Metallointercalator Complexes For Use As Dna Damage Probes, Eli Schwartz

Honors Program Theses and Projects

Cancer is a disease that affects millions around the globe. Treatments such as radiation and chemotherapy are often used to treat cancerous tumors, but are not 100% effective in curing the patient. Alternatively, some research groups have proposed the use of metallointercalators, metal complexes consisting of a rhodium, ruthenium, or iridium metal center with bidentate supporting ligands such as bipyridine and phenanthroline, or tridentate ligands such as terpyridine. The complexes also contain a bidentate intercalating group such as dipyridophenazine. These complexes possess the ability to directly bind to DNA through intercalation between the stacks of DNA base pairs, giving them …

Caspases And Cancer: Connections Through Circular Dichroism Spectroscopy, Sarah M. Hethcox May 2019

Caspases And Cancer: Connections Through Circular Dichroism Spectroscopy, Sarah M. Hethcox

Honors College Theses

While excessive cell death inevitably leads to negative effects, the endurance of damaged cells in the presence of death signals can be equally detrimental to health. Apoptosis, or programmed cell death, is a highly regulated process in which cues from within or from outside a cell can trigger an irreversible sequence of signals that carry out cell destruction known as the apoptotic cascade. A group of enzymes called caspases play a vital role in this cascade with some participating as initiators and others acting as effectors of protein cleavage and intracellular breakdown. Although it is normal for the activity of …

Machine Learning Classification Of Primary Tissue Origin Of Cancer From Dna Methylation Markers, Sravani Gannavarapu Surya Naga May 2019

Machine Learning Classification Of Primary Tissue Origin Of Cancer From Dna Methylation Markers, Sravani Gannavarapu Surya Naga

UNLV Theses, Dissertations, Professional Papers, and Capstones

Cancer is one of the leading causes of death globally and was responsible for approximately 9.6 million deaths in 2018. One of the main reason for deaths from cancer is late-stage presentation and inaccessible diagnosis and treatment. Cancer often spreads from the part of the body where it started (primary site) to a different part of the body (metastatic site). Identifying the primary site of cancer plays a key role as it directs the appropriate treatment. Cancer which spreads needs the same treatment as its origin. Having this knowledge can help doctors to decide the type of treatment.

All cancers …

Using Fundamental Properties Of Light To Investigate Photonic Effects In Condensed Matter And Biological Tissues, Laura A. Sordillo Jan 2019

Using Fundamental Properties Of Light To Investigate Photonic Effects In Condensed Matter And Biological Tissues, Laura A. Sordillo

Dissertations and Theses

Light possesses characteristics such as polarization, wavelength and coherence. The interaction of light and matter, whether in a semiconductor or in a biological sample, can reveal important information about the internal properties of a system. My thesis focuses on two areas: photocarriers in gallium arsenide and biomedical optics. Varying the excitation wavelength can be used to study both biological tissue and condensed matter. I altered the excitation wavelengths to be in the longer near-infrared (NIR) optical windows, in the shortwave infrared (SWIR) range, a wavelength region previously thought to be unusable for medical imaging. With this method, I acquired high …

Design, Synthesis, Characterization And Anti-Cancer Effects Evaluation Of Interchain Cysteine Linked Antibody Drug Conjugates, Ke Li Jan 2019

Design, Synthesis, Characterization And Anti-Cancer Effects Evaluation Of Interchain Cysteine Linked Antibody Drug Conjugates, Ke Li

Doctoral Dissertations

"Antibody drug conjugates (ADCs) are emerging therapeutic products specially designed for the treatment of cancers. Potent cytotoxic agents are linked to antibodies via linkers. Antibody drug conjugates, which take advantage of both antibodies and cytotoxic agents, showed high selectivity towards cancer cells. Cytotoxic agents are delivered to the cancer cells selectively and kill them from inside while leaving low or non-toxicity towards normal cells. Antibody drug conjugates are actually heterogeneous mixtures. The major heterogeneities mainly lie in the drug to antibody ratio (DAR) and drug linking positions, both of which potentially affect the therapeutic index of ADCs. In this research, …

Simulating An Immune Response With A Combined Agent-Based Model Of A Triple-Negative Breast Cancer Tumor And Vascular Network, Michael J. Ventoso Jan 2019

Simulating An Immune Response With A Combined Agent-Based Model Of A Triple-Negative Breast Cancer Tumor And Vascular Network, Michael J. Ventoso

Senior Projects Fall 2019

Cytotoxic T-cells (CTLs) are one mechanism the immune system employs to eliminate cancer cells. In this study, I expand upon a previous 3-dimmensional agent-based model of triple-negative breast cancer to include a therapy simulating an immune response in the form of a CTL insertion into the tumor. The model consists of the tumor, comprised of progenitor cells, cancer stem cells, and tumor-associated macrophages, as well as an expanding vascular network. I investigate the effects of inserting different amounts of CTLs into the space, and their effect on tumor size in the short and longer terms. The results show that while …

Microarray Data Analysis And Classification Of Cancers, Grant Gates Jan 2019

Microarray Data Analysis And Classification Of Cancers, Grant Gates

Williams Honors College, Honors Research Projects

When it comes to cancer, there is no standardized approach for identifying new cancer classes nor is there a standardized approach for assigning cancer tumors to existing classes. These two ideas are known as class discovery and class prediction. For a cancer patient to receive proper treatment, it is important that the type of cancer be accurately identified. For my Senior Honors Project, I would like to use this opportunity to research a topic in bioinformatics. Bioinformatics incorporates a few different subjects into one including biology, computer science and statistics. An intricate method for class discovery and class prediction is …

Killing Breast Cancer One Porphyrin At A Time, Taylor C. Lymburner Jan 2019

Killing Breast Cancer One Porphyrin At A Time, Taylor C. Lymburner

Honors Theses

New treatments for cancer are continuously being developed and improved. One such treatment is Photodynamic Therapy, more commonly referred to as PDT. PDT is quickly becoming more popular due to its relative lack of side effects that are present in other treatments. In PDT, light-sensitive agents are required and are activated by light in the targeted cells. There are many types of PDT agents but the one focused on in this research is a four-pyrrole ring structure known as a porphyrin. The combination of H2 TPPC with 3- amino-oxetane-3-yl-methanol created the final product ofH2TPP-Oxo-MeOH. Once the porphyrin was formed, it …
