The goal of this study is to determine whether or not there is an appreciable difference between concentrations of various metal ions present in conventionally grown and processed and organically grown and processed baby foods. Two prominent, commercially available brands were chosen to undergo comparative studies between both their own conventional and organic varieties of second stage green beans and carrots. Samples were tested for cadmium, calcium, iron, lead, nickel, and zinc. Two containers of each variety of baby food were sampled in triplicate, with purchases of foods being made in separate areas to ensure that each set came from …

Reported in this dissertation are the results of investigations performed at the New Orleans Center for Mass Spectrometry at the University of New Orleans. The projects that are detailed in the coming pages take on a variety of subjects, but a common thread is that each employs matrix-assisted laser desorption/ionization (MALDI) mass spectrometry to solve a problem. Fundamental aspects of MALDI in-plume ionization are implicated in the introduction of a newly developed electron-transfer secondary ionization matrix. This research is done to determine the distribution of flavonoids in diverse varieties of onions (commercial and wild onions). Reflux extraction method was done on dried onions and the abstracts were analyzed subsequently by high performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GCMS). Statistical calculations were done to see if there are significant differences in the varieties of onions studied from the chromatographic profiles obtained. The chromatograms obtained were patterned using visual observations, scatter plot study, correlation co-efficient, and ANOVA to evaluate the significant difference of the distribution of flavonoids in varieties of onions. Liquid chromatography tandem mass spectrometry has become a powerful tool for investigating biological systems. Herein we describe the development of both isotope dilution mass spectrometry methods and targeted metabolomics methods for the study of metabolic and cell-cell signaling applications.
A putative yeast enzyme was characterized by discovery metabolite profiling, kinetic flux profiling, transcriptomics and structural biology. These experiments demonstrated that the enzyme shb17 was a sedoheptulose bisphosphatase that provides a thermodynamically dedicated step towards riboneogenesis, leading to the redefinition of the canonical pentose phosphate pathway.

In forensic science antipsychotics are drugs of considerable interest because of their potential abuse, involvement in suicides, and they are frequently associated with sudden death investigations. Polyaniline (PANI) has been extensively investigated due to its unique conductivity, which is primarily dependent on the oxidation state of the material and the solution pH. PANI is an insulator when it is oxidized, and the solution pH exceeds four. Numerous attempts to extend PANI's conductivity to neutral and alkaline environments have been unsuccessful. However, our previous study has demonstrated that PANI/Pd composite is conductive and catalytically active in a basic solution, pH ∼ 12. Knowledge of low-energy cross sections for stable and unstable nuclei in neutron-, γ- and ion induced reactions is important in both the simulation and radiochemistry diagnosis of nuclear device performance. This dissertation explores extraction chromatography and ion chromatography methods for the intragroup separation of trivalent actinides and lanthanides in support of cross section measurements for isotopes of interest. Redox cycling is an electrochemical technique that cycles the reversible redox species between its oxidative states repeatedly on generator and collector electrodes. Two or more individually-addressable microelectrodes located close to each other allow redox cycling to be possible. Electrochemical behavior of a biologically important molecule, dopamine is examined under redox cycling conditions. To our knowledge, this is the first report on detection of physiological concentration of dopamine in presence of up to 100 times excess ascorbate with the use of redox cycling, without the involvement of polymer coating such as Nafion®.

Biomaterial implantation induces the foreign body response (FBR). Development of longer-term implants relies on the thorough understanding of the FBR. The progression of the FBR is regulated by a number of biomolecules including cytokines, chemokines, and matrix metalloproteinases (MMPs). The nature of the FBR requires the spatial and temporal regulation of these mediators. MMPs are an extremely large and diverse group of enzymes that play key roles in regulating the FBR. This thesis presents work on the mechanism and kinetics of the surface film reactions involving the conversion of aqueous species on solid surfaces, and specifically, the reactions of iodide and bromide on silver oxide on silver substrate. These reactions provide a method of immobilizing unwanted halides and are particularly suited to control of hazardous radioactive iodine. Potential applications of this research include nuclear reactor safety and post-accident radioiodine management, and the safe production and use of the medical isotope 131I.

One of the most important developments of the last decade has been the emergence of new methods to dynamically resolve racemic organolithiums using stoichiometric amounts of the chiral ligand. When this concept is implemented successfully, it obviates the need for covalently attached chiral auxiliary based methods, asymmetric deprotonation, and asymmetric synthesis of a precursor stannane as ways to access enantioenriched organolithium compounds for use in asymmetric synthesis. Membrane proteins continue to represent a major challenge for most analytical techniques. Using bacteriorhodopsin (BR) as model system, this work aims to develop mass spectrometry (MS)-based approaches for exploring the structure, dynamics and folding of membrane proteins.
As the first step, BR in its native lipid environment was exposed to hydroxyl radicals, which were produced by laser photolysis of hydrogen peroxide. It was found that the resulting methionine (Met) labeling pattern was consistent with the known BR structure. Though mass spectrometry has earned a central role in the field of proteomics due to its versatility in a wide range of experiments, challenges and complications are still encountered when using mass spectrometry to characterize protein structures, post-translational modifications (PTMs), and abundances. This dissertation explores lanthanide speciation in liquid solution systems related to separation schemes involving the acidic ligands: bis(2-ethylhexyl) phosphoric acid (HDEHP), lactate, and 8-hydroxyquinoline. Equilibrium speciation of neodymium (Nd3+ ), sodium (Na+ ), HDEHP, water, and lactate in the TALSPEAK liquid-liquid extraction system was explored under varied Nd3+ loading of HDEHP in the organic phase and through extraction from aqueous HCl and lactate media. Bacterial spores have remarkable resistance to a variety of harsh conditions, causing spoilage in food industry and becoming the primary bacterial agent in biowarfare and bioterrorism. In this study, inactivation mechanisms of Bacillus amyloliquefaciens (BA) spores by non-thermal plasma (NTP) were investigated by using Fourier-transform infrared spectroscopy (FTIR) as a major tool to exam spores after NTP treatment. Drug delivery is one of the most important concepts for cancer patients because it is oftentimes very difficult or nearly impossible to exclusively target cancerous cells. In this study, the aim was to target receptors on cancerous cells, specifically the Folate Receptor (FR). This specific receptor was targeted because it has been proven to enhance intracellular delivery being that folic acid is an integral species in normal cell function and also that cancerous cells tend to exhibit an abnormally high concentration of Folic Acid on cell surfaces.

The purpose of this research is to develop a unique nanoparticulate system that is capable of delivering antibiotics in a locally applied and extended-release manner for patients receiving cranial replacement implants and bone grafting. This study involves three major components: first, encapsulate both hydrophobic antibiotics (chloramphenicol and rifampicine) and hydrophilic antibiotics (vancomycine and acyclovir) into nanoparticles. Chronic wounds are considered as a pandemic health problem. Approximately 1% to 2% of the population in developing countries has experienced a chronic wound during their lifetime.
