Telomeres form the ends of linear chromosomes and usually comprise protein complexes that bind to simple repeated sequence motifs that are added to the 3′ ends of DNA by the telomerase reverse transcriptase (TERT). One of the primary functions attributed to telomeres is to solve the “end-replication problem” which, if left unaddressed, would cause gradual, inexorable attrition of sequences from the chromosome ends and, eventually, loss of viability. Telomere-binding proteins also protect the chromosome from 5′ to 3′ exonuclease action, and disguise the chromosome ends from the double-strand break repair machinery whose illegitimate action potentially generates catastrophic chromosome aberrations. Telomeres …

Non-Transgenic Crispr-Mediated Knockout Of Entire Ergot Alkaloid Gene Clusters In Slow-Growing Asexual Polyploid Fungi, Simona Florea, Jolanta Jaromczyk, Christopher L. Schardl Feb 2021

Non-Transgenic Crispr-Mediated Knockout Of Entire Ergot Alkaloid Gene Clusters In Slow-Growing Asexual Polyploid Fungi, Simona Florea, Jolanta Jaromczyk, Christopher L. Schardl

Computer Science Faculty Publications

The Epichloë species of fungi include seed-borne symbionts (endophytes) of cool-season grasses that enhance plant fitness, although some also produce alkaloids that are toxic to livestock. Selected or mutated toxin-free endophytes can be introduced into forage cultivars for improved livestock performance. Long-read genome sequencing revealed clusters of ergot alkaloid biosynthesis (EAS) genes in Epichloë coenophiala strain e19 from tall fescue (Lolium arundinaceum) and Epichloë hybrida Lp1 from perennial ryegrass (Lolium perenne). The two homeologous clusters in E. coenophiala—a triploid hybrid species—were 196 kb (EAS1) and 75 kb (EAS2), and …

White Pine Blister Rust Distribution In New Hampshire 1900-2018: Exploring The Impacts Of An Exotic Pathogen On Forest Composition And Succession, Janine Marr Jan 2021

White Pine Blister Rust Distribution In New Hampshire 1900-2018: Exploring The Impacts Of An Exotic Pathogen On Forest Composition And Succession, Janine Marr

Antioch University Dissertations & Theses

White pine blister rust (WPBR) has been affecting New Hampshire’s white pines for more than a century, yet no data exist on the long-term effects of the non-native disease on the state’s forests, particularly with respect to the regeneration and sustainability of white pine, and forest succession. This study aimed to address the gaps in the literature by exploring: 1) the current distribution, incidence, and severity of WPBR in New Hampshire; 2) the application of two historical hazard ratings models, one climatic, and one biotic; and 3) the long-term effects of the disease on forest composition, structure, and succession. Historical …

Arbuscular Mycorrhizal Fungi And Exogenous Glutathione Mitigate Coal Fly Ash (Cfa)-Induced Phytotoxicity In Cfa-Contaminated Soil, Olushola M. Awoyemi, Ekundayo O. Adeleke, Kudjo E. Dzantor May 2019

Arbuscular Mycorrhizal Fungi And Exogenous Glutathione Mitigate Coal Fly Ash (Cfa)-Induced Phytotoxicity In Cfa-Contaminated Soil, Olushola M. Awoyemi, Ekundayo O. Adeleke, Kudjo E. Dzantor

Agricultural and Environmental Sciences Faculty Research

Coal fly ash (CFA) makes a bulk of the coal combustion wastes generated from coal-fired power plants. There are several environmental mishaps due to coal ash spills around the world and in the United States. Management of CFA-polluted sites has proven inefficient resulting in soil infiltration, leaching, and phytotoxicity. This study assessed the mitigation strategies for CFA-induced phytotoxicity using biological [arbuscular mycorrhizal fungi (AMF)] and chemical [exogenous glutathione (GSH)] agents. Indices of phytotoxicity include seed germination, plant morphometrics, lipid peroxidation and genomic double-stranded DNA (dsDNA) in switchgrass plant (Panicum virgatum). Experiments include laboratory screening (0, 5, 10, 15 and 20% …

Gwas For Fusarium Head Blight Related Traits In Winter Wheat (Triticum Aestivum L.) In An Artificially Warmed Treatment, Elisane W. Tessmann, David A. Van Sanford

Plant and Soil Sciences Faculty Publications

Global temperature increases will affect Fusarium head blight (FHB) levels in wheat (Triticum aestivum L.). A pressing question is whether current sources of resistance will be effective in a warmer environment. We evaluated phenotypic response to disease in 238 soft winter wheat breeding lines and cultivars grown in 2015–2016 and 2016–2017 under control and warmed (+3 °C) conditions. Warming was achieved with heating cables buried 3 cm in the rhizosphere. We measured heading date, plant height, yield, FHB rating, Fusarium damaged kernels (FDK), deoxynivalenol (DON), leaf blotch rating, powdery mildew rating and leaf rust rating. There were significant ( …

Climate Change And The Integrity Of Science, Peter H. Gleick, James L. Van Etten, Members Of The U.S. National Academy Of Sciences Nov 2016

Climate Change And The Integrity Of Science, Peter H. Gleick, James L. Van Etten, Members Of The U.S. National Academy Of Sciences

James Van Etten Publications

We are deeply disturbed by the recent escalation of political assaults on scientists in general and on climate scientists in particular. All citizens should understand some basic scientific facts. There is always some uncertainty associated with scientific conclusions; science never absolutely proves anything. When someone says that society should wait until scientists are absolutely certain before taking any action, it is the same as saying society should never take action. For a problem as potentially catastrophic as climate change, taking no action poses a dangerous risk for our planet.

Scientific conclusions derive from an understanding of basic laws supported by …

Genetic Engineering And Sustainable Crop Disease Management: Opportunities For Case-By-Case Decision-Making, Paul Vincelli May 2016

Genetic Engineering And Sustainable Crop Disease Management: Opportunities For Case-By-Case Decision-Making, Paul Vincelli

Plant Pathology Faculty Publications

Genetic engineering (GE) offers an expanding array of strategies for enhancing disease resistance of crop plants in sustainable ways, including the potential for reduced pesticide usage. Certain GE applications involve transgenesis, in some cases creating a metabolic pathway novel to the GE crop. In other cases, only cisgenessis is employed. In yet other cases, engineered genetic changes can be so minimal as to be indistinguishable from natural mutations. Thus, GE crops vary substantially and should be evaluated for risks, benefits, and social considerations on a case-by-case basis. Deployment of GE traits should be with an eye towards long-term sustainability; several …

Identifying Rare Fhb-Resistant Segregants In Intransigent Backcross And F2 Winter Wheat Populations, Anthony J. Clark, Daniela Sarti-Dvorjak, Gina Brown-Guedira, Yanhong Dong, Byung-Kee Baik, David A. Van Sanford Mar 2016

Identifying Rare Fhb-Resistant Segregants In Intransigent Backcross And F2 Winter Wheat Populations, Anthony J. Clark, Daniela Sarti-Dvorjak, Gina Brown-Guedira, Yanhong Dong, Byung-Kee Baik, David A. Van Sanford

Plant and Soil Sciences Faculty Publications

Fusarium head blight (FHB), caused mainly by Fusarium graminearum Schwabe [telomorph: Gibberella zeae Schwein.(Petch)] in the US, is one of the most destructive diseases of wheat (Triticum aestivum L. and T. durum L.). Infected grain is usually contaminated with deoxynivalenol (DON), a serious mycotoxin. The challenge in FHB resistance breeding is combining resistance with superior agronomic and quality characteristics. Exotic QTL are widely used to improve FHB resistance. Success depends on the genetic background into which the QTL are introgressed, whether through backcrossing or forward crossing; QTL expression is impossible to predict. In this study four high-yielding soft red …

Developing The Framework For A Risk Map For Mite Vectored Viruses In Wheat Resulting From Pre-Harvest Hail Damage, Anthony L. Nguy-Robertson, Arthur Zygielbaum, Anthony J. Mcmechan, Gary L. Hein, Stephen N. Wegulo, Abby R. Stilwell, Travis M. Smith Jan 2016

Developing The Framework For A Risk Map For Mite Vectored Viruses In Wheat Resulting From Pre-Harvest Hail Damage, Anthony L. Nguy-Robertson, Arthur Zygielbaum, Anthony J. Mcmechan, Gary L. Hein, Stephen N. Wegulo, Abby R. Stilwell, Travis M. Smith

Department of Plant Pathology: Faculty Publications

There is a strong economic incentive to reduce mite-vectored virus outbreaks. Most outbreaks in the central High Plains of the United States occur in the presence of volunteer wheat that emerges before harvest as a result of hail storms. This study provides a conceptual framework for developing a risk map for wheat diseases caused by mite-vectored viruses based on pre-harvest hail events. Traditional methods that use NDVI were found to be unsuitable due to low chlorophyll content in wheat at harvest. Site-level hyperspectral reflectance from mechanically hailed wheat showed increased canopy albedo. Therefore, any increase in NIR combined with large …

A Potential Plan Of Action For Emerald Ash Borer In Nebraska, Lee Wheeler Apr 2010

A Potential Plan Of Action For Emerald Ash Borer In Nebraska, Lee Wheeler

Department of Environmental Studies: Undergraduate Student Theses

Abstract Emerald Ash Borer (Agrilus planipennis) (EAB) is an invasive insect pest. It feeds on the cambium tissues of ash tree species. It was first discovered in the United States in 2002 in Detroit, Michigan. Their effects on ash trees are deadly, and it is quickly spreading across the Midwest. Nebraska has not yet been invaded, but confirmed findings continue getting closer and closer. The major problem facing Nebraskans, with regards to EAB, is how to begin preparations to prevent a dramatic economic loss when an infestation does occur. So, to address this problem, I have conducted street and park …
