Leveraging The 1,3-Azadiene-Anhydride Reaction For The Synthesis Of Functionalized Piperidines Bearing Up To Five Contiguous Stereocenters, Jorge Garcia, Jane Eichwald, Jayme Zesiger, Timothy K. Beng

Student Published Works

A modular and scalable strategy, which remodels 3-methylglutaric anhydride to 2-oxopiperidines bearing at least three contiguous stereocenters is described. The approach relies on the chemoselective and stereocontrolled annulation of 1,3-azadienes with the anhydride component. The resulting acid-tethered allylic 2-oxopiperidines are then engaged in several selective fragment growth processes, including catalytic denitrative alkenylation, halolactonization, and Vilsmeier–Haack functionalization.

Landfills contain a trove of valuable materials, such as critical, precious, and rare earth metals, that are integral to the United State’s economy and national security. The leachate that filters through landfills picks up these materials, which allows for the possibility of recovery. For this research, samples will be analyzed from landfills throughout the Midwestern United States to provide a baseline on water quality constituents, elements present, and microbial activity. Preliminary data for this study was acquired by analyzing samples of landfill leachate from a landfill in northern Indiana. pH readings indicate that the leachate is slightly basic. It also …

Cyclodextrins (CD) are a family of oligosaccharides that can non-covalently and precisely bind small hydrophobic drug molecules, such as antibiotics and anticancer drugs, through well-defined host-guest interactions inside the inner cavity of the macrocycle. Ring-opening metathesis polymerization (ROMP) is a versatile, functional group tolerant method of polymerization that is underexplored in biomedical applications relative to the more common controlled chain-growth polymerizations. Electrochemistry provides an alternative pathway to handle redox organic reactions, especially for certain chemical process that can be only achieved through electrochemical process. Herein, there research projects are introduced to enrich the whole synthetic community, including an anodic oxidative cyclization studies to develop methodology on heterocyclic ring skeleton constructions, an electroreduction pathway exploration to more effectively obtain hydrazine from nitroso compounds, as well as a study in paired electrolysis process on which both anode and cathode are simultaneously employed to maximize overall atom and energy efficiency. Given the growing issue in healthcare of antibiotic resistance, effective and safe alternative treatment methods are required. One of these possible alternative treatment methods is bacteriotoxins including bacteriocins and tailocins. The focus of this study is a bacteriotoxin from Burkholderia cenocepacia (ATCC 25608), which was induced for toxin using a modified UV light induction procedure and tested against a variety of Pseudomonas and Burkholderia for its killing capacity. Various other pathogenic strains were then induced with UV light and tested. N-heterocyclic carbenes are essential for the synthesis and stabilization of various metal complexes due to the tendency to act as sigma-donor of sp2 hybridized lone pair in carbene into sigma-accepting orbital of metals. 1,2-Diazetidine is a four-membered ring heterocyclic compound which has two adjacent nitrogen atoms. However, the syntheses of C-unsubstituted 1,2-diazetidines are rarely reported in the literature. C-unsubstituted 1,2-diazetidines were synthesized through an operationally simple intermolecular vicinal disubstitution reaction between 1,2-dibromoethane and hydrazine with N-arylsulfonyl as the protecting group. Several different types of C-unsubstituted 1,2-diazetidines derivatives were synthesized with either two of the same or two different N-arylsulfonyl groups. The electronic and steric properties were analyzed using Raman spectroscopy and computational calculations. Complexes of the type [M(Cp/Cp*)(MeCN)(PR2NR′2)]PF6 (M = Fe, Ru) were synthesized and/or tested towards acceptorless dehydrogenation (AD) of amines. The primary amine benzylamine can undergo AD to form a mixture of products, and a selection of catalysts were employed in an attempt to control product selectivity. As a result, trends in product distribution were observed by modifying the primary and secondary coordination spheres on the catalyst. Human fibroblast growth factor one (hFGF1) belongs to a family of 22 FGF members produced by fibroblast cells. Cell signaling during physiological processes of angiogenesis and wound healing occurs when hFGF1 binds to its receptor (FGFR). In their adaptive response, plants use signaling molecules that are trafficked throughout the plant. This research focuses on the chemical synthesis and assessment of analogs of the plant signal phytosulfokine (PSK, a sulfated pentapeptide), its delivery to plants and its observation using a fiber-optic fluorescence microscope. A popular method for the enantioselective synthesis of substituted malonic half esters is hydrolysis via Pig Liver Esterase (PLE), however some substrates produce low enantiomeric excess, namely benzyl-substituted malonic esters. Presented here are alternative methods explored for this synthesis, the first being phase-transfer catalyzed hydrolysis via N-benzyl quaternary ammonium salts derived from cinchona alkaloids. The second method utilized chiral auxiliary directed benzylation with auxiliaries including menthol and oxazolidinones. Though unsuccessful, this research provided valuable groundwork in the investigation for the enantioselective asymmetric synthesis of benzyl-substituted malonic acid esters.

This dissertation details the development of rationally designed dioxomolybdenum catalyst active for deoxydehydration (DODH), the net reduction of diols and polyols into alkenes and dienes. Catalyst design involved variations on dioxomolybdenum(VI) supported by a dianionic meridional pincer ligand. Rational substrate scope was explored using aliphatic diols, aromatic diols, and biomass derived diols. Various reductants were tested for ability to catalyze the reaction. Low molecular weight gelators (LMWGs) are small molecules that self-assemble in appropriate solvents to form three dimensional networks that immobilize the solvent, creating a supramolecular gel. The self-assembly of LMWGs occurs through non-covalent interactions such as hydrogen bonding, aromatic interactions, donor-acceptor interactions, Van der Waals interactions, hydrophobic forces, halogen bonding, etc. Due to self-assembly occurring through reversible non-covalent interactions, supramolecular gels can undergo a gel to solution transformation. The sample was structurally verified, high purity, and 2.98 mmol/L in dissolved DMSO. The clinical success of MRI is in large part due to the employment of gadolinium-based contrast agents (GBCAs). Due to sensitivity limitations, GBCAs are indicated at markedly high dosages (~1 g Gd3+ per exam). A simple and efficient protocol for the construction of 1-aminoisoquinolines was achieved by treating 2-(2-oxo-2- phenylethyl)benzonitriles with amines in the presence of Me 3 Al. The reaction proceeds via a domino nucleophilic addition with subsequent intramolecular cyclisation. This method provides a wide variety of substituted 1-aminoisoquinolines with good func- tional group tolerance. Furthermore, the synthetic utility of this protocol was demonstrated in the successful synthesis of the anti- tumor agent CWJ-a-5 in gram scale.

The Marquis reagent is a well-established and widely used chemical presumptive test for 3,4-Methylenedioxymethamphetamine (MDMA) and methamphetamine. It is composed of concentrated sulfuric acid and 40% formaldehyde, which act upon alkaloids causing them to complex into larger molecules. This complexation causes a color change that can be visually interpreted as a positive or negative result. Almost any sugar molecule can be complexed in this way as well, due to their many OH groups. Mercury (Hg) separation and sensing is of high significance due to Hg(II) environmental mobility and toxicity. Furthermore, the use of Hg in nuclear applications has resulted in its accumulation in several DOE sites, such as in Oak Ridge and Savannah River reservations. Organic mercury species have been found in low activity waste (LAW) streams resulting from high-level waste (HLW) processing at the Savannah River Site (SRS), therefore posing a threat to humans and the environment.
