Organic Copper Binding Ligands And Thiol Compounds Produced By Bacteria And In The Elizabeth River, Virginia, Christina Louise Dryden

This dissertation presents work focusing on copper and organic copper binding ligands in laboratory cultures and the Elizabeth River, Virginia. Laboratory cultures of the marine bacterium Vibrio parahaemolyticus were used to demonstrate the influence of elevated copper concentrations on copper-complexing ligand and thiol production. Copper-complexing ligands similar in binding strength to the strongest natural ligands were detected in V. parahaemolyticus cultures (log K′CuL = 11.8–13.2). A strong correlation (r2 = 0.973) was found between total thiol and copper-complexing ligand concentrations at all copper concentrations examined.

Cyanobacteria (blue-green algae) are associated with production of potent hepatotoxins (Microcystis; microcystin) and dense surface and benthic mats (Lyngbya), which impede critical water resource usages. Water resource managers are in need of effective and efficient treatment techniques for these problematic algae in field situations. Applications of algaecides are considered in situations where algal problems become acute or when critical water usages are threatened. However, laboratory data are needed that accurately predict responses of algae prior to field-scale algaecide applications. Presented here are the first data on copper complexation and speciation in the South Atlantic Ocean. Samples were collected at six vertical profile stations and along the surface (1m) transect during the 1996 IOC Baseline Contaminant Survey. Copper complexation measurements were made for the surface transect (1m) samples and three of the six vertical profile stations (1m - ~500m) using differential pulse anodic stripping voltammetry (DPASV) with a thin mercury film, rotating glassy carbon disk electrode (TMFRGCDE).

Anchovys, croaker, spat, summer flounder and hogchokers were collected monthly to seasonally from November 1972 to October 1973 at three locations in the southern Chesapeake Bay region of Virginia.
The concentrations of mercury, copper and zinc were determined by atomic absorption spectrophotometry in whole fishes. Metal concentration varied with species and location. Anchovys had the highest mean concentrations of mercury and zinc while hog- chokers had the highest mean copper concentration. Mercury levels exceeding O.5 ppm were observed in species from all three loca- tions, but maximum heavy metal concentrations per species were highest in Craney Island fishes.
