MicroRNA (miRNA) expression varies in association with different tissue types and in diseases. Having been found in body fluids including blood and cerebrospinal fluid (CSF), miRNAs constitute potential biomarkers. CSF miRNAs have been proposed as biomarkers for neurodegenerative diseases; however, there is a lack of consensus about the best candidate miRNA biomarkers and there has been variability in results from different research centers, perhaps due to technical factors. Here, we sought to optimize technical parameters for CSF miRNA studies. We examined different RNA isolation methods and performed miRNA expression profiling with TaqMan® miRNA Arrays. More specifically, we developed a customized …

Exposure to environmental toxicants namely polychlorinated biphenyls (PCBs) is correlated with multiple health disorders including liver and cardiovascular diseases. The liver is important for both xenobiotic and endobiotic metabolism. However, the responses of an injured liver to subsequent environmental insults has not been investigated. The current study aims to evaluate the role of a compromised liver in PCB-induced toxicity and define the implications on overall body homeostasis. Male C57Bl/6 mice were fed either an amino acid control diet (CD) or a methionine-choline deficient diet (MCD) during the 12-week study. Mice were subsequently exposed to either PCB126 (4.9 mg/kg) or the …

Understanding the role of differential gene expression in cancer etiology and cellular process is a complex problem that continues to pose a challenge due to sheer number of genes and inter-related biological processes involved. In this paper, we employ an unsupervised topic model, Latent Dirichlet Allocation (LDA) to mitigate overfitting of high-dimensionality gene expression data and to facilitate understanding of the associated pathways. LDA has been recently applied for clustering and exploring genomic data but not for classification and prediction. Here, we proposed to use LDA inclustering as well as in classification of cancer and healthy tissues using lung cancer …
