Nearshore coastal waters are highly dynamic in both space and time. They can be difficult to sample using conventional methods due to their shallow depth, tidal variability, and the presence of breaking waves. High resolution satellite sensors can be used to provide synoptic views of Surface Temperature (ST), but the performance of such ST products in the nearshore zone is poorly understood. Close to the shoreline, the ST pixels can be influenced by mixed composition of water and land, as a result of the sensor’s spatial resolution. The accuracy and precision of satellite sea surface temperature (SST) products in nearshore coastal waters are not well known, owing to a lack of in-situ data available for validation. It has been suggested that recreational watersports enthusiasts, who immerse themselves in nearshore coastal waters, be used as a platform to improve sampling and fill this gap. One tool that has been used worldwide by surfers is the Smartfin, which contains a temperature sensor integrated into a surfboard fin. Background: As more protein atomic structures are determined from cryo-electron microscopy (cryo-EM) density maps, validation of such structures is an important task. Methods: We applied a histogram-based outlier score (HBOS) to six sets of cryo-EM atomic structures and five sets of X-ray atomic structures, including one derived from X-ray data with better than 1.5 Å resolution. Cryo-EM data sets contain structures released by December 2016 and those released between 2017 and 2019, derived from resolution ranges 0–4 Å and 4–6 Å respectively. Water‐borne hormone measurement is a noninvasive method suitable for amphibians of all sizes that are otherwise difficult to sample. For this method, containment‐water is assayed for hormones released by the animal. Originally developed in fish, the method has expanded to amphibians, but requires additional species‐specific validations. We wanted to determine physiological relevance of water‐borne corticosterone in spotted salamanders (Ambystoma maculatum) by comparing concentrations to those taken using established corticosterone sampling methods, such as plasma. Cryo-electron microscopy (cryo-EM) is a structure determination method for large molecular complexes. As more and more atomic structures are determined using this technique, it is becoming possible to perform statistical characterization of side-chain conformations. Two data sets were involved to characterize block lengths for each of the 18 types of amino acids. One set contains 9131 structures resolved using X-ray crystallography from density maps with better than or equal to 1.5 Å resolutions, and the other contains 237 protein structures derived from cryo-EM density maps with 2-4 Å resolutions. Age information derived from calcified structures is commonly used to estimate recruitment, growth, and mortality for fish populations. Validation of daily or annual marks on age structures is often assumed, presumably due to a lack of general knowledge concerning the status of age validation studies. Therefore, the current status of freshwater fish age validation studies was summarized to show where additional effort is needed, and increase the accessibility of validation studies to researchers. In total, 1351 original peer-reviewed articles were reviewed from freshwater systems that studied age in fish. Periodicity and age validation studies were found for 88 freshwater species …

Reproductive stage determination of male gonads has received sparse attention in fish biology literature with few studies detailing the building of gross anatomical- and histologic scales. The meagre (Argyrosomus regius) is one of the world's largest sciaenids and supports a significant regional fishery in European and North African waters whose reproductive patterns are yet to be fully investigated. In the present study, we derive a macroscopic grading system for meagre testis using semi-quantitative graphs that feature the testis variability along the species size range and time of the year. Liquid chromatography coupled with tandem mass spectrometry (LC–MS/MS) has revolutionized the proteomics analysis of complexes, cells, and tissues. In a typical proteomic analysis, the tandem mass spectra from a LC–MS/MS experiment are assigned to a peptide by a search engine that compares the experimental MS/MS peptide data to theoretical peptide sequences in a protein database. The peptide spectra matches are then used to infer a list of identified proteins in the original sample. Validation of new approaches in regulatory toxicology is commonly defined as the independent assessment of the reproducibility and relevance (the scientific basis and predictive capacity) of a test for a particular purpose. In large ring trials, the emphasis to date has been mainly on reproducibility and predictive capacity (comparison to the traditional test) with less attention given to the scientific or mechanistic basis. Assessing predictive capacity is difficult for novel approaches (which are based on mechanism), such as pathways of toxicity or the complex networks within the organism (systems toxicology). This report describes a validation study of Greenhouse gases Observing Satellite (GOSAT) data processing using ground-based measurements of the Total Carbon Column Observing Network (TCCON) as reference data for column-averaged dry air mole fractions of atmospheric carbon dioxide (X CO2). We applied the photon path length probability density function method to validate X CO2 retrievals from GOSAT data obtained during 22months starting from June 2009. Carbon monoxide (CO) is retrieved daily and globally from space-borne IASI radiance spectra using the Fast Optimal Retrievals on Layers for IASI (FORLI) software developed at the Universit´e Libre de Bruxelles (ULB). Column-averaged volume mixing ratios of carbon dioxide and methane retrieved from the Greenhouse gases Observing SATellite (GOSAT) Short-Wavelength InfraRed observation (GOSAT SWIR XCO2 and XCH4 ) were compared with the reference calibrated data obtained by ground-based high-resolution Fourier Transform Spectrometers (g-b FTSs) participating in the Total Carbon Column Observing Network (TCCON). Preliminary results are as follows: the GOSAT SWIR XCO2 and XCH4 (Version 01.xx) are biased low by 8.85 ±4.75 ppm (2.3±1.2 %) and 20.4±18.9 ppb (1.2±1.1 %), respectively. The Bremen Optimal Estimation differential optical absorption spectroscopy (DOAS) (BESD) algorithm for satellite based retrievals of XCO 2 (the column-average dry-air mole fraction of atmospheric CO 2) has been applied to Scanning Imaging Absorption Spectrometer for Atmospheric Cartography (SCIAMACHY) data. It uses measurements in the O 2-A absorption band to correct for scattering of undetected clouds and aerosols. A confirmatory method has been developed to allow for the analysis of fourteen prohibited medicinal additives in pig and poultry compound feed. These compounds are prohibited for use as feed additives although some are still authorised for use in medicated feed. Feed samples are extracted by acetonitrile with addition of sodium sulphate. The extracts undergo a hexane wash to aid with sample purification. The extracts are then evaporated to dryness and reconstituted in initial mobile phase. The samples undergo an ultracentrifugation step prior to injection onto the LC-MS/MS system and are analysed in a run time of 26 minutes. The …

A confirmatory method has been developed to allow for the analysis of eleven nitroimidazoles and also chloramphenicol in milk and honey samples. These compounds are classified as A6 compounds in annex IV of council regulation 2377/90 and therefore prohibited for the use in animal husbandry. Milk samples are extracted by acetonitrile with addition of NaCl, while honey samples are first dissolved in water before a similar extraction. Honey extracts undergo a hexane wash to remove impurities. Both milk and honey extracts are evaporated to dryness and reconstituted in initial mobile phase. Column-averaged volume mixing ratios of carbon dioxide and methane retrieved from the Greenhouse gases Observing SATellite (GOSAT) Short-Wavelength InfraRed observation (GOSAT SWIR XCO2 and XCH4) were compared with the reference data ob- 5 tained by ground-based high-resolution Fourier Transform Spectrometers (g-b FTSs) participating in the Total Carbon Column Observing Network (TCCON). Through calibrations of g-b FTSs with airborne in-situ measurements, the uncertainty of XCO2 and XCH4 associated with the g-b FTS was determined to be 0.8 ppm (0.2%) and 4 ppb (0.2%), respectively. Abstract. This paper presents a validation study of SCanning Imaging Absorption spectroMeter for Atmospheric CHartographY (SCIAMACHY) carbon monoxide (CO) total column measurements from the Iterative Maximum Likelihood Method (IMLM) algorithm using ground-based spectrometer observations from twenty surface stations for the five year time period of 2003–2007. Overall we find a good agreement between SCIAMACHY and ground-based observations for both mean values as well as seasonal variations. For high-latitude Northern Hemisphere stations absolute differences between SCIAMACHY and ground-based measurements are close to or fall within the SCIAMACHY CO 2 precision of 0.2×1018 molecules/cm2 (10%) indicating that SCIAMACHY can observe CO accurately …

This paper presents extensive bias determination analyses of ozone observations from the Atmospheric Chemistry Experiment (ACE) satellite instruments: the ACE Fourier Transform Spectrometer (ACE-FTS) and the Measurement of Aerosol Extinction in the Stratosphere and Troposphere Retrieved by Occultation (ACE-MAESTRO) instrument. Here we compare the latest ozone data products from ACE-FTS and ACE-MAESTRO with coincident observations from nearly 20 satellite-borne, airborne, balloonborne and ground-based instruments, by analysing volume mixing ratio profiles and partial column densities. The ENVISAT validation programme for the atmospheric instruments MIPAS, SCIAMACHY and GOMOS is based on a number of balloon-borne, aircraft, satellite and ground-based correlative measurements. In particular the activities of validation scientists were coordinated by ESA within the ENVISAT Stratospheric Aircraft and Balloon Campaign or ESABC.

The Atmospheric Chemistry Experiment (ACE) satellite was launched on 12 August 2003. Its two instruments measure vertical profiles of over 30 atmospheric trace gases by analyzing solar occultation spectra in the ultraviolet/visible and infrared wavelength regions. The reservoir gases HNO3, ClONO2, and N2O5 are three of the key species provided by the primary instrument, the ACE Fourier Transform Spectrometer (ACE-FTS). This paper describes the ACE-FTS version 2.2 data products, including the N2O5 update, for the three species and presents validation comparisons with available observations. New results of CO global total column measurements using the Atmospheric Infrared Sounder (AIRS) aboard the Aqua satellite in comparison with Measurements of Pollution in the Troposphere (MOPITT) sensor aboard the Terra satellite are presented. Both data sets are validated using ground-based total column measurements in Russia and Australia. A quality parameter based on the Profile Percent A Priori values from the standard MOPITT product is introduced. Vertical profiles of NO2 and NO have been obtained from solar occultation measurements by the Atmospheric Chemistry Experiment (ACE), using an infrared Fourier Transform Spectrometer (ACE-FTS) and (for NO2) an ultraviolet-visible-near-infrared spectrometer, MAESTRO (Measurement of Aerosol Extinction in the Stratosphere and Troposphere Retrieved by Occultation). The ACE-FTS (Atmospheric Chemistry Experiment Fourier Transform Spectrometer) solar occultation instrument that was launched onboard the Canadian SCISAT-1 satellite in August 2003 is measuring vertical profiles from the upper troposphere to the lower mesosphere for a large number of atmospheric constituents. Methane is one of the key species. Hydrogen chloride (HCl) and hydrogen fluoride (HF) are respectively the main chlorine and fluorine reservoirs in the Earth's stratosphere. Their buildup resulted from the intensive use of man-made halogenated source gases, in particular CFC-11 (CCl3F) and CFC-12 (CCl2F2), during the second half of the 20th century. It is important to continue monitoring the evolution of these source gases and reservoirs, in support of the Montreal Protocol and also indirectly of the Kyoto Protocol. Nitric acid (HNO3) is one of the key products that are operationally retrieved by the European Space Agency (ESA) from the emission spectra measured by the Michelson Interferometer for Passive Atmospheric Sounding (MIPAS) onboard ENVISAT. The product version 4.61/4.62 for the observation period between July 2002 and March 2004 is validated by comparisons with a number of independent observations from ground-based stations, aircraft/balloon campaigns, and satellites. Individual HNO3 profiles of the ESA MIPAS level-2 product show good agreement with those of MIPAS-B and MIPAS-STR (the balloon and aircraft version of MIPAS, respectively), and the balloon-borne infrared spectrometers MkIV and SPIRALE, …

The Michelson Interferometer for Passive Atmospheric Sounding (MIPAS), on-board the European ENVIronmental SATellite (ENVISAT) launched on 1 March 2002, is a middle infrared Fourier Transform spectrometer measuring the atmospheric emission spectrum in limb sounding geometry. The instrument is capable to retrieve the vertical distribution of temperature and trace gases, aiming at the study of climate and atmospheric chemistry and dynamics, and at applications to data assimilation and weather forecasting.

Altitude profiles of ClONO2 retrieved with the IMK (Institut fur Meteorologie und Klimaforschung) science-oriented data processor from MIPAS/Envisat (Michelson Interferometer for Passive Atmospheric Sounding on Envisat) mid-infrared limb emission measurements between July 2002 and March 2004 have been validated by comparison with balloon-borne (Mark IV, FIRS2, MIPASB), airborne (MIPAS-STR), ground-based (Spitsbergen, Thule, Kiruna, Harestua, Jungfraujoch, Izana, Wollongong, Lauder), and spaceborne (ACE-FTS) observations. With few exceptions we found very good agreement between these instruments and MIPAS with no evidence for any bias in most cases and altitude regions. We present a model study of carbon monoxide for 1988-1997 using the GEOS-Chem 3-D model driven by assimilated meteorological data, with time-varying emissions from biomass burning and from fossil fuel and industry, overhead ozone columns, and methane. The hydroxyl radical is calculated interactively using a chemical parameterization to capture chemical feedbacks. We document the inventory for fossil fuels/industry and discuss major uncertainties and the causes of differences with other inventories that give significantly lower emissions. We find that emissions hardly change from 1988 to 1997, as increases in Asia are offset by decreases elsewhere. The model reproduces the 20% decrease …
