Boxwood blight, a fungal disease of ornamental plants (Buxus spp.), is caused by two sister species, Calonectria pseudonaviculata (Cps) and C. henricotiae (Che). Compared to Cps, Che is documented to display reduced sensitivity to fungicides, including the azole class of antifungals, which block synthesis of a key fungal membrane component, ergosterol. A previous study reported an ergosterol biosynthesis gene in Cps, CYP51A, to be a pseudogene, and RNA-Seq data confirm that a functional CYP51A is expressed only in Che. The lack of additional ergosterol biosynthesis genes showing significant differential expression suggests that the functional CYP51A in Che could contribute to …

Biofilm infections are a global public health threat, necessitating new treatment strategies. Biofilm formation also contributes to the development and spread of multidrug-resistant (MDR) bacterial strains. Biofilm-associated chronic infections typically involve colonization by more than one bacterial species. The co-existence of multiple species of bacteria in biofilms exacerbates therapeutic challenges and can render traditional antibiotics ineffective. Polymeric nanoparticles offer alternative antimicrobial approaches to antibiotics, owing to their tunable physico-chemical properties. Here, we report the efficacy of poly(oxanorborneneimide) (PONI)-based antimicrobial polymeric nanoparticles (PNPs) against multi-species bacterial biofilms. Aim Ecological relationships between species richness and biomass production are increasingly thought to be pervasive across the globe. Yet, diversity-production relationships have not been explored extensively for freshwater fish communities even though fisheries production provides key services to humans. Our aim was to evaluate the diversity-production relationship of fish communities inhabiting freshwater streams across the Appalachian Mountain range and examine how diversity-production relationships varied across streams possessing different thermal signatures. Achieving carbon net neutrality at UMass by the year 2032 would require an unprecedented scale of renovation. However, we stand before an opportunity to set a nationwide standard in college campus sustainability. The usage of water also requires energy, as both are inextricable commodities, high in demand and often non-renewable. The central heating plant (CHP) distributes heat across the 1,400+ acre campus and consumes copious amounts of natural gas. The Pioneer Valley is home to a network of food producers, from farmers and cheesemakers to brewers and beekeepers. The objective of today’s work will be to generate wrinkle patterns on very thin polymer films using the forces generated by the surface tension of a water drop. You’ll do a few sizes of water drop and we’ll give you a couple of thicknesses of film. This case study will detail the background and influences for a plastic bag ban in the town of Amherst, outline the steps taken from planning to passage, and address roadblocks and missteps that may be avoided with the implementation of future bans. It is my hope that this document will not only serve as a guide, but also an inspiration for additional local action in Massachusetts and across the country. No matter what the national political climate or attitude towards environmental issues, local action is always possible, and change is usually easier than you think. Think globally, act locally!

African elephants (Loxodonta africana) are imperiled by poaching and habitat loss. Despite global attention to the plight of elephants, their population sizes and trends are uncertain or unknown over much of Africa. To conserve this iconic species, conservationists need timely, accurate data on elephant populations. Here, we report the results of the Great Elephant Census (GEC), the first continent-wide, standardized survey of African savannah elephants. We also provide the first quantitative model of elephant population trends across Africa. We estimated a population of 352,271 savannah elephants on study sites in 18 countries, representing approximately 93% of all savannah …

Background
Low-cost DNA sequencing allows organizations to accumulate massive amounts of genomic data and use that data to answer a diverse range of research questions. Presently, users must search for relevant genomic data using a keyword, accession number of meta-data tag. However, in this search paradigm the form of the query – a text-based string – is mismatched with the form of the target – a genomic profile.
Results
To improve access to massive genomic data resources, we have developed a fast search engine, GEMINI, that uses a genomic profile as a query to search for similar genomic profiles. GEMINI …

This study was conducted by the University of Massachusetts Amherst public policy masters students for Springfield based Wellspring Cooperative, a nonprofit focused on cooperative job creation and training. The project assesses three potential scale options for Wellspring in order to use organic material to heat and/or generate electricity to power its hydroponic greenhouse. Though the greenhouse is not constructed as of yet, its source of energy is an important element for Wellspring. When conducting high-throughput biological experiments, it is often necessary to develop a protocol that is both inexpensive and robust. Standard approaches are either not cost-effective or arrive at an optimized protocol that is sensitive to experimental variations. Here, we describe a novel approach that directly minimizes the cost of the protocol while ensuring the protocol is robust to experimental variation. Our approach uses a risk-averse conditional value-at-risk criterion in a robust parameter design framework. We demonstrate this approach on a polymerase chain reaction protocol and show that our improved protocol is less expensive than the standard protocol and more robust …

The Tan Brook is a small 3.91 square kilometer watershed that runs through and encompasses the highly urbanized areas of Amherst, MA. It is part of the Connecticut River basin and the Mill River sub-basin. In order to meet the high demands of the local middle and high school, and heavy runoff from high amounts of impervious cover from downtown Amherst and the University of Massachusetts, the Tan Brook has been highly culverted.

The Tan Brook Watershed is both a daylighted and diverted underground stream that runs through the Town Center of Amherst and the campus of UMass Amherst. Various stormwater management practices have been used to infiltrate stormwater runoff from streets, lots, buildings and/or vehicles. These systems are custom-designed relative to the surrounding permeability of the soil, vegetation and geographical topography. Some systems have higher success rates than others.

Eastern hemlock (Tsuga canadensis) is in decline because of infestation by the invasive hemlock woolly adelgid (Adelges tsugae; ‘HWA’) and, to a lesser extent, the elongate hemlock scale (Fiorinia externa; ‘EHS’). Many conifers respond to insect herbivory by inducing oleoresin-based defenses, however it is unknown whether eastern hemlock is capable of this inducible response. We conducted a plantation setting study of artificially infested saplings to determine if feeding by HWA or EHS induces changes in the tree’s volatile chemistry. The induced changes in volatiles we found were unlike the terpenoid-based defenses of related conifers. Only HWA feeding …

Predators are key components of aquatic ecosystems and innovative approaches to understanding their spatial distribution are imperative for research, effective management, and conservation. Discontinuities, created by abrupt changes between two unlike entities, are irregularly-distributed, intermediate-scale features that can have a disproportionate effect on organismal distribution within the seascape. Here I use the discontinuity concept to relate the distribution of a predator, striped bass (Morone saxatilis), to physical features within Plum Island Estuary (PIE), MA. Diving Behavior And Identification Of Sex Of Breeding Atlantic Puffins (Fratercula Arctica), And Nest-Site Characteristics Of Alcids On Petit Manan Island, Maine, Sarah M. Spencer

During 2008 – 2009, we quantified foraging behavior of adult Atlantic puffins (Fratercula arctica) by deploying time-depth recorders (TDRs) on 18 adults and collected morphological measurements from 40 adults nesting on Petit Manan Island, Maine. Dive data were successfully retrieved from 5 birds foraging for 14 days in 2008, and 8 birds foraging for 18 days in 2009. Pooling across all birds, a total of 8,097 dives were recorded, with peaks in activity during 0400-0800 and 1600-2000, and no diving between 2100 and 0400. Mean (± SD) dives/bird/day was 276.4 (± 84.7), with dives grouped into bouts lasting …
