A severely damaged 19th-century oil painting depicting a portrait of a woman was treated at Patricia H. and E. Garman Art Conservation Department. A typed letter provided by the owner mentioned that it has been previously restored yet returned with unsatisfactory results. After further examination, the painting appeared to have been previously treated multiple times by different people. There was overpaint distinctly present on the face and later discovered to be present overall. The full state of condition of the painting was initially unknown due to the sum of the surface being overpainted. However, there were evidence of paint loss …

With the ever-increasing threat of antibiotic resistant bacteria, alternative treatment methods have been developed including photodynamic therapy (PDT). Within the PDT process, photosensitizers are used to generate reactive oxygen species (ROS) and facilitate the cell termination process. This work lays the conceptual foundation for the functionalization of the photosensitizer methylene blue (MB) with gold nanoparticles (AuNPs) and INF-55 as a potential inhibitor of the AcrAB-TolC efflux pump to enhance the effectivity of the PDT process.

Rapid docking of flexible biological macromolecules remains a significant open challenge in protein structure determination. While rigid docking is relatively simple with toolkits such as TagDock, a key obstacle to rapid flexible docking is the complexity and roughness of the free energy surface associated with protein conformational motion (often termed the many-minima problem), meaning conventional molecular dynamics methods do not effectively sample protein conformations near the interaction complex in accessible timescales. Ever since the first image of a coral reef was captured in 1885, people worldwide have been accumulating images of coral reefscapes that document the historic conditions of reefs. However, these innumerable reefscape images suffer from perspective distortion, which reduces the apparent size of distant taxa, rendering the images unusable for quantitative analysis of reef conditions. Here we solve this century-long distortion problem by developing a novel computer-vision algorithm, ReScape, which removes the perspective distortion from reefscape images by transforming them into top-down views, making them usable for quantitative analysis of reef conditions. Studying the involvement of nicotinic acetylcholine receptors (nAChRs), specifically α7-nAChRs, in neuropsychiatric brain disorders such as autism spectrum disorder (ASD) has gained a growing interest. The flavonoid apigenin (APG) has been confirmed in its pharmacological action as a positive allosteric modulator of α7-nAChRs. However, there is no research describing the pharmacological potential of APG in ASD. Knowledge about adaptive capacity and its determinants has increased significantly over the last decade. However, most research on adaptive capacity has been static, not considering how adaptive capacity might change over time, particularly after severe disturbances. We studied the adaptive capacity dynamics of Asian-Pacific reef tourism operators affected by coral bleaching and tropical cyclones compared with a control group with non-affected operators. We found that impacts from tropical cyclones were associated with frequent changes in adaptive capacity. Notably, we found a reduction in tangible attributes (assets and flexibility) of adaptive capacity, whereas intangible attributes (agency and social organization) increased. Our …

Coastal mangrove forests provide a suite of environmental services, including sequestration of anthropogenic contamination. Yet, research lags on the environmental fate and potential human health risks of mangrove-sequestered contaminants in the context of mangrove removal for development and range shifts due to climate change. To address this, we conducted a study on Moloka'i, Hawai'i, comparing microplastic and pesticide contamination in coastal compartments both at areas modified by non-native red mangroves (Rhizophora mangle) and unmodified, open coastline. An Ecological Monitoring Program (EMP) has been established at the Virginia Institute of Marine Science Eastern Shore Laboratory (VIMS ESL) for the coastal environment near the Wachapreague lab. Currents in the ocean distribute water masses and everything floating in them. To understand how fast the water (and its flotsam) travels and how effectively it mixes, we need to study the flow statistics. There are two types of measurements of ocean currents: Eulerian measurements that look at how the ocean velocities change at a stationary point on the Earth; and Lagrangian measurements that follow a water mass. Dispersion and mixing is generally best captured by Lagrangian measurements. California's demand-side urban water management policies, such as shifting water pricing structures from non-conservation to conservation-based rates, have received much attention in terms of meeting the state's short- and long-term water conservation policies. This paper quantifies the effect of pricing structure changes on residential water consumption using a survey dataset of 189 major California water utilities from 1994 to 2019. Results of our study demonstrate that residential per capita per day water consumption was reduced by an average of 2.6% when water agencies switched from non-conservation-based to conservation-based pricing structures. Climate change has myriad impacts on ecosystems, but the mechanisms by which it affects individual species can be difficult to pinpoint. One strategy to discover such mechanisms is to identify a specific ecological factor related to survival or reproduction and determine how that factor is affected by climate. Here we used Landsat imagery to calculate water clarity for 127 lakes in northern Wisconsin from 1995 to 2021 and thus investigate the effect of clarity on the body condition of an aquatic visual predator, the common loon (Gavia immer). Sulfur containing compounds including glucosinolates (GLS), sulforaphane (SFN) and S-methyl-L-cysteine sulfoxide (SMCSO) have been proposed to be partly responsible for the beneficial health effects of cruciferous vegetables. As such, greater understanding of their measurements within foods is important to estimate intake in humans and to inform dietary intervention studies. Herein is described a simple and sensitive method for simultaneous analysis of 20 GLS, SFN and SMCSO by liquid chromatography mass spectrometry. Bacterial antibiotic persistence is a phenomenon where bacteria are exposed to an antibiotic and the majority of the population dies while a small subset enters a low metabolic, persistent, state and are able to survive. Once the antibiotic is removed the persistent population can resuscitate and continue growing. Several different molecular mechanisms and pathways have been implicated in this phenomenon. A common mechanism that may underly bacterial antibiotic persistence is perturbations in protein synthesis. The long-run outlook for the corn ethanol industry is questionable, due to a transition to electric and hybrid vehicles. One source of hope for the long run is the potential demand for producing Sustainable Aviation Fuel (SAF). SAF is a key component in the United States Aviation Climate Action Plan, a path to net-zero greenhouse gas (GHG) emissions in the aviation industry by 2050. Demand for ethanol for SAF offers hope to the ethanol industry, but it depends a great deal on policy decisions that are being made now. Here we sketch out this story.

Avena sativa phototropin 1 light-oxygen-voltage 2 domain (AsLOV2) is a model protein of Per-Arnt-Sim (PAS) superfamily, characterized by conformational changes in response to external environmental stimuli. This conformational change begins with the unfolding of the N-terminal A'α helix in the dark state followed by the unfolding of the C-terminal Jα helix. The light state is characterized by the unfolded termini and the subsequent modifications in hydrogen bond patterns. Tuberculosis (TB)-causing bacterium Mycobacterium tuberculosis (Mtb) utilizes mycolic acids for building the mycobacterial cell wall, which is critical in providing defense against external factors and resisting antibiotic action. MmpL3 is a secondary resistance nodulation division transporter that facilitates the coupled transport of mycolic acid precursor into the periplasm using the proton motive force, thus making it an attractive drug target for TB infection. In 2019, X-ray crystal structures of MmpL3 from M. smegmatis were solved with a promising inhibitor SQ109, which showed promise against drug-resistant TB in Phase II clinical trials. Mediterranean forest ecosystems will be increasingly affected by hotter drought and more frequent and severe wildfire events in the future. However, little is known about the longer-term responses of these forests to multiple disturbances and the forests' capacity to maintain ecosystem function. This is particularly so for below-ground organisms, which have received less attention than those above-ground, despite their essential contributions to forest function. Neuroinflammation and brain oxidative stress are recognized as significant contributors to hypertension including salt sensitive hypertension. Extracellular vesicles (EVs) play an essential role in intercellular communication in various situations, including physiological and pathological ones. Based on this evidence, we hypothesized that EVs derived from the brains of hypertensive rats with salt sensitivity could trigger neuroinflammation and oxidative stress during hypertension development. Long-term irrigation management research has been conducted from 2014 to 2023 for corn and soybean at the Eastern Nebraska Research, Extension, and Education Center (ENREEC) Variable Rate Irrigation (VRI) Field located in subhumid east-central Nebraska (in the Lower Platte North Natural Resources District). Tropical forest root characteristics and resource acquisition strategies are underrepresented in vegetation and global models, hampering the prediction of forest–climate feedbacks for these carbon-rich ecosystems. Lowland tropical forests often have globally unique combinations of high taxonomic and functional biodiversity, rainfall seasonality, and strongly weathered infertile soils, giving rise to distinct patterns in root traits and functions compared with higher latitude ecosystems. We provide a roadmap for integrating recent advances in our understanding of tropical forest belowground function into vegetation models, focusing on water and nutrient acquisition.
