Variation In Personality Among Semi-Wild Myanmar Timber Elephants, Sateesh Venkatesh

This study examines two personality traits: exploration and neophobia, which could influence human-elephant conflicts. Thirty-one semi-wild elephants were tested over two trials using a custom novel puzzle tube containing three tasks and three rewards. Our studies show that elephants do vary significantly between individuals in both exploration and neophobia.

A Mechanistic Investigation Of Cytochrome C Nitrite Reductase Catalyzed Reduction Of Nitrite To Ammonia: The Search For Catalytic Intermediates, Shahid Shahid

Cytochrome c Nitrite Reductase (ccNiR) is a periplasmic homodimeric decaheme enzyme that catalyzes the reduction of nitrite to ammonium in a process that involves six electrons and eight protons. Under standard assay conditions, which use a strong reducing agent as an electron source, catalysis takes place rapidly without producing detectable intermediates. However, intermediates do accumulate when weaker reducing agents are employed, allowing the ccNiR mechanism to be studied. Herein, the early stages of Shewanella oneidensis ccNiR-catalyzed nitrite reduction were investigated in isolation by using the weak reducing agents N,N,N’,N’-tetramethyl-p-phenylenediamine (TMPD) and the 2-electron reduced form of indigo trisulfonate. X-ray free-electron lasers (XFELs) open the possibility of obtaining diffraction information from a single biological macromolecule. This is because XFELs can generate extremely intense X-ray pulses which are so short that diffraction data can be collected before the sample is destroyed. By collecting a sufficient number of single-particle diffraction patterns from many tilts of a molecule relative to the X-ray beam, the three-dimensional electron density can be reconstructed ab-initio. The resolution and therefore the information content of the data will ultimately depend largely on the number of patterns collected at the experiment. This paper investigates how the snow-albedo feedback mechanism of the arctic is changing in response to rising climate temperatures. Specifically, the interplay of vegetation and snowmelt, and how these two variables can be correlated. This has the potential to refine climate modelling of the spring transition season. Research was conducted at the ecoregion scale in northern Alaska from 2000 to 2020. Each ecoregion is defined by distinct topographic and ecological conditions, allowing for meaningful contrast between the patterns of spring albedo transition across surface conditions and vegetation types. Aquaponics is an emerging method of agriculture in which fish and plants are grown in an enclosed and recirculating environment. The method mimics a relationship found in nature where fish waste provides nutrients for plants and plants cleanse the water for the benefit of the fish. This symbiotic relationship has proven to be a sustainable method of agriculture in which there is less water use, no need for pesticides or herbicides, recycling of nutrient waste and a smaller spatial footprint. There are roughly 80 ocelots (Leopardus pardalis) remaining in the United States, with the entire population constrained to south Texas, with roadkill being a predominant source of mortality. To prevent additional roadkill and maintain wildlife movement, Texas Department of Transportation constructed 11.9 kilometers of wildlife exclusion fencing, 5 wildlife crossing structures (WCS), and 18 wildlife guards on State Highway 100. This project demonstrates two applications of remote sensing in agricultural and rangeland environments. In the first, an unmanned aerial system (UAS) equipped with a multi-spectral sensor was used to estimate canopy cover across four different cover crop trials at four time periods. In the second, a local database of stationary camera trap images of wildlife was used to train a convolutional neural network to automatically catalogue images by identifying the animal in those images. Genes are segments of DNA that provide a blueprint for cells and organisms to effectively control processes and regulations within individuals. There have been many attempts to quantify these processes, as a greater understanding of how genes operate could have large impacts on both personalized and precision medicine. Roads can greatly impact wildlife. Wildlife vehicle collisions contribute to population declines while disturbance from human activity may prevent wildlife from using areas around roads. The construction of mitigation structures may lessen these effects through fine scale modification of animal use of roadside areas. In this thesis, how the construction of wildlife mitigation structures impacted the fine scale distribution of wildlife on State Highway 100 in Cameron County, Texas was examined. The multiple experiments conducted in an Avicennia germinans forest in Laguna Vista, TX was focused on determining if facilitation theory could explain the behavior observed between the herbaceous halophyte Batis maritima and A. germinans. I hypnotized that a positive interaction exists but through which mechanisms of facilitation I did not know. The overarching theme of the study is the presence or absence of B. maritima. Facilitation theory tells us that positive interactions are most often found under stressful conditions such as those on the coastal tidal flat of the Laguna Madre. Gold nanoparticles (AuNPs) have been exploited in the various domains of science such as drug delivery, bio-sensing, immunoassays and environmental sensors, due to their optical properties and intriguing surface chemistry. Different scientific procedures have been used to effectively immobilize antibodies onto AuNPs. Although acceptable outcomes have been achieved in the immobilization of antibodies onto AuNPs, the sensitivity of these immobilized antibodies to target antigen or binding sites is limited due to improper orientation of the antibodies. Also, the possibility of nanoparticle aggregation when exposed to proteins limits its biomedical applicability.

Polychlorinated biphenyls (PCBs) are a group of persistent organic pollutants known to contribute to several adverse health conditions in humans including cancers and a suite of liver diseases. While there exist 209 unique PCB congeners, the World Health Organization has identified 12 that pose the greatest health risk to humans due to these congeners’ functional similarity to dioxins, another notoriously toxic class of contaminants. Cadmium ion causes toxicity in humans, most prominently in the kidney. This thesis focuses on mechanisms by which Cd2+ harms kidney proximal tubule cells. Previous experiments have shown the time-dependent distribution of Cd2+ and Zn2+ within supernatant of pig kidney, LLC-PK1 cells after exposure to Cd2+ plus the ionophore pyrithione1. The first part of this research describes the trafficking of cadmium within LLC-PK1 proximal tubule cells, including the time dependent distribution, speciation, and quantification of Cd2+ and Zn2+ in cells exposed to Cd2+ and Cd2+ plus pyrithione. Roads are known to negatively impact wildlife by fragmenting habitat and mortality caused by wildlife-vehicle collisions. Road mitigation structures, such as wildlife crossing structures (WCS), wildlife guards (WG), and fencing are commonly used to address the issue of roads. In South Texas, such structures were built or modified along State Highway (SH) 100 in Cameron County as an effort to conserve the endangered ocelot (Leopardus pardalis).
