Vernal pools are small seasonal wetlands that are a common landscape feature that contribute to biodiversity in northeastern North American forests. However, even basic information about their biogeochemical functions, such as carbon cycling, is limited. Dissolved gas concentrations (CH4, CO2) and other water chemistry parameters were monitored weekly at the bottom and surface of four vernal pools in central and eastern Maine, USA, from April to August 2016. Introduction: Differential scanning calorimetry (DSC) is used to determine thermally-induced conformational changes of biomolecules within a blood plasma sample. Recent research has indicated that DSC curves (or thermograms) may have different characteristics based on disease status and, thus, may be useful as a monitoring and diagnostic tool for some diseases. Since thermograms are curves measured over a range of temperature values, they are often considered as functional data. Previous work in Integro-Difference models have generally considered Allee effects and over-compensation separately, and have either focused on bounded domain problems or asymptotic spreading results. Some recent results by Sullivan et al. (2017 PNAS 114(19), 5053-5058) combining Allee and over-compensation in an Integro-Difference framework have shown chaotic fluctuating spreading speeds. In this thesis, using a tractable parameterized growth function, we analytically demonstrate that when Allee and over-compensation are present solutions which persist but essentially remain in a compact domain exist. Although black bears (Ursus americanus, Ursus americanus luteolus) were once found throughout the south-central United States, unregulated harvest and habitat loss resulted in severe range retractions and by the beginning of the twentieth century populations in Oklahoma, Louisiana, Texas and Arkansas were nearing extirpation. In response to these losses, translocation programs were initiated in Arkansas (1958-1968 & 2000-2006) and Louisiana (1964-1967 & 2001-2009). These programs successfully restored bears to portions of Louisiana and Arkansas, and, as populations in Arkansas began dispersing, to Oklahoma. In contrast, east Texas remains unoccupied despite the existence of suitable habitat in the region.

Anadromous sea lamprey Petromyzon marinus are native to Atlantic coastal systems and serve as a functional link between marine and freshwater ecosystems. Sea lamprey spend 1–2 years in the ocean parasitizing marine vertebrates before migrating into freshwaters during the spring to spawn. There they construct nests, spawn, then die shortly afterwards. Larvae hatch, bury into fine sediments and reside in streams for generally 6–8 years, but up to 14. Larvae then undergo metamorphosis, a non-feeding period characterized by a series of physical and physiological changes. The juveniles (macropthalmia) then migrate to the ocean to begin the parasitic juvenile phase.

The coexistence of species is probably one of the most interesting and complex phenomenon in nature. We constructed an agent based model to study the coexistence dynamics of prey - predator populations by varying productivity levels of producers in fragmented and connected habitats along with different levels of quality of predators. Our results indicated that productivity levels of producers in fragmented and connected habitats along with levels of predator quality are significantly responsible for overall predator - prey population size and survivorship. Visual Acuity (VA) examinations are one of the most commonly conducted medical assessment throughout the world. Recent advances in computer technology allows for new forms of visual assessment to be conducted. In Part I of this thesis I demonstrate the capability of an automated computer program named VISION to assess human visual acuities. Different color combinations of an object against a background emitted from a computer screen are used to examine a variety of human color vision acuities. Protein identification using tandem mass spectrometry (MS/MS) has proven to be an effective way to identify proteins in a biological sample. An observed spectrum is constructed from the data produced by the tandem mass spectrometer. A protein can be identified if the observed spectrum aligns with the theoretical spectrum. However, data generated by the tandem mass spectrometer are affected by errors thus making protein identification challenging in the field of proteomics. Lilium grayi (Gray’s Lily), a southern Appalachian endemic species, is threatened by a Lilium-specific fungal pathogen, Pseudocercosporella inconspicua. The disease is characterized by tan lesions that can cause early senescence, while also lowering seed production and viability. This project tested for P. inconspicua conidia and accessed health at nine locations. The disease was present and ubiquitous across the range of L. grayi. Through identification of P. inconspicua conidia in the field, L. superbum (Turk’s Cap Lily) was identified as an additional host, while L. michauxii (Michaux’s Lily) was disease-free. Despite being separated by millions of years of evolution, black bears (Ursus americanus) and brown bears (Ursus arctos) can be difficult to distinguish based on skeletal and dental material alone. Complicating matters, some Late Pleistocene U. americanus are significantly larger in size than their modern relatives, obscuring the identification of the two bears. In the past, fossil bears have been identified based on differences in dental morphology or size. Fossil Agamidae from Western Australia have been the subject of limited study. To aid in fossil agamid identification, Hocknull (2002) examined the maxilla and dentary of several extant species from Australia and determined diagnostic characters for various species groups. In the study here, fossil agamids from two localities in Western Australia, Hastings Cave and Horseshoe Cave, were examined, grouped, and identified to the lowest unambiguous taxonomic level. Morphometric analyses were conducted to compare morphotypes, and find additional diagnostic characters. From Hastings Cave there were two maxilla morphotypes and three dentary morphotypes. From 2015-2017, we evaluated a newly established bighorn sheep (Ovis canadensis) herd in the Deadwood Region of the Black Hills, South Dakota. Our objectives were to 1a) determine annual survival rates, 1b) determine cause-specific mortality, 1c) estimate population size, 2a) assess genetic diversity, 2b) assess disease prevalence, 3) evaluate movement patterns post-release, 4a) evaluate 3rd-order habitat selection, and 4b) estimate herbaceous biomass at foraging sites post-release of translocated bighorn sheep. Irrigation water is scarce. Hence, accurate estimation of crop water use is necessary for proper irrigation managements and water conservation. Satellite-based remote sensing is a tool that can estimate crop water use efficiently. Animals that co-occur in sympatry with multiple closely-related species use reproductive mate attraction signals not only to assess the quality of a potential conspecific mate (sexual selection), but also to discriminate conspecifics from heterospecifics (species recognition). However, the extent to which sexual selection and species recognition may interact, or even conflict, is poorly known. Neotropical electric fish offer unrivaled opportunities for understanding this problem. Recent work indicates that the Y-chromosome of the fruit fly Drosophila melanogaster can influence gene regulation on the autosomes and X chromosome. This newly discovered function of the Y has the potential to dramatically shape the regulatory evolution of numerous genes that reside throughout the genome; even for genes that code for both male and female traits. Mycofactocin is a putative peptide-derived redox cofactor in Mycobacterium family. Its putative biosynthetic pathway is encoded by the operon mftABCDEF. The initial step of this pathway is a posttranslational modification of a peptide precursor MftA, which is catalyzed by MftC enzyme. This modification only occurs in the presence of chaperone MftB. Here, we demonstrate that MftC is a radical S-adenosyl L-methionine (SAM) enzyme and we examine its catalytic mechanism. Breeding Bird Survey (BBS) data for grassland bird species has shown the most rapid population decline of any other bird group. Current roadside survey techniques, however, may fall short of providing accurate numbers of rare grassland bird species such as chestnut-collared longspur (Calcarius ornatus), lark bunting (Calamospiza melanocorys), Sprague’s pipit (Anthus spragueii), and Baird’s sparrow (Ammodramus bairdii). Trends resulting from roadside data for grassland birds are oftentimes determined to be statistically insignificant because many grassland bird species occur on too few routes, occur in low numbers per route, and show high annual fluctuations in number. Swift foxes (Vulpes velox) were reintroduced into Badlands National Park between 2003 and 2006 after being nearly extirpated from South Dakota in the early 1900’s. Genetic analysis provided strong evidence that the reintroduction was successful, but viability analysis indicated the population may be in jeopardy with a high probability of extinction. Recently, the population has declined due to various biotic and abiotic factors (e.g., recent weather patterns, effects of plague [Yersinia pestis], and increased coyote [Canis latrans] numbers). Landscape homogenization and the degradation of riparian areas has greatly impaired stream ecosystems throughout North America. Conservation programs may repair riparian ecosystems to indirectly improve water quality and instream habitat heterogeneity in hopes to elicit biological responses. However, focused manipulations on isolated stream fragments have rarely achieved biological goals. Prairie streams with restored riparian areas were appraised (chemical, physical, and biological variables) to quantify the indirect effects of prevalent grassland conservation practices on aquatic resources. The northern Great Plains provide critical breeding habitat for waterfowl and gamebirds in the United States. Peak commodity prices in the late 2000s resulted in increased agricultural production and large-scale conversions of grassland habitat to monoculture row crops. However, recent declines in commodity prices have created a renewed interest for private landowners to diversify crops and enroll in government subsidized conservation programs that convert idle grassland and unproductive cropland to wildlife friendly perennial grassland plantings.
