Over-complete bases offer the flexibility to represent much wider range of signals with more elementary basis atoms than signal dimension. The use of over-complete dictionaries for sparse representation has been a new trend recently and has increasingly become recognized as providing high performance for applications such as denoise, image super-resolution, inpaiting, compression, blind source separation and linear unmixing. This dissertation studies the dictionary learning for single or coupled feature spaces and its application in image restoration tasks. A Bayesian strategy using a beta process prior is applied to solve both problems.

Unique microfluidic control actuated by simply turning off and on microfabricated electrodes in a small-volume system was investigated for lab-on-a-chip applications. This was accomplished using a relatively new pumping technique of redox-magnetohydrodynamics (MHD), which as shown in this dissertation generated the important microfluidic features of flat flow profile and fluid circulation. MHD is driven by the body force, FB = j × B, which is the magnetic part of the Lorentz force equation, and its direction is given by the right hand rule. The aftermath of a natural disaster is typically characterized by lack of a reliable medium for dissemination of information to survivors. Current state-of-the-art emergency response systems rely on satellite radio-enabled devices, but survivors, unlike first responders, do not have access to such devices. To mitigate this problem, we present PERPETUU a solar-powered portable GIS microserver. The microserver node can be deployed in a disaster scene, and can serve maps to survivors viewable on browsers of off-the-shelf mobile systems. Water-soluble Quantum Dots (QDs) are highly sensitive fluorescent probes that are often used to study biological species. One of the most common ways to render QDs water-soluble for such applications is to apply hydrophilic thiolated ligands to the QD surface. However, these ligands are labile and can be easily exchanged on the QD surface, which can severely limit their application. As one way to overcome this limitation while maintaining a small colloidal size of QDs, we developed a method to stabilize hydrophilic thiolated ligands on the surface of QDs through the formation of a crosslinked shell using a photocrosslinking approach. …

Optical pyrometry is the sensing of thermal radiation emitted from an object using a photoconductive device to convert photons into electrons, and is an important diagnostic tool in shock physics experiments. Data obtained from an optical pyrometer can be used to generate a blackbody curve of the material prior to and after being shocked by a high speed projectile. The sensing element consists of an InGaAs photodiode array, biasing circuitry, and multiple transimpedance amplifiers to boost the weak photocurrent from the noisy dark current into a signal that can eventually be digitized. Hence, image denoising is an essential processing step preceding visual and automated analyses. The kinematics and dynamics for plane wave optics are derived for a massive electrodynamic field by utilizing Proca's theory. Atomic spectroscopy is also examined, with the focus on the 21 cm radiation due to the hyperfine structure of hydrogen. The modifications to Snell's Law, the Fresnel formulas, and the 21 cm radiation are shown to reduce to the familiar expressions in the limit of zero photon mass.

Despite tremendous scientific achievements, cancer remains the second leading cause of death in the United States. Metabolic reprogramming has been increasingly recognized as a core hallmark of cancer. My dissertation work identified novel diagnostic markers and therapeutic targets for human cancers through the study of cholesterol in cancer cells.
Enabled by label-free Raman spectromicroscopy, we performed the first quantitative analysis of lipogenesis at single cell level in human patient cancerous tissues. As applications evolve, they continue to operate over a large monolithic framework of networking and computing equipment built from general-purpose microprocessors and Application Specific Integrated Circuits (ASICs) that offers few architectural choices. This dissertation presents techniques to diversify the next-generation Internet infrastructure by integrating Field-programmable Gate Arrays (FPGAs), a class of reconfigurable integrated circuits, with general-purpose microprocessor-based techniques. Impact pile driving can produce extremely high underwater sound levels, which are of increasing environmental concern due to their deleterious effects on marine wildlife. Prediction of underwater sound levels is important to the assessment and mitigation of the environmental impacts caused by pile driving. Current prediction methods are limited and do not account for the dynamic pile driving source, inhomogeneities in bathymetry and sediment, or physics-based sound wave propagation.

A DC microgrid is selected for the efficiency benefits, lack of reactive power in the system, and ease of connecting to an AC grid. In this dissertation, the relationship between the geometry of ion-beam sculpted solid-state nanopores and their ability to analyze single DNA molecules using resistive pulse sensing is investigated. To accomplish this, the three dimensional shape of the nanopore is determined using energy filtered and tomographic transmission electron microscopy. It is shown that this information enables the prediction of the ionic current passing through a voltage biased nanopore and improves the prediction of the magnitude of current drop signals when the nanopore interacts with single DNA molecules. In this work, computational investigation of plasmonic nanostructures was conducted using the commercial finite element electromagnetics solver Ansys® HFSS. Arrays of silver toroid nanoparticles located on the surface of an amorphous silicon thin-film absorbing layer were studied for particle sizes ranging from 20 nm to 200 nm in outer diameter. Parametric optimization by calculating an approximation of the photocurrent enhancement due to the nanoparticles was performed to determine optimal surface coverage of the nanoparticles.
