Changes In Enzyme Structural Dynamics Studied By Hydrogen Exchange-Mass Spectrometry: Ligand Binding Effects Or Catalytically Relevant Motions?, Courtney S Fast, Siavash Vahidi, Lars Konermann

Chemistry Publications

It is believed that enzyme catalysis is facilitated by conformational dynamics of the protein scaffold that surrounds the active site, yet the exact nature of catalytically relevant protein motions remains largely unknown. Hydrogen/deuterium exchange (HDX) mass spectrometry (MS) reports on backbone H-bond fluctuations. HDX/MS thus represents a promising avenue for probing the relationship between enzyme dynamics and catalysis. A seemingly straightforward strategy for such studies involves comparative measurements during substrate turnover and in the resting state. We examined the feasibility of this approach using rabbit muscle pyruvate kinase (rM1-PK) which catalyzes the conversion of phosphoenolpyruvate and Mg-ADP to pyruvate and …

Development Of Pre-Clinical Assays Based On Tandem Mass Spectrometry To Investigate Gabaa Receptor Modulators, Margaret Laurel Guthrie Dec 2017

Drugs are compounds that interact with a biological system to produce a specific biological response. The goal of drug discovery is to design and synthesize a pharmaceutical agent that will produce a desired biological effect. However, the in-vivo effects of a drug molecule cannot be predicted without a variety of test results prior to human clinical trials. In order to designate any drug compound as a …

Fatty Acid Biomarker Detection For Breast Cancer Using Differential Mobility Spectrometry With Non-Radioactive Ion Source, James Joseph Alberti Mar 2017

Fatty Acid Biomarker Detection For Breast Cancer Using Differential Mobility Spectrometry With Non-Radioactive Ion Source, James Joseph Alberti

USF Tampa Graduate Theses and Dissertations

Differential Mobility Spectrometry (DMS) using a non-radioactive ion source (NRIS) is investigated as a possible medical diagnostic instrument for near real-time detection of breast cancer biomarkers. In previous clinical studies, concentrations of Linoleic, Palmitic and Stearic fatty acids have been observed at different levels in women with carcinoma breast cancer versus women with benign tumors or healthy women showing no signs of breast cancer. Present diagnostic methods require a biopsy of the suspect tissue and a microscopic lab analysis performed to determine its disease state. This process can take hours or days before the patient and doctor are informed of …

Mechanisms Of Heterogeneous Oxidations At Model Aerosol Interfaces By Ozone And Hydroxyl Radicals, Elizabeth A. Pillar-Little Jan 2017

Mechanisms Of Heterogeneous Oxidations At Model Aerosol Interfaces By Ozone And Hydroxyl Radicals, Elizabeth A. Pillar-Little

Theses and Dissertations--Chemistry

Atmospheric aerosols play an important role in climate by scattering and absorbing radiation and by serving as cloud condensation nuclei. An aerosol’s optical or nucleation properties are driven by its chemical composition. Chemical aging of aerosols by atmospheric oxidants, such as ozone, alters the physiochemical properties of aerosol to become more hygroscopic, light absorbing, and viscous during transport. However the mechanism of these transformations is poorly understood. While ozone is a protective and beneficial atmospheric gas in the stratosphere, it is a potent greenhouse gas in the troposphere that traps heat near the Earth’s surface. It also impacts human heath …

Rapid Quantification Of Trypsin Inhibitors In Food And Feed Formulations With Electrospray Ionization Mass Spectrometry, Radheshyam Panta Jan 2017

Rapid Quantification Of Trypsin Inhibitors In Food And Feed Formulations With Electrospray Ionization Mass Spectrometry, Radheshyam Panta

Doctoral Dissertations

"Presence of anti- nutrients deteriorate the nutritional value of animal foods and feed formulations. The important one being trypsin inhibitors. Trypsin inhibitors in food and feed formulations lower the protein supplement and hence the ultimate consequence is the decrease in growth and development of animals. It is therefore, crucial to monitor trypsin inhibitors in animal food and feed formulations. Research work mentioned in this dissertation is based on the development of rapid, more accurate and robust method for determination of trypsin inhibitor in food and feed formulations. The developed method includes extraction of trypsin inhibitor from food and feed formulation …
