Characterization Of Shinella 6-Hydroxy-(L)-Nicotine Oxidase (6hlno-Se), Kaitrin Funckes

Honors Theses

6-hydroxy-(L)-nicotine oxidase, isolated from the organism Shinella sp. HZN7 (6HLNO-SE), is an enzyme that during the catabolism of nicotine catalyzes the oxidation of (S)-6-hydroxynicotine to 6-hydroxypseudeooxynicotine. This enzyme is a member of the monoamine oxidase (MAO) structural family of flavoproteins, which contain a highly conserved FAD-binding domain. Within the binding domain is a conserved lysine residue that can be traced throughout evolutionary history in this family. 6HLNO isolated from Arthrobacter nicotinovorans serves as a model enzyme for the one isolated from Shinella as both catalyze the same reaction in this biochemical pathway and much has been studied about the Arthrobacter …

Optimization Of Cation Exchange Chromatography Purification Of A Small Peptide: An Industrial And Practical Approach, Jamin Willoughby Oct 2020

Optimization Of Cation Exchange Chromatography Purification Of A Small Peptide: An Industrial And Practical Approach, Jamin Willoughby

Honors Theses

Ion exchange chromatography (IEX) is a common chemical separation technique that is used to separate ionizable target molecules from a liquid mixture mobile phase by adsorption of the molecule to a stationary phase. In chromatography, the stationary phase is a porous solid that the target molecule and mobile phase flows through, and is commonly referred to as the resin. Resins are commonly packed into hollow cylindrical columns. The mobile phase is the liquid solvent which flows through the column containing the stationary phase. The target molecule will have an affinity for the resin and adsorb to it, while impurities in …

Biological Evaluation Of Fda-Approved Drugs As Mirna-31 Inhibitors Using Real Time Qrt-Pcr And A Luciferase Assay, Nicholas Sienkiewicz

Honors Theses

MicroRNAs (miRNAs) are a group of post-transcriptional, negative regulators of gene expression, consisting of ~19-24 nucleotides. Since their discovery in 1993, miRNAs have been found to be involved in a diverse amount of cellular and disease pathways, including certain types of cancer. Overexpression of certain oncogenic miRNAs, including miRNA-31, have been shown to play a role in cancer cell development. Recent findings have suggested that miRNAs play a significant role in every stage of colorectal cancer (CRC) initiation, progression, and development, as well as induce resistance to chemotherapy drugs like 5-FU. In particular, research has shown that increased levels of …

Best Organic Chemistry Practices In The Midwest, Claire Van Der Bosch Apr 2019

Best Organic Chemistry Practices In The Midwest, Claire Van Der Bosch

Honors Theses

Organic Chemistry is a gateway course to a handful of courses in higher chemical education. Unfortunately, it is notorious for its high-fail, high-withdrawal, and high-drop rate and has a reputation for being time-consuming, with difficult concepts that are oftentimes new to students. It is often taught in a two-semester sequence, with the courses named Organic I and Organic II, and both courses have a lab component. At Western Michigan University, the fail rate for these two courses are 53.5% and 36.9%, respectively. Most higher education institutions are certified with the American Chemical Society (ACS) and given a skeleton of a …

The Effects Of Climate Change And Forest Disturbances On Terpene Production Of White Pine (Pinus Strobus) And Red Pine (Pinus Resinosa) In Northern Michigan, Troy Dehagen Apr 2019

The Effects Of Climate Change And Forest Disturbances On Terpene Production Of White Pine (Pinus Strobus) And Red Pine (Pinus Resinosa) In Northern Michigan, Troy Dehagen

Honors Theses

The secondary impacts that climate change may have on the interactions between forests and the atmosphere through forest disturbances is of increasing interest as the climate continues to change. Anthropogenic climate change, which is a direct result of coal and fossil fuel emissions, is likely to impact forest ecology through increased temperatures and extreme precipitation events, thus increasing the frequency and severity of climate-induced forest disturbances. Forest disturbances may have a secondary impact on the production and emission of biogenic volatile organic compounds (BVOC) within forests. In the presence of oxides of nitrogen (NOx) produced from fossil fuel …

Extraction Studies Of Toxic Multiply Charged Oxoanions From Aqueous Media Using Nanojars, Hartman Christian Apr 2017

Extraction Studies Of Toxic Multiply Charged Oxoanions From Aqueous Media Using Nanojars, Hartman Christian

Honors Theses

In this paper, the potential of nanojars as extracting agents of chromate and arsenate from aqueous media into a non-polar organic medium is investigated. Nanojars are a class of compounds that consist of multiple [cis-CuII(μ-OH)(μ-pz)]n cyclic polymerization isomers (n = 6, 8, 9, 10, 12, 14; pz = pyrazolato anion) that form from self-assembly reactions around a multiply-charged oxoanion core with capping counterions to render an overall neutral complex. This incarcerates the anion, preventing it from escaping the complex without the use of any covalent bonding, making it a potentially highly effective and unique extracting agent. We have concluded that …

Effects Of Creatine, Leucine, And Ethanol On Glial Cell Line-Derived Neurotrophic Factor In C2c12 Skeletal Muscle Cells, Daniel Waters Apr 2016

Effects Of Creatine, Leucine, And Ethanol On Glial Cell Line-Derived Neurotrophic Factor In C2c12 Skeletal Muscle Cells, Daniel Waters

Honors Theses

Glial Cell Line-Derived Neurotrophic Factor (GDNF) is a protein expressed throughout the mammalian nervous system that serves several protective functions. Originally discovered in the brain, where it supports the survival of dopaminergic neurons, GDNF has since been shown to be expressed in skeletal muscle and is vital for the maintenance of motor neurons. The relationship between muscle hypertrophy, and GDNF is not well understood. The purpose of this study was to examine the relationship between hypertrophy and GDNF by inducing hypertrophy in C2C12 skeletal muscle cells using leucine and creatine, inhibiting hypertrophy using ethanol and measuring GDNF content. As measured …

Functional Variants Of Metal-Binding Domains 5 And 6 Of The Human Wilson Protein, Sandra Weitzner Mar 2014

Functional Variants Of Metal-Binding Domains 5 And 6 Of The Human Wilson Protein, Sandra Weitzner

Honors Theses

Wilson's disease is caused by mutations in atp7b, a gene that encodes a copper-transporting P1B-type ATPase, otherwise known as the Wilson protein. Over 300 different mutations cause Wilson's disease, and we focused on missense mutations occurring in the 650 amino acid N-terminus of the Wilson protein. This portion of the protein includes six metal-binding domains that function to azquire Cu(I) from the cytosol, prior to passing it through the ATP-driven membrane-spanning pump. Of particular interest are metal-binding domains 5 and 6 (WLN5-6) since we know that they are absolutely required for function of the enzyme. Several missense mutations …

Investigation Of Saccharide Release Through Acidification, Sonication, Thermal, And Microwave Treatments, William K. Storck Apr 2012

Investigation Of Saccharide Release Through Acidification, Sonication, Thermal, And Microwave Treatments, William K. Storck

Honors Theses

With diminishing fossil fuels, renewable sources are becoming much higher in demand. Algae are becoming an increasingly important source of biofuels. Production of biofuels from algae exceeds yields from every other source, most notably corn, per acre. It also provides a highly viable method of reusing carbon dioxide and does not compete with other crops for growth. The production of ethanol through fermentation requires the release of saccharides produced by the algae. The more saccharides extracted, the more ethanol can be produced. This project investigates for a more effective method of maximum saccharide release.

The purpose of this project is …

Interaction Of Pharmaceutical Compounds With Dissolved Organic Matter, Carlen Nigl Smith Apr 2012

Interaction Of Pharmaceutical Compounds With Dissolved Organic Matter, Carlen Nigl Smith

Honors Theses

Highly prescribed pharmaceutical compounds and their metabolites used to treat conditions ranging from infections to depression are of environmental concern as these excreted compounds are increasingly appearing in wastewater influent. As pharmaceuticals enter treatment plants they also can interact with dissolved organic matter (DOM) present. This potential interaction between pharmaceutical compounds and DOM may inhibit the advanced oxidation process (AOP) based removal of drugs during water treatment. The focus of this research was to utilize multinuclear magnetic resonance and hydroxyl radical kinetics to assess whether an interaction between influent pharmaceuticals and DOM does occur, the extent of the interaction, and …

Stability Testing Of Mint Oils Through Gas Chromatography Analysis, Sarah Wolf May 2007

Stability Testing Of Mint Oils Through Gas Chromatography Analysis, Sarah Wolf

Honors Theses

In this experiment, the stability of mint oil was measured through gas chromatography analysis. Eight different oils were tested. Levels of key components of mint oil, including menthol and menthone for peppermint oil, and carvone and limonene for spearmint oil, was compared over time. Several packaging materials were immersed in oils in order to determine their effects, if any, on the composition of the oils. Additionally, oil was subjected to either 25 degree or 40 degree Celsius temperatures to determine if temperature affected the oil composition.

A Method For The Determination Of Purity Of Pnu-10483 Bulk Drug By Capillary Zone Electrophoresis, Gavin Gibson Apr 1998

A Method For The Determination Of Purity Of Pnu-10483 Bulk Drug By Capillary Zone Electrophoresis, Gavin Gibson

Honors Theses

In this investigation, a method was developed for the determination of purity of the drug PNU-10483 using the technique of capillary zone electrophoresis (CZE). CZE was seen as a potential alternative to the formerly used procedure of ion-pair reversed phase highperformance liquid chromatography (HPLC). Nearly five years earlier, a similar study was performed, in which an attempt was made to develop a method that used CZE to determine the purity of bulk drug and the potency of drug tablets. While this development was mostly successful, sporadically unpredictable results reduced the method's overall viability. These outliers were thought to be caused …

Nickel Catalyzed Cyclizations Of Alkynyl-Etones And Their Application To The Formal Synthesis Of Pentalenic Acid, Kevin Schlosser Dec 1996

Nickel Catalyzed Cyclizations Of Alkynyl-Etones And Their Application To The Formal Synthesis Of Pentalenic Acid, Kevin Schlosser

Honors Theses

Pentalenic acid is a very important molecule because it is a biosynthetic intermediate of pentalenolactone, which has antibiotic effects on Grampositive/ negative bacteria and pathogenic fungi. These effects are due to its ability to inhibit glyceraldehyde 3-phosphate dehydrogenase.1 However, the total synthesis of pentalenic acid is very lengthy.2 This project will begin the process of finding a more convenient route to pentalenic acid. Previously in this laboratory there have been experiments conducted in the cyclization of enones with nickel (0)3 It is hypothesized that this method can be used to shorten the preparation of pentalenic acid. As illustrated by the …

Sds Non-Acrylamide Polymeric Gel-Filled Capillary Electrophoresis For Molecular Size Separation Of Protein, Devon Andres

Honors Theses

Sodium dodecyl sulfide (SDS) non-acrylamide gel-filled capillary columns are a new technology being used for analysis and separation of biotechnology-derived proteins. This research was to compare this new technology to the current methods of SDS polyacrylamide gel electrophoresis (SDS-PAGE) and high-performance size-exclusion chromatography (HPSEC). The molecular mass of four different recombinant proteins were determined by two commercialized SDS non-acrylamide gel-filled capillary columns, SDS-PAGE, and HPSEC. The data obtained showed that the SDS non-acrylamide gel-filled capillary columns were compatible with the SDS-PAGE technique for molecular mass determination. HPSEC was shown to be unreliable for molecular weight determination. SDS non-acrylamide gel-filled capillary …

Determination Of The Rate Of Formation Of Hcn, Mitchell Hull Nov 1977

Determination Of The Rate Of Formation Of Hcn, Mitchell Hull

Honors Theses

Nitric oxide is a major air pollutant arising from fossil fuel combustion, yet the fuel contribution to its formulation is uncertain. Since, in combustion, fossil fuels undergo disintegration before oxidation takes place, the study of the pyrolsis of a model constituent of fossil fuels can shed light on NO formation. The rate of inert pyrolytic disappearance of pyridine and the rate of formation of the principal Nitrogen-containing pyrolytic product, HCN, was studied in an inert atmosphere. The rate constant for the formation of HCN and its temperature dependence were determined.
