The increasing prevalence of drug-resistant pathogens is an urgent problem that requires novel methods of bacterial control. Plant extracts inhibit bacterial pathogens and could contain antibacterial compounds with novel mechanisms of action. Yerba mate, a common South American beverage made from Ilex paraguariensis, has antibiotic activity against a broad range of bacterial pathogens. In this work, an attempt was first made to characterize the antibacterial source of an aqueous yerba mate extract by generating a series of extract fractions, collecting GC-MS and antibacterial activity profiles, and then ranking the hundreds of compounds by their presence in fractions with high antibacterial …

This work studies a variety of molecules and the systems they are involved in to further understand disease and treatment of diabetes mellitus, malaria, and castrate resistant prostate cancer. The first system studied, diabetes mellitus, involves synthesis of 5 different steroidal compounds, while the other two systems, malaria and castrate-resistant prostate cancer, apply mass spectrometric-based untargeted metabolomics to study both known metabolites and unknown spectral features.

Diabetes mellitus is a result of the dysfunction and death of islet beta cells caused by the transcription of pro-inflammatory cytokines resulting in reduced insulin secretion and uncontrollable blood glucose levels. Glucocorticoids can reduce …

Metabolomics aims to identify and quantify all of the small molecules within a cell. The relative concentrations of metabolites within cells grown under different experimental conditions, or different species, can be compared using pool size data. Exposing cellular cultures to isotopically labeled compounds allows the progress of metabolites through metabolic pathways to be tracked, known as flux. In the first experiment, the metabolic effects of exposure to lower than optimal temperatures in wild type Salmonella enterica, i.e. cold shock, were determined. Quorum sensing (QS) is a phenomenon that allows bacteria to communicate with each other. Small molecules known as autoinducers are synthesized and released by bacteria, and once enough members of the community are around to ensure survival, i.e. quorum, a phenotype, e.g. bioluminescence, is expressed. There are two types of QS molecules, intra- and inter-species.

Two biological systems were studied using LC/ESI/MS/MS on a triple quadrupole operated in SRM (selected reaction monitoring) scan mode. The first bacterium system is aquatic and microscopic in size known as Roseobacter. The second mammalian system is terrestrial and large in size relative to humans known as Holstein cows.

Roseobacter is a clade of marine bacteria abundant in the ocean. Roseophages are viruses that infect Roseobacter and cause viral lysis. Liquid chromatography tandem mass spectrometry has become a powerful tool for investigating biological systems. Herein we describe the development of both isotope dilution mass spectrometry methods and targeted metabolomics methods for the study of metabolic and cell-cell signaling applications.

A putative yeast enzyme was characterized by discovery metabolite profiling, kinetic flux profiling, transcriptomics and structural biology. These experiments demonstrated that the enzyme shb17 was a sedoheptulose bisphosphatase that provides a thermodynamically dedicated step towards riboneogenesis, leading to the redefinition of the canonical pentose phosphate pathway.

Many mixtures important to research consist of hundreds or even thousands of individual components of interest. These types of mixtures are far too complex to separate by a single chromatographic dimension in any reasonable amount of time. However, if a multidimensional approach is used, where a complex mixture is separated by an initial dimension, simpler fractions of that separation are collected and each of those fractions are analyzed individually, highly complex mixtures can be resolved in relatively short amounts of time.
