Volatile Profiles And Resistance To Herbivory In Eastern Hemlock, Elizabeth A. Mckenzie Nov 2014

Volatile Profiles And Resistance To Herbivory In Eastern Hemlock, Elizabeth A. Mckenzie

Masters Theses

Eastern hemlock hosts the hemlock woolly adelgid, an introduced sap-feeding insect that causes rapid deterioration of the host. Like most conifers, eastern hemlock produces a variety of constitutive and induced defenses, primarily terpenoids. To explore the relationship of terpenoid defenses with adelgid infestations, we artificially infested hemlocks at a forest site and a plantation site, and compared their terpenoid concentrations to those in control trees. Infested trees showed lower terpenoid concentrations than control trees, suggesting that eastern hemlock not only fails to induce production of terpenoids in response to adelgid infestation, but becomes less able to produce carbon-based defenses due …

Modification Of Gold Nanoparticles For Sers Application In Emulsion And Lipid Systems, Michael J. Driver Nov 2014

Modification Of Gold Nanoparticles For Sers Application In Emulsion And Lipid Systems, Michael J. Driver

Masters Theses

Gold nanoparticles produced using the Turkevich method were able to have their hydrophobicity modified using octanethiol in a novel method for SERS application. Both amphiliphic GNPs and hydrophobic GNPs were produced and differentiated by Raman signals. The amphiliphic GNPs were able to enhance the SERS signals of the protein emulsifier in the emulsion in situ and the hydrophobic GNPs were able to enhance the SERS signals from canola oil. Further purification of the hydrophobic GNPs proved to have higher enhancement and sensitivity, but still poor consistency which is typical of SERS. Monitoring lipid oxidation using Raman and SERS using alternative …

Cell-Based Sensing Of Endocrine Disrupting Substances Using Fluorescent Protein-Gold Nanoparticle Complexes, Xian Wang Aug 2014

Cell-Based Sensing Of Endocrine Disrupting Substances Using Fluorescent Protein-Gold Nanoparticle Complexes, Xian Wang

Masters Theses

Developing a sensitive and effective in vitro bioassay to detect endocrine disrupting chemicals (EDCs) would reduce the cost, eliminate the possibility of low dose effects, detect the non-monotonic dose responses, and identify mechanisms of actions. The “chemical nose” sensing method using supramolecular complexes composed of cationic monolayer functionalized gold nanoparticles (AuNPs) and fluorescent proteins (FPs) can successfully distinguish serum proteins, mammalian cells, tissue lysates, and chemotherapeutic drug mechanisms. EDCs regulate target cells via genomic or non-genomic pathways in terms of proliferative effect and response time. In this thesis, green fluorescent protein-gold nanoparticle (GFP-AuNP) sensors were used to detect the proliferative …

Chemistry Graduate Teaching Assistants: A Comparison Of The Classroom Discourse Within Expository And Problem-Based Learning Laboratories, Kelley M. Current Jun 2014

Chemistry Graduate Teaching Assistants: A Comparison Of The Classroom Discourse Within Expository And Problem-Based Learning Laboratories, Kelley M. Current

Masters Theses

Graduate Teaching Assistants (GTAs) commonly function as instructors within undergraduate chemistry laboratories. This study sought to explore and describe GTA classroom discourse within two distinct instructional modes, using discourse analysis as the theoretical framework. Theclassroom discourse within a series of verification style labs was comparedto the classroom discourse produced within a set of Problem-Based Learning (PBL) labs. The results suggest three primary findings: (1) the apparent relationship between the instructional mode and form of GTA classroom discourse, (2) the patterns in classroom discourse observed within a given instructional mode repeat, irrespective of content, and (3) the classroom discourse observed within …

Using Ultra Performance Liquid Chromatography-Mass Spectrometric Based Metabolomics To Determine The Metabolic Differences Due To Low Temperature Or Genetic Mutation In Salmonella Enterica, Nicole Rhonda Mooney May 2014

Using Ultra Performance Liquid Chromatography-Mass Spectrometric Based Metabolomics To Determine The Metabolic Differences Due To Low Temperature Or Genetic Mutation In Salmonella Enterica, Nicole Rhonda Mooney

Masters Theses

Metabolomics aims to identify and quantify all of the small molecules within a cell. The relative concentrations of metabolites within cells grown under different experimental conditions, or different species, can be compared using pool size data. Exposing cellular cultures to isotopically labeled compounds allows the progress of metabolites through metabolic pathways to be tracked, known as flux. In the first experiment, the metabolic effects of exposure to lower than optimal temperatures in wild type Salmonella enterica, i.e. cold shock, were determined. The metabolites involved in glycolysis, the citric acid cycle, and the pentose phosphate pathway had less efficient labeling …

Characterizing The Morphology Of Solvent Annealed Pcbm/P3ht Bilayers Using Gixrd And Raman Spectroscopy, Tyler Evan Mcpherson May 2014

Characterizing The Morphology Of Solvent Annealed Pcbm/P3ht Bilayers Using Gixrd And Raman Spectroscopy, Tyler Evan Mcpherson

Masters Theses

A growing demand for new forms of renewable energy has been at the forefront of scientific research. Organic photovoltaics (OPVs) provide a promising alternative to the expensive inorganic solar cells used today. Poly(3-hexylthiophene) (P3HT) and [6,6]-phenyl C61-butyric acid methyl ester (PCBM) are common donor and acceptor materials used in the active layer of an OPV device. These systems have reached power conversion efficiencies of approximately 12 %. However, little is known about the actual morphology. A better understanding of the morphology would allow researchers to optimize device fabrication to increase the device performance. As-cast devices are limited by the processing …

Gold-Magnetite Nanoparticle-Biomolecule Conjugates: Synthesis, Properties And Toxicity Studies, Akshay Pariti

Masters Theses

"This thesis study focuses on synthesizing and characterizing gold-magnetite optically active magnetic nanoparticle and its conjugation with biomolecules for biomedical applications, especially magnetic fluid hyperthermia treatment for cancerous tissue. Gold nanoparticles have already displayed their potential in the biomedical field. They exhibit excellent optical properties and possess strong surface chemistry which renders them suitable for various biomolecule attachments. Studies have showed gold nanoparticles to be a perfect biocompatible vector. However, clinical trials for gold mediated drug delivery and treatment studied in rat models identified some problems. Of these problems, the low retention time in bloodstream and inability to maneuver externally …

Synthesis And Characterization Of Molecular Components For The Preparation Of Poly-Rotaxanes, Austin P. Welti Jan 2014

Synthesis And Characterization Of Molecular Components For The Preparation Of Poly-Rotaxanes, Austin P. Welti

Masters Theses

The self-sorting ability of certain crown ethers make them important players in the field of supramolecular chemistry, especially those studying mechanically interlocked molecules (MIMs). Of these MIMs, rotaxanes and catenanes are the most common. Our approach in the synthesis of poly-rotaxane systems involves the preparation of axles containing multiple recognition motifs to determine the self-sorting principles that each motif shows. These principles include narcissistic self-sorting, and also type I and type II social self-sorting principles. Using these techniques, poly-rotaxane formation can be made readily available.

A hetero-poly-rotaxane equipped with a naphthalene diimide (NDI) site and two ammonium salt sites has …

Computational And Experimental Studies Of Non-Covalent Interactions: Characterization Of Ch...Π Interactions And Structure Of 1,1-Difluoroethylene...Carbon Dioxide, Ashley M. Anderton

Masters Theses

Weakly bound CH/π interactions, due to the abundance of C-H bonds and π systems that exist in larger organic molecules, are an important driving force in the construction of biomacromolecules, supramolecular assemblies and crystal packing of organic compounds. To assist in the characterization of CH/π interactions, a density functional theory (DFT) study has been conducted to determine a quick and cheap method for accurate determination of the binding energy and rotational constants for CH/π interactions. Seven complexes were used in this study: five benzene-HY complexes (where Y= C≡CH, Cl, F, Br and C≡N) and two fluorobenzene-HY complexes (where Y= C≡CH …

Investigations Towards Synthesis Of Cinnamaldehyde Semicarbazone Derivatives And Their Photochromicity By Modifying The Ring, The Linker, And The Semicarbazone, Xiao Xiao Jan 2014

Investigations Towards Synthesis Of Cinnamaldehyde Semicarbazone Derivatives And Their Photochromicity By Modifying The Ring, The Linker, And The Semicarbazone, Xiao Xiao

Masters Theses

Cinnamaldehyde semicarbazone was varied in three ways, by altering the ring, the conjugated linker, and the semicarbazone, to investigate how changing the structure affects the photochomicity. The synthesis of 7 hetero- and 2 polycyclic- aromatic semicarbazone variants involved three steps, (a Wittig reaction, hydrolysis, and condensation to the semicarbazone) with cumulative yields of 15% to 52%. Generally, the pyridine series gave lower yields than other synthesized semicarbazones. The Wittig reaction gave cis/trans mixtures in ratios of 2 : 1 to 1 : 7 for the 6 acetals, except for the 3 that went directly to the aldehyde, due to the …
