Background

Black men who have sex with men (MSM) in the United States (US) are affected by HIV at disproportionate rates compared to MSM of other race/ethnicities. Current HIV incidence estimates in this group are needed to appropriately target prevention efforts.

Methods

From July 2009 to October 2010, Black MSM reporting unprotected anal intercourse with a man in the past six months were enrolled and followed for one year in six US cities for a feasibility study of a multi-component intervention to reduce HIV infection. HIV incidence based on HIV seroconversion was calculated as number of events/100 person-years. Multivariate proportional …

Speer, Vittorio Addona, Christy L. Hanson, Joseph K. Sitienei, Hillary K. Kipruto, Martin Muhingo Githiomi, Brenda Nyambura Mungai

Vittorio Addona

" Despite high tuberculosis (TB) treatment success rate, treatment adherence is one of the major obstacles to tuberculosis control in Kenya. Our objective was to identify patient-related factors that were associated with time to TB treatment interruption and the geographic distribution of the risk of treatment interruption by county. Data of new and retreatment patients registered in TIBU, a Kenyan national case-based electronic data recording system, between 2013 and 2014 was obtained. Kaplan-Meier curves and log rank tests were used to assess the adherence patterns. Mixed-effects Cox proportional hazards modeling was used for multivariate analysis. Records from 90,170 patients were …

Non-Human Primates In Neuroscience Research: The Case Against Its Scientific Necessity, Jarrod Bailey, Katy Taylor Sep 2016

Non-Human Primates In Neuroscience Research: The Case Against Its Scientific Necessity, Jarrod Bailey, Katy Taylor

Jarrod Bailey, PhD

Public opposition to non-human primate (NHP) experiments is significant, yet those who defend them cite minimal harm to NHPs and substantial human benefit. Here we review these claims of benefit, specifically in neuroscience, and show that: a) there is a default assumption of their human relevance and benefit, rather than robust evidence; b) their human relevance and essential contribution and necessity are wholly overstated; c) the contribution and capacity of non-animal investigative methods are greatly understated; and d) confounding issues, such as species differences and the effects of stress and anaesthesia, are usually overlooked. This is the case in NHP …

An Assessment Of The Role Of Chimpanzees In Aids Vaccine Research, Jarrod Bailey Sep 2016

An Assessment Of The Role Of Chimpanzees In Aids Vaccine Research, Jarrod Bailey

Jarrod Bailey, PhD

Prior to Simian Immunodeficiency Virus (SIV)-infected macaques becoming the ‘model of choice’ in the 1990s, chimpanzees were widely used in AIDS vaccine research and testing. Faced with the continued failure to develop an effective human vaccine, some scientists are calling for a return to their widespread use. To assess the past and potential future contribution of chimpanzees to AIDS vaccine development, databases and published literature were systematically searched to compare the results of AIDS vaccine trials in chimpanzees with those of human clinical trials, and to determine whether the chimpanzee trials were predictive of the human response. Protective and/or therapeutic …

The Other Side Of Silence: Rachel Carson’S Views Of Animals, Marc Bekoff, Jan Nystrom Sep 2016

The Other Side Of Silence: Rachel Carson’S Views Of Animals, Marc Bekoff, Jan Nystrom

Marc Bekoff, PhD

The publication of Rachel Carson’s Silent Spring in 1962 is widely regarded as one of the major events that launched the modern environmental movement. Silent Spring is a compelling blend of stories, natural history, human values, and biological facts. In this essay we consider Carson’s attitude toward animals in Silent Spring and in other texts. Despite the facts that she was raised to love Nature and animals, little direct attention has been given to Carson’s views about our moral responsibilities to, and the moral standing of animals. Carson favored responsible stewardship, was more of an animal welfarist and environmentalist/conservation biologist …

Compassion As A Practical And Evolved Ethic For Conservation, David Ramp, Marc Bekoff Sep 2016

Compassion As A Practical And Evolved Ethic For Conservation, David Ramp, Marc Bekoff

Marc Bekoff, PhD

The ethical position underpinning decisionmaking is an important concern for conservation biologists when setting priorities for interventions. The recent debate on how best to protect nature has centered on contrasting intrinsic and aesthetic values against utilitarian and economic values, driven by an inevitable global rise in conservation conflicts. These discussions have primarily been targeted at species and ecosystems for success, without explicitly expressing concern for the intrinsic value and welfare of individual animals. In part, this is because animal welfare has historically been thought of as an impediment to conservation. However, practical implementations of conservation that provide good welfare outcomes …

Generalizing Evidence From Randomized Trials Using Inverse Probability Of Sampling Weights, Ashley L. Buchanan, Michael G. Hudgens, Stephen R. Cole, Katie R. Mollan, Paul E. Sax, Eric S. Daar, Adaora A. Adimora, Joseph J. Eron, Michael J. Mugavero Sep 2016

Generalizing Evidence From Randomized Trials Using Inverse Probability Of Sampling Weights, Ashley L. Buchanan, Michael G. Hudgens, Stephen R. Cole, Katie R. Mollan, Paul E. Sax, Eric S. Daar, Adaora A. Adimora, Joseph J. Eron, Michael J. Mugavero

Ashley Buchanan

Results obtained in randomized trials may not easily generalize to target populations. Whereas in randomized trials the treatment assignment mechanism is known, the sampling mechanism by which individuals are selected to participate in the trial is typically not known and assuming random sampling from the target population is often dubious. We consider an inverse probability of sampling weighted (IPSW) estimator for generalizing trial results to a target population. The IPSW estimator is shown to be consistent and asymptotically normal. A consistent sandwich-type variance estimator is derived and simulation results are presented comparing the IPSW estimator to a previously proposed stratified …

Alamethicin In Lipid Bilayers: Combined Use Of X-Ray Scattering And Md Simulations, Jianjun Pan, D. Peter Tieleman, John F. Nagle, Norbert Kučerka, Prof. Stephanie Tristram-Nagle Ph.D. Aug 2016

Alamethicin In Lipid Bilayers: Combined Use Of X-Ray Scattering And Md Simulations, Jianjun Pan, D. Peter Tieleman, John F. Nagle, Norbert Kučerka, Prof. Stephanie Tristram-Nagle Ph.D.

John Copeland Nagle

We study fully hydrated bilayers of two di-monounsaturated phospholipids diC18:1PC (DOPC) and diC22:1PC with varying amounts of alamethicin (Alm). We combine the use of X-ray diffuse scattering and molecular dynamics simulations to determine the orientation of alamethicin in model lipids. Comparison of the experimental and simulated form factors shows that Alm helices are inserted transmembrane at high humidity and high concentrations, in agreement with earlier results. The X-ray scattering data and the MD simulations agree that membrane thickness changes very little up to 1/10 Alm/ DOPC. In contrast, the X-ray data indicate that the thicker diC22:1PC membrane thins with added …

Institutional Animal Care And Use Committees: A Flawed Paradigm Or Work In Progress?, John P. Gluck, F. Barbara Orlans Aug 2016

Institutional Animal Care And Use Committees: A Flawed Paradigm Or Work In Progress?, John P. Gluck, F. Barbara Orlans

John P. Gluck, PhD

In his challenging article, Steneck (1997) criticized the creation of the Institutional Animal Care and Use Committee (IACUC) system established by the 1985 amendments to the Animal Welfare Act. He saw the IACUC review and approval of biomedical and behavioral research with animals as an unnecessary "reassignment" of duties from existing animal care programs to IACUC committees. He argued that the committees are unable to do the work expected of them for basically three reasons: (a) the membership lacks the expertise in matters relevant to animal research and care, (b) there exists an inherent and disabling conflict of interest, and …

Nicholas Whiting

Silicon-based nanoparticles are ideally suited for use as biomedical imaging agents due to their biocompatibility, biodegradability, and simple surface chemistry that facilitates drug loading and targeting. A method of hyperpolarizing silicon particles using dynamic nuclear polarization, which increases magnetic resonance imaging signals by several orders-of-magnitude through enhanced nuclear spin alignment, has recently been developed to allow silicon particles to function as contrast agents for in vivo magnetic resonance imaging. The enhanced spin polarization of silicon lasts significantly longer than other hyperpolarized agents (tens of minutes, whereas <1  min for other species at room temperature), allowing a wide range of potential …

Impacts Of Cover Crops On Phosphorus And Nitrogen Loss With Surface Runoff, Antonio Mallarino, Richard Cruse, Dan Jaynes, John Sawyer, Pablo Barbieri Jul 2016

Impacts Of Cover Crops On Phosphorus And Nitrogen Loss With Surface Runoff, Antonio Mallarino, Richard Cruse, Dan Jaynes, John Sawyer, Pablo Barbieri

John E. Sawyer

Iowa research has demonstrated that cover crops can improve soil productivity and water quality by increasing soil organic matter and reducing nitrate nitrogen (N) leaching. Other research has investigated and is investigating the agronomic and economic viability of using cereal rye cover crops in continuous corn or corn-soybean rotations. However, no Iowa research has evaluated under natural rainfall the impact of cover crops on phosphorus (P) and N loss with surface runoff interacting with other management practices. The need for this type of research was indicated in the Iowa Nutrient Reduction Strategy documents. This effort assessed what would be needed …

Impact Of 4r Management On Crop Production And Nitrate-Nitrogen Loss In Tile Drainage, Matthew Helmers, John Sawyer, Josh Sievers

John E. Sawyer

Corn Belt corn and soybean producers are increasingly challenged to maximize crop production while addressing the contributions farm practices make to Gulf hypoxia. Based on the need for nitrate-N reductions to meet water quality goals, new management practices are needed to reduce nitrate-N losses at minimal cost and maximum economic benefits. This three-year field research and demonstration project is evaluating various promising N management methods and technologies by documenting the nitrate-N export and crop yield from various systems.

Lifetime Prevalence Of Non-Melanoma And Melanoma Skin Cancer In Australian Recreational And Competitive Surfers, Michael Climstein, James Furness, Wayne Hing, Joe Walsh

Wayne Hing

Background/Purpose Surfing is one of the most popular outdoor aquatic activities in Australia with an estimated 2.7 million recreational surfers; however, Australia has long been recognized as having the highest incidence of melanoma in the world, and it is the most common type of cancer in young Australians. The aim of this study was to investigate the lifetime prevalence of non-melanoma [basal cell carcinoma (BCC), squamous cell carcinoma (SCC)] and melanoma skin cancers in Australian recreational and competitive surfers. Methods Australian surfers were invited to complete an online surveillance survey to determine the lifetime prevalence of non-melanoma and melanoma skin …

Food For Thought … Mechanistic Validation, Thomas Hartung, Sebastian Hoffman, Martin Stephens Jul 2016

Food For Thought … Mechanistic Validation, Thomas Hartung, Sebastian Hoffman, Martin Stephens

Martin Stephens, PhD

Validation of new approaches in regulatory toxicology is commonly defined as the independent assessment of the reproducibility and relevance (the scientific basis and predictive capacity) of a test for a particular purpose. In large ring trials, the emphasis to date has been mainly on reproducibility and predictive capacity (comparison to the traditional test) with less attention given to the scientific or mechanistic basis. Assessing predictive capacity is difficult for novel approaches (which are based on mechanism), such as pathways of toxicity or the complex networks within the organism (systems toxicology). This is highly relevant for implementing Toxicology for the 21st …

Noncompliance With Public Health Service (Phs) Policy On Humane Care And Use Of Laboratory Animals: An Exploratory Analysis, Leah M. Gomez, Kathleen Conlee, Martin Stephens

Martin Stephens, PhD

The National Institutes of Health (NIH) is a major biomedical research-funding body in the United States. Approximately 40% of NIH-funded research involves experimentation on nonhuman animals (Monastersky, 2008). Institutions that conduct animal research with NIH funds must adhere to the Public Health Service (PHS) care and use standards of the Office of Laboratory Animal Welfare (OLAW, 2002a). Institutions deviating significantly from the PHS’s animal care and use standards must report these incidents to the NIH’s OLAW. This study is an exploratory analysis of all the significant deviations reported by animal-research facilities to OLAW during a 3-month period. The study identifies …

Addressing Distress And Pain In Animal Research: The Veterinary, Research, Societal, Regulatory And Ethical Contexts For Moving Forward, Kathleen Conlee, Martin Stephens, Andrew N. Rowan Jul 2016

Addressing Distress And Pain In Animal Research: The Veterinary, Research, Societal, Regulatory And Ethical Contexts For Moving Forward, Kathleen Conlee, Martin Stephens, Andrew N. Rowan

Martin Stephens, PhD

While most people recognize that biomedical scientists are searching for knowledge that will improve the health of humans and animals, the image of someone deliberately causing harm to an animal in order to produce data that may lead to some future benefit has always prompted an uncomfortable reaction outside the laboratory. However, proponents of animal research have usually justified the practice by reference to greater benefits (new knowledge and medical treatments) over lesser costs (in animal suffering and death). Given that one of the costs of animal research is the suffering experienced by the animals, the goal of eliminating distress …

The Usefulness Of Systematic Reviews Of Animal Experiments For The Design Of Preclinical And Clinical Studies, Rob B.M. De Vries, Kimberley E. Weaver, Marc T. Avey, Martin Stephens, Emily S. Sena, Marlies Leenaars Jul 2016

The Usefulness Of Systematic Reviews Of Animal Experiments For The Design Of Preclinical And Clinical Studies, Rob B.M. De Vries, Kimberley E. Weaver, Marc T. Avey, Martin Stephens, Emily S. Sena, Marlies Leenaars

Martin Stephens, PhD

The question of how animal studies should be designed, conducted, and analyzed remains underexposed in societal debates on animal experimentation. This is not only a scientific but also amoral question. After all, if animal experiments are not appropriately designed, conducted, and analyzed, the results produced are unlikely to be reliable and the animals have in effect been wasted. In this article, we focus on one particular method to address this moral question, namely systematic reviews of previously performed animal experiments. We discuss how the design, conduct, and analysis of future (animal and human) experiments may be optimized through such systematic …

The Minimization Of Research Animal Distress And Pain: Conclusions And Recommendations, Kathleen Conlee, Martin Stephens, Andrew N. Rowan Jul 2016

The Minimization Of Research Animal Distress And Pain: Conclusions And Recommendations, Kathleen Conlee, Martin Stephens, Andrew N. Rowan

Martin Stephens, PhD

While the attention given to preventing, assessing, and alleviating pain in research animals has increased noticeably in recent decades, much remains to be done both in terms of implementing best practices and conducting studies to answer outstanding questions. In contrast, the attention to distress (particularly non-pain induced distress) has shown no comparable increase. There are many reasons for this discrepancy, including the conceptual untidiness of the distress concept, the paucity of pharmacological treatments for distress, and perceived lack of regulatory emphasis on distress. These are challenges that need to be addressed and overcome. This book is intended to help meet …

Animal Model Research: The Apples And Oranges Quandary, Kenneth J. Shapiro Apr 2016

Animal Model Research: The Apples And Oranges Quandary, Kenneth J. Shapiro

Kenneth J. Shapiro, PhD

In this paper, I explore the premises underlying the problem of the evaluation of animal models. I argue that the presence of similarities and differences between the model and the modelled, although historically and currently a dominant antinomy framing evaluation, is not a bottom-line consideration. What is critical is 1) whether we learn and 2) whether we improve treatment through the animal model research. Similarity between model and modelled and the closely related concept of validity are not coterminus with these critical evaluative measures. In fact, differences between the model and modelled also can provide impetus to new understanding and …

A Sodium Salt Of The Dimer Of Boronoterephthalic Acid Anhydride, Scott Simmons, Albert Fratini, Vladimir Benin Mar 2016

A Sodium Salt Of The Dimer Of Boronoterephthalic Acid Anhydride, Scott Simmons, Albert Fratini, Vladimir Benin

Albert Fratini

The title compound, sodium bis(6-carboxy-1-hydroxy-3-oxo-1,3-dihydro-2,1-benzoxaborol-1-yl)oxidanium, Na+·C16H15B2O13-, was prepared in two steps from 2-bromo-p-xylene. Its crystal structure was determined at 140 K and has triclinic (P) symmetry. The compound presents a unique structural motif, including two units of the cyclic anhydride of boronoterephthalic acid, joined by a protonated, and thereby trivalent, oxonium center. Association in the crystal is realized by complementary hydrogen bonding of the carboxyl groups, as well as by coordination of the sodium cations to the oxygen centers on the five-membered rings.

A Data-Driven Behavior Modeling And Analysis Framework For Diabetic Patients On Insulin Pumps, Sanjian Chen, Lu Feng, Michael Rickels, Amy Peleckis, Oleg Sokolsky, Insup Lee Mar 2016

A Data-Driven Behavior Modeling And Analysis Framework For Diabetic Patients On Insulin Pumps, Sanjian Chen, Lu Feng, Michael Rickels, Amy Peleckis, Oleg Sokolsky, Insup Lee

Oleg Sokolsky

About 30%-40% of Type 1 Diabetes (T1D) patients in the United States use insulin pumps. Current insulin infusion systems require users to manually input meal carb count and approve or modify the system-suggested meal insulin dose. Users can give correction insulin boluses at any time. Since meal carbohydrates and insulin are the two main driving forces of the glucose physiology, the user-specific eating and pump-using behavior has a great impact on the quality of glycemic control.

In this paper, we propose an “Eat, Trust, and Correct” (ETC) framework to model the T1D insulin pump users’ behavior. We use machine learning …
