Background/aim: To evaluate the influence of caffeic acid phenethyl ester (CAPE) on cigarette smoke (CS)-induced cell damage, oxidative stress, and inflammation in human alveolar epithelial cells. Materials and methods: A549 alveolar epithelial cells were divided into control, CS exposure, CAPE, and CS+CAPE treatment groups. Undiluted CS-exposed medium (100%) and three dilutions (50%, 25%, and 10%) of CS-exposed media were applied to cultured A549 cells, which were analyzed after 3 h of incubation. Viability was measured by MTT assay, the gene expressions were evaluated by real-time PCR, and spectrophotometric techniques were used for biochemical assessments. Background/aim: Methotrexate (MTX) has been widely used for treatment of cancer and rheumatoid arthritis, but its use has been limited by its nephrotoxicity. This study was carried out to determine whether garlic exerts a protective effect against MTX-induced nephrotoxicity. Materials and methods: Nephrotoxicity was induced in rats after a single i.p. injection of MTX (20 mg/kg). Garlic extract (1 mL/100 g b.w.) was given orally for 7 days before and after MTX administration. Serum samples were collected to evaluate urea, creatinine, sodium, phosphorous, potassium, and calcium. Human alveolar epithelial cells play a critical role in the pathogenesis of lung diseases. The objective of this study is to determine the contribution of beraprost sodium, a prostaglandin I_2 (PGI_2) analogue, to inflammatory and oxidative events in response to lipopolysaccharide (LPS) in airway epithelial cells. Salivary Glucose And Antioxidant Defense Markers In Type Ii Diabetes Mellitus, Sayeeda Mussavira, Mala Dharmalingam, Bindhu Omana Sukumaran

To evaluate salivary antioxidant defense markers, their correlation with salivary glucose, and glycemic status in type II diabetes mellitus (DM). Materials and methods: The study included 53 diabetic patients and 40 healthy subjects. Salivary glucose, blood glucose, and uric acid (UA) were determined by specific enzymatic methods. Total antioxidant activity (AOA), glutathione (GSH), catalase (CAT), superoxide dismutase (SOD), and total protein were determined spectrophotometrically. Results: Salivary UA (3.12 vs. 1.89 mg/dL), GSH (47 vs. 11.92 µg/mL), and total protein (375.12 vs. 202.23 mg/dL) were significantly higher (P < 0.001; r = 0.455, 0.735, 0.498 respectively) and AOA (653.1 vs. 897.3 µmol/L) was significantly lower in the DM group (P < 0.001, r = -0.431) compared to healthy controls. Among the antioxidant enzymes, CAT was significantly lower (1214 vs. 9468.9 kat) in the DM group (P < 0.001, r = -0.886). Spearman correlation analyses within the diabetic group showed a strong positive association between salivary glucose and blood glucose (P < 0.001, r = 0.9), salivary glucose and GSH, and salivary glucose and UA. Salivary glucose showed a negative correlation with AOA and CAT (P = 0.008, r = -0.447) in the diabetic group. Conclusion: Findings of this study, showing a strong correlation between salivary glucose and blood glucose as well as changes in antioxidant components in the DM group, suggest that saliva can be used for the diagnosis and management of DM.

To evaluate the effects of oxidant/antioxidant mechanisms and levels of trace elements on trauma-stimulated moderate pulmonary contusions after vitamin E administration. Materials and methods: Sixty-three male Sprague Dawley rats were used. Animals were studied in 4 groups. Vitamin E (150 mg/kg) was injected intraperitoneally 30 min after trauma and on the first and second days. Blood samples were obtained for nitric oxide (NO) levels and superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities. Background/aim: The aims of this study were to investigate the significance of oxidative stress parameters in the pathogenesis of ischemic stroke and hemorrhagic stroke and to investigate their effects on stroke severity using the National Institutes of Health Stroke Scale (NIHSS). Recent studies have demonstrated that inhibition of poly(ADP-ribose) polymerase (PARP) decreases tissue necrosis and inflammation. We evaluated the efficacy of 3-aminobenzamide (3-AB), a PARP inhibitor, in a rodent model of APAP-induced hepatotoxicity. Background/aim: The effects of coadministration of melatonin and theanine (Mel/Thea) on pentylenetetrazole (PTZ)-induced seizures and brain tissue oxidative damage were investigated in ovariectomized (OVX) and sham-operated rats. Materials and methods: The rats were divided into the following groups: 1) sham, 2) ovariectomized (OVX), 3) sham-PTZ, 4) OVX-PTZ, 5) sham-Mel/Thea-PTZ, and 6) OVX-Mel/Thea-PTZ.
