The In Vitro Determination Of Genotoxicity In Peripheral Lymphocytes Of Welders Exposed To Fumes From Metal Arc Welding, Ceylan Şener, Hali̇l Erhan Eroğlu

Turkish Journal of Medical Sciences

Gas metal arc welding is a widely used method in many industrial areas. In this study, the peripheral blood lymphocytes of 23 welders and 25 nonexposed subjects were monitored for genotoxicity. Materials and methods: The blood samples obtained from the subjects were incubated with a culture medium at 37 °C for 72 h. Cytochalasin-B and BrdU were added for micronucleus and replication index. After incubation, the cultures were harvested, stained, and examined. Results: The micronucleus rate (2.165 ± 1.645) of the welders was quite higher than that of those not exposed (1.352 ± 0.978) (P < 0.01). There was a significant difference between micronucleus rates of smoking and nonsmoking subjects. It was found that as the time of exposure to welding fumes increased, so did the micronucleus rates (2.938 ± 1.723) (P < 0.05). A positive correlation was observed between micronucleus frequency and age. The replication index rate (1.026 ± 0.020) of the welders was higher than that of those not exposed (1.019 ± 0.016) (P = 0.01). Conclusion: It is advised that welders who work with gas metal arc welding be instructed and that necessary protective measures be taken because of the risk caused by metal arc welding fumes.

To investigate the effect of insulin on receptor activator of nuclear factor kappa-B ligand (RANKL) and osteoprotegerin (OPG) expression in human osteosarcoma MG63 cells using real-time polymerase chain reaction. Materials and methods: MG63 cells were exposed to different concentrations of human recombinant insulin (1, 10, and 100 nM) for 24 h, and then the expression of RANKL and OPG mRNA was identified. After that, MG63 cells were exposed to tensile stress combined with 0 or 10 nM insulin respectively for 0, 3, 6, and 12 h, and then the expression of RANKL and OPG mRNA was identified. Human monocytes were cultured …

Double-J Stent Placement With Grasping Forceps Through Ureteroscope Working Channel: A Novel Technique, Aki̇f Koç, Ömer Bayrak

Turkish Journal of Medical Sciences

To report a new technique for the retrograde placement of a double-J stent with grasping forceps through the ureteroscope working channel. Materials and methods: In our technique, the ureteroscope is advanced to the renal pelvis or proximal location of the obstruction in the ureteral lumen; a double-J stent is then introduced into the ureteroscope working channel. The double-J stent is pushed forward until its proximal tip is seen; then the clamp over the distal tip of the pusher is opened and the guide-wire and pusher are removed from the channel, respectively. The grasping forceps is inserted into the working channel …

To investigate the effect of hypercholesterolemia on in vitro contractions of the detrusor smooth muscle stimulated by an electrical field in rats and the relative contributions of cholinergic and noncholinergic neurotransmission. Materials and methods: Sprague-Dawley rats were fed with standard diet (NC group) or with 4% cholesterol diet (HC group). After 4 weeks, the urinary bladder was excised under anesthesia and 4 detrusor strips were prepared. Blood samples were collected for serum lipid profile, and aortic arches were dissected for histopathological examination. In vitro contractile function of the detrusor smooth muscle was evaluated with: 1) electrical field stimulation (EFS) only, …
