The purpose of this project was to study the antibacterial effect of the short exposure of 5.25% NaOCl and 17% EDTA on eight strains of Enterococcus faecalis in extracted human teeth. The crowns of 240 single rooted teeth were removed and the canals were instrumented to ISO 0.36. The canals were irrigated with 5.25% NaOCl and 17% EDTA, autoclaved and separated into eight groups of 30 teeth consisting of 20 experimental teeth, 5 positive and 5 negative controls. The purpose of this investigation was to determine the antimicrobial effect of Biopure MTAD (a mixture of a tetracycline isomer, an acid, and a detergent) as a final irrigant on eight strains of Enterococcus faecalis (E. faecalis) in contaminated root canals of extracted human teeth. In addition, the Minimum Inhibitory Concentration (MIC) and Minimum Lethal Concentration (MLC) of MTAD were also measured on the same strains of bacteria. Enterococcus faecalis is the most commonly isolated bacteria in failed root canal treatment. Endodontic intracanal medicaments are commonly tested using standardized bovine cylinders infected with bacteria. The literature is not clear on whether calcium hydroxides are able to remove E. faecalis from the dentinal tubules. PURPOSE: To compare the ability of three-to four-year-old children in performing toothbrushing with manual versus battery-powered toothbrushes with and without oral hygiene instructions.

METHODS: This study was a randomized, controlled, examiner-blind, 4-period crossover design performed on 50 healthy children, ages 3 to 4 years old. Children were assigned either a manual or power toothbrush at visit one and alternated between the two types of toothbrushes each week, for four weeks. While previous studies have shown that presence of bacteria in systemic circulation can cause infective endocarditis, there is no information on the specific amount of bacteria necessary to cause this condition. The purpose of this study was to establish the minimum level of circulating bacteria that will cause colonization of damaged heart valves in rats. Fifty-two Sprague-Dawley rats were anesthetized and their heart valves were damaged using an established protocol. A clinical isolate of Enterococcus faecalis was transformed with a plasmid bearing the luxF and Chloramphenicol cassettes to label the bacteria with the light-emitting protein. Bacteria have been shown to play an important role in the development of pulp and periradicular disease. The source of the infective bacteria is most often the oral cavity. Previous studies have detected bacteria in teeth that are structurally intact that seemingly have no communication intraorally. Anachoresis has been proposed as a mechanism for localizing circulating bacteria to such teeth. In several previous bacteriologic anachoresis studies, bacteria have been found to localize in inflamed dental tissues. The criticism most commonly associated with these investigations has been the potential contribution of contaminant bacteria resulting in false positive findings.

Children often present with dental trauma to pediatric emergency departments and pediatric dental offices. There is a wide spectrum of types of dental trauma. Although research has been done concerning the treatment of avulsed teeth, the current recommendations in the published guidelines might still be improved for better long term prognoses. One may speculate that ultrasonification of an avulsed tooth prior to replantation may dislodge existing surface bacteria and improve the prognosis for the tooth. A possible cause for root canal failure is the persistence of bacteria that have colonized dentinal tubules. To reduce this risk and, if possible, to also shorten the time consuming endodontic therapy, new equipment and materials are constantly being introduced. The aim of the present investigation was to study the effect of disinfection of dentinal tubules by intracanal laser irradiation using an in vitro model. Enamel demineralization due to increased plaque accumulation is a well-recognized problem associated with fixed orthodontic appliances. The purpose of this study was to evaluate the efficacy of a proprietary product to be marketed by 3M Unitek for the reduction of plaque accumulation and thereby reduce enamel demineralization.

Sixty four bovine teeth were divided equally into an untreated control group and a group treated with the proprietary sealant product. Ozone is a major component of photochemical smog. High levels of this pollutant, sufficient to affect human health are found in many urban areas worldwide. Though limited studies in humans are supported by extensive findings from animal experiments, a difficulty in interpreting the results of these experiments has lead to an ambiguity on the biochemical mechanism of ozone toxicity. The purpose of this study was to examine in vitro plaque formation on sealant coated enamel. The plaque growth on different types of sealants placed on enamel was compared to each other and to an unsealed enamel control. The antimicrobial properties of sealants were also studied.

Sealant coated bovine enamel disks were immersed in a S. mutans culture medium for four days. The amount of bacterial plaque formation was quantified by removing the deposit, suspending it in solution with subsequent serial dilution. The aim of this investigation was to characterize peri-implant tissue breakdown following ligature-induced plaque accumulation around osseointegrated implants in the rhesus monkey. Two hydroxyapatite-coated cylindrical implants were inserted bilaterally in the second premolar and first molar region of both mandible and maxilla in four non-human primates (Macaca mulatto). The implants were left submerged to osseointegrate for one year. Eight weeks after healing abutment connection, a two-month plaque control program was initiated. At the end of this period, clinical and radiographic recordings were carried out, and peri-implant subgingival microbial samples were obtained (baseline). This study investigated the site of ATP utilization in the signal transduction pathway of bacterial chemotaxis and localized the point of convergence of a methylation-independent system of chemotaxis with the methylation-dependent system. The identity of the signal originating from the phosphotransferase system was investigated by substituting the fructose-inducible HPr-like protein FPr for HPr in transport and chemotaxis. In addition, a novel chemoattractant, glycerol, was identified for Salmonella typhimurium. Histidine-auxotrophic S. typhimurium strains ST23 (hisF) and ST171 (hisF cheB) were depleted for ATP. Klebsiella pneumoniae is an enteric opportunistic pathogen which commonly causes pulmonary, urinary tract and kidney infections. It has also been widely used for the study of nitrogen fixation. In this study, a macrorestriction map of K. pneumoniae M5a1 chromosomal DNA was constructed using pulsed field gel electrophoresis (PFGE) and Southern hybridization techniques. The genome DNA was digested with two rare cutting restriction enzymes (XbaI and BlnI) and the genome size was estimated from the sum of the fragment sizes as 5,121 Kb (+2.8%). B. subtilis, like Escherichia coli and Salmonella typhimurium, responded to a step increase in oxygen concentration by swimming smoothly (~ 35 s duration) and to a decrease in oxygen by tumbling. In a spatial gradient of oxygen in liquid media, a band of cells congregated near the air interface. Aerotaxis required a functioning respiratory chain. Adaptation of B. subtilis, E. coli and S. typhimurium to media containing amino acids requires methylation of receptors/methyl-accepting chemotaxis proteins. Adaptation to oxygen and phosphotransferase sugar substrates in B. subtilis was dependent on methylation, unlike similar adaptation in E. coli and S. typhimurium. …

The pathogenesis of periapical lesions is the interaction of the oral flora with the existing host defenses. The role of bacteria has been established in endodontic disease but the contribution of the host defenses in periapical lesion formation is less clear. The purpose of this study was to compare periapical lesion development in immunosuppressed rats with that in normal rats. Thirty-five, eight-week old Sprague-Dawley rats were equally divided into seven groups. Groups 1, 2, and 3 were given weekly injections of Cytoxan, an immunosuppressive agent. The 5-lipoxygenase metabolites of arachidonic acid such as leukotrienes (LT) B4 and C4, are potent mediators of inflammation. The purpose of this investigation was to determine the concentrations of LTB4 and LTC4 in human periradicular lesions. Specimens from asymptomatic and symptomatic patients were obtained and immediately frozen in liquid nitrogen. Periradicular tissues from unerupted third molars as well as chronically inflamed gingival tissues were also obtained, frozen and were used as negative and positive controls respectively. Enzyme IImtl of Escherichia coli was purified from mtlA-overexpressing E. coli strain LJ1112 extraction of isolated membranes with deoxycholate, followed by hexyl agarose and ω-amino hexyl agarose chromatography in Lubrol PX. A 10-fold increase in total pure protein and 2.5-fold increase in specific activity over previously reported procedures was obtained. Tryptic fragments of Enzyme IImtl were also purified and identified as the membrane and cytoplasmic domains by protein sequencing and SDS-PAGE. Endodontic or root canal infection results in immunologic changes in periapical region, which causes the destruction of pulpal and periapical tissue. No definitive study has identified a specific organism or a group of organisms as a etiological agents. Past studies have indicated that immunoglobulin E play a significant role in the pathogenesis of periapical lesions. The presence of this factor suggests that some of the endodontic flare-ups may be IgE-mediated hypersensitivity reactions. The effects of a 12-week period of oral hygiene alone on gingival conditions and subgingival microflora in 15 patients with severe periodontitis were investigated. In addition, subgingival microsamples were compared during this oral hygiene phase with those obtained 1 week after debridement. Clinical measurements and subgingival samples from selected sites were taken at week 0 (baseline), week 6, and week 12. Endodontic (root canal) infections are generally considered to be caused by the invasion of microorganisms which destroy pulp tissue and extend to the periapical region. No definitive study has identified a specific bacterium or group of bacteria as etiological agents. We quantitively measured antibodies present in the serum of 30 normal individuals and patients with either chronic (n = 30) or acute (n = 30) periapical lesions with the indirect fluorescent antibody (IFA) and enzyme-linked immunosorbent assay (ELISA) for IgG and IgM against 11 oral microorganisms routinely isolated from such lesions. A group of 48 enteric bacteria was selected from a total of 170 at Loma Linda University Medical Center because of resistance to three or more antibiotics. They were examined to see if any of the antibiotic resistances were transferred by conjugation to either of two recipient bacteria. These were a restrictionless, antibiotic sensitive, azide resistant mutant of Escherichia coli K 12 designated E. coli 1228 and a similar phenotypic strain of Salmonella typhimurium LT2 designated LB 5000. This investigation evaluated the gingival condition of areas with minimal (less than or equal to 1.0 millimeter) and appreciable (greater than or equal to 2.0 millimeters) widths of keratinized gingiva.

Sixteen dental, dental hygiene, and dental assisting students and dental faculty who had contralateral or unilateral areas of minimal and appreciable widths of keratinized gingiva on the mid-buccal surface of mandibular bicuspids were examined. Gingival exudate, plaque, gingival inflammation based on color change and/or swelling and bleeding on probing were evaluated. The results showed that all areas examined were plaque free. An in vitro study using polyethylene tubes filled with Grossman's sealer, Rickert's sealer, and N2 permanent was conducted over a six month period in an effort to determine their lasting antimicrobial properties against eight different microorganisms.

From the study it was observed that N2 permanent possesses a long-lasting antimicrobial effect, but this effect appears to diminish greatly with time. Further investigation is needed to determine whether or not the effect is actually permanent in nature. In order to determine the duration of the effect of glucose, fructose, sucrose, honey, starch, and orange juice on neutrophilic phagocytosis, 10 subjects underwent a complete series of carbohydrate tolerance tests. Blood samples were drawn at specific time periods following ingestion of 100 grams of each carbohydrate. Blood was then analyzed for glucose and phagocytic index (the mean number of bacteria engulfed per leukocyte); a red blood cell and white blood cell count and differential white cell count were done also on about 1/2 of the samples. Two group A phages, P9c/2 and P4/2, were studied. Their density, sedimentation velocity, and biochemical properties were compared. Their densities were determined by density gradient centrifugation in 38.3% cesium chloride. Sedimentation velocity was used to determine their molecular weights. To accomplish this, the DNA of P9c/2 and P4/2 was labeled with tritium and T2 DNA, labeled with 32P, was used as a standard.
