Functional Analysis Of Chromatin Remodeler Csb In Parp1/2- Mediated Oxidative Dna Lesion Repair, Rabeya Bilkis

Biomedical Sciences ETDs

The ATP-dependent chromatin remodeler Cockayne syndrome protein B (CSB) is essential for maintaining genome integrity. Mutations in CSB are the primary cause of Cockayne syndrome (CS), a premature aging disorder with severe sun sensitivity and neurodevelopmental symptoms. However, the precise mechanisms underlying the disease etiology remain poorly understood. Given the critical roles of oxidative stress in aging and neurodegenerative disorders, this research sought to determine the mechanisms of CSB’s functions in oxidative DNA damage repair.

In the first aim, we investigated how CSB participates in PARP1/2-mediated oxidative DNA repair. We showed that PARP1 and PARP2 recruit CSB to oxidized chromatin …

The Enzymatic Regulation Of Contraction-Induced Toll-Like Receptor 4 Shedding From C2c12 Myotubes, Rebekah C. Gridley

Health, Exercise, and Sports Sciences ETDs

Stimulated contractions are shown to increase toll-like receptor 4 (TLR4) release from C2C12 myotubes in vitro. The purpose of this study was to determine if the release of TLR4 from myotubes is controlled by proteolytic membrane cleavage. A broad-spectrum metalloprotease inhibitor was used to test this hypothesis in a C2C12 cell culture model, and TLR4 concentrations in the cell culture supernatant were measured via enzyme-linked immunosorbent assay (ELISA) following stimulated myotube contractions.

ELISA analysis revealed TLR4 concentrations below the sensitivity threshold of the ELISA kit for all experimental conditions. In confirming the effectiveness of the electrical pulse stimulation (EPS) treatment, …

The Role Of Spreading Depolarizations In Mild Traumatic Brain Injuries, Natalie J. Pinkowski Apr 2024

The Role Of Spreading Depolarizations In Mild Traumatic Brain Injuries, Natalie J. Pinkowski

Biomedical Sciences ETDs

Mild traumatic brain injuries (mTBIs) often lead to acute symptoms like disorientation and discoordination, but the underlying mechanisms are unknown. Although most patients recover quickly from a single mTBI, repeated injuries can be debilitating. After an mTBI, a neurometabolic cascade produces metabolic burden and vulnerability. Spreading depolarizations (SDs) have been observed after mTBIs. Here we investigated SDs’ role in short-term motor behavioral deficits post-mTBI. We hypothesized that SDs contribute to the deficits, and exacerbated symptoms after repeated mTBIs. To test this, we used acute motor behavioral tests and long-term behavioral and cognitive tests after initiating SDs by mTBI, chemical, or …

Since its introduction, vaccination has improved health outcomes tremendously. However, modern vaccination efforts are still difficult to implement, particularly in developing countries with warm climates. This includes complex and expensive cold-chain management of vaccines to maintain stability, a lack of medical personnel to administer parental vaccines, and needle phobia. As an alternative, dissolvable microneedles have recently been studied for their simple and painless application. In this thesis, I present a novel manufacturing process for polymer-based dissolvable microneedles, loaded with virus-like particles vaccines conjugated to TRIO or Sialokinin peptides to target malaria and arboviruses, respectively. We have performed multiple characterization studies …

Pharmaceutical Sciences ETDs

Adults with Cystic Fibrosis (CF) are a unique and expanding patient population in the context of massive progress in disease screening and treatment. However, exacerbations remain a significant complication of the disease associated with its decline. CF pathophysiology promotes chronic respiratory bacterial infections, which are implicated in exacerbations. An important finding in research surrounding the respiratory microbiome has correlated the loss of diversity being with a worse prognosis in chronic lung diseases. We developed an aerosol formulation towards supporting the existing, diverse respiratory microbiome. Our formulation of prebiotics with and without antibiotics was designed to fit within the complex, multistep, …

Interrogation Of The Molecular Mechanisms That Finetune Fcεri Signaling And Mast Cell Outcomes, Derek A. Rinaldi

Biomedical Sciences ETDs

FcεRI is the ITAM receptor primarily responsible for the allergic response and immune hypersensitivity diseases such as asthma. Regulation of the strength of both FcεRI signaling, and the resulting outcomes, is accomplished via a complex array of molecular mechanisms whose interactions remain unclear. Here, I explored how the biophysical properties of antigens, crosstalk between FcεRI and integrin signaling and Arf6-mediated control of trafficking and exocytosis tune mast cell outcomes. I first found significant differences in FcεRI aggregation, phosphorylation and granule secretion after crosslinking with the multivalent antigen DNP-BSA or the small, trivalent antigen DF3. I also revealed a bi-directional, Syk-mediated …

Determining The Existence And Regulation Of Microlipophagy In Primary Brown And White Adipocytes, Sara C. Arenas De Leon

Biomedical Sciences ETDs

Lipids are one of life’s four main macromolecules and provide essential functions to cells.

The degradation of lipid droplets and mobilization of lipids occurs through lipolysis. Emerging evidence demonstrates evidence of a selective form of autophagy in lipolysis. The process of microlipophagy has only recently been described, and many features of its regulation are still poorly understood. Our recent study showed that inhibiting mammalian target of rapamycin complex 1 (mTORC1) pathway by depletion of Raptor in adipocytes led to an influx of lysosomes and accumulation of lipid droplets within lysosome. Here, we expand on these previous findings. We were able …

Effects Of Acute And Chronic Gut Injury On Secretory Lineage Differentiation, Roger Atanga Aug 2023

Effects Of Acute And Chronic Gut Injury On Secretory Lineage Differentiation, Roger Atanga

Biomedical Sciences ETDs

Acute and chronic injury to the gut has several detrimental consequences. However, dysregulation of secretory lineage cells is a common observation. To model acute gut injury, we use non-fissile uranium bearing dust (UBD), an environmental toxicant prevalent in communities adjacent abandoned uranium mines, majority of which are located in the Southwestern United States. Repeated exposure to environmental toxins and heavy metals has been obscurely linked to intestinal inflammation, increased susceptibility to pathogen-induced diseases, and higher incidences of colorectal cancer. However, there is no molecular evidence that non-fissile uranium can cause damage to the intestinal epithelia. Human colonoids exposed to UBD …
